The development of smartphones, specifically their cameras, and imaging technologies has enabled their use as sensors/measurement tools. Here we aimed to evaluate the applicability of a fast and noninvasive method for the estimation of total chlorophyll (Chl), Chl a, Chl b, and carotenoids (Car) content of soybean plants using a smartphone camera. Single leaf disc images were obtained using a smartphone camera. Subsequently, for the same leaf discs, a Chl meter was used to obtain the relative index of Chl and the photosynthetic pigments were then determined using a classic method. The RGB, HSB and CIELab color models were extracted from the smartphone images and correlated to Chl values obtained using a Chl meter and by a standard laboratory protocol. The smartphone camera was sensitive enough to capture successfully a broad range of Chl and Car contents seen in soybean leaves. Although there was a variation between color models, some of the proposed regressions (e.g., the S and b index from HSB and Lab color models and NRI [RGB model]) were very close to the Chl meter values. The effect of ultraviolet B radiation (UV-B) on cellular ultrastructure, chlorophyll (Chl), carotenoids, and total phenolics of Acrostichum danaeifolium gametophytes was analyzed. The control group of spores was germinated under standard conditions, while the test group of spores was germinated with additional UV-B for 30 min every day for 34 d. The cell characteristics were preserved in gametophytes irradiated with UV-B, but the number of starch grains increased in the chloroplasts and the more developed grana organization in contrast to the chloroplasts of the control group. Chl a content decreased, while Chl b content increased in the gametophytes cultivated with UV-B for 34 d. Contents of lutein and zeaxanthin decreased and trans-β-carotene concentration was enhanced in the gametophytes irradiated with UV-B. The content of total phenolic compounds increased in the gametophytes cultivated with UV-B. Therefore our data suggest that the gametophytes of A. danaeifolium, a fern endemic to the mangrove biome, were sensitive to enhancement of UV-B radiation at the beginning of their development and they exhibited alterations in their ultrastructure, pigment contents, and protective mechanisms of the photosynthetic apparatus, when exposed to this radiation., A. Over last decades, several studies have been focused on
short-term high light stress in lichens under laboratory conditions. Such studies reported a strong photoinhibition of photosynthesis accompanied by a partial photodestruction of PSII, involvement of photoprotective mechanisms, and resynthetic processes into gradual recovery. In our paper, we applied medium [800 μmol(photon) m-2 s-1] light stress to induce negative changes in PSII funcioning as well as pigment and glutathione (GSH) content in two Antarctic fruticose lichen species. Chlorophyll (Chl) fluorescence parameters, such as potential and effective quantum yield of photosynthetic processes and fast transients (OJIP) recorded during high light exposition and recovery, revealed that Usnea antarctica was less susceptible to photoinhibition than U. aurantiaco-atra. This might be supported by a more pronounced high light-induced reduction in Chl a and b contents in U. aurantiaco-atra compared with U. antarctica. In both experimental species, total GSH showed an initial increase during the first 30-40 min of high light treatment followed by a decrease (60 min) and an increase during dark recovery. Full GSH recovery, however, was not finished in U. aurantiaco-atra even after 5 h indicating lower capacity of photoprotective mechanisms in the species. OJIP curves showed high light-induced decrease in both species, however, the recovery of the OJIPs shape to pre-photoinhibitory values was faster and more apparent in U. antarctica than in U. aurantiaco-atra. PSI trimer to monomer ratio in intact cyanobacterial cells and isolated thylakoids was analysed by two noninvasive, in vivo methods; low-temperature fluorescence emission and circular dichroism spectroscopy. We measured fluorescence emission spectra of cells upon chlorophyll (Chl, 436 nm) excitation. All three species - Synechocystis sp. PCC 6803, Anabaena sp. PCC 7120, and Spirulina platensis - showed shifted Chl peak, indicating they have different spectral properties. CD spectroscopy revealed the highest intensity at 515 nm (PSI peak) in Spirulina platensis cells, which may originate from PSI multi-oligomerisation. The most sensitive response to heat treatment in this strain was the oligomerisation of PSI RCs. PSI dimers and tetramers in Anabaena cells showed smaller changes of the CD signal upon the heat treatment compared to that of Synechocystis WT. The acclimation to high light, elevated temperature, and combination of both factors was evaluated in tomato (Solanum lycopersicum cv. M82) by determination of photochemical activities of PSI and PSII and by analyzing 77 K fluorescence of isolated thylakoid membranes. Developed plants were exposed for six days to different combinations of temperature and light intensity followed by five days of a recovery period. Photochemical activities of both photosystems showed different sensitivity towards the heat treatment in dependence on light intensity. Elevated temperature exhibited more negative impact on PSII activity, while PSI was slightly stimulated. Analysis of 77 K fluorescence emission and excitation spectra showed alterations in the energy distribution between both photosystems indicating alterations in light-harvesting complexes. Light intensity affected the antenna complexes of both photosystems stronger than temperature. Our results demonstrated that simultaneous action of high-light intensity and high temperature promoted the acclimation of tomato plants regarding the activity of both photosystems in thylakoid membranes., A. Ca2+ has been considered as a necessary ion for alleviation of stress-induced damages in plants. We investigated effects of exogenous Ca2+ on waterlogging-induced damage to pepper and its underlying mechanisms. Pepper seedlings under stress were treated by spraying of 10 mM CaCl2. Applying exogenous Ca2+ increased the biomass of pepper leaves and roots, improved photosynthetic characteristics, membrane permeability, root activity, osmotic substance contents, antioxidant enzyme and alcohol dehydrogenase activities, while it reduced lactate dehydrogenase activity. It maintained hydroxide radical contents and activities of malate dehydrogenase and succinate dehydrogenase relatively high. Light-emitting diodes (LEDs) are a promising technology with a potential to improve the irradiance efficiency, light quality, and the light spectrum for increasing plant yield and quality. In this experiment, we investigated the impacts of various LED light qualities, including 100% red, 100% blue, 70% red + 30% blue, and 100% white, on the growth and photosynthesis, phytochemical contents, and mineral element concentrations in lettuce (Lactuca sativa L. cv. 'Grizzly') in comparison to normal greenhouse conditions. Photon flux of 300 µmol m-2 s-1 was provided for 14 h by 120 LEDs set on a 60 cm × 60 cm sheet of aluminum platform in the growth chambers, where plants were grown for 60 d. Fresh mass per plant was significantly higher when grown under 100% blue and 70% red + 30% blue LEDs compared to the other environments including greenhouse conditions. Phytochemical concentrations and a nutritive value of lettuce were also significantly affected by the light treatments. Chlorophyll and carotenoid concentrations increased in the plants grown under 70% red + 30% blue LEDs compared to those grown in the greenhouse. Vitamin C content was 2.25-fold higher in the plants grown under 100% blue LEDs compared to those grown in the greenhouse. Higher photosynthesis and maximal quantum yield of PSII photochemistry were also observed in the plants treated with LED lights. The application of LED light led to the elevated concentrations of macro-and micronutrients in lettuce possibly because of the direct effect of LED light and lower stress conditions in the growth chambers compared to the greenhouse. This paper reports effects of ultraviolet B (UVB) radiation on leaf anatomy and contents of chlorophyll and carotenoids, as well as photosynthetic parameters, in young sporophytes of Acrostichum danaeifolium Langsd. & Fisch. (Polypodiopsida, Pteridaceae) exposed to UV radiation treatments for 1 h daily for six weeks. The leaves showed large aerenchyma and present chloroplasts in both epidermises. After cultivation under PAR + UVA + UVB, leaves showed curling and malformed stomata on the abaxial face. After the UV treatment, chloroplasts in leaves were arranged against the inner wall of the epidermal cells. Transmission electron microscopy analysis showed some dilated thylakoids and plastoglobuli in chloroplasts and vesicles containing phenolic compounds in the cytoplasm. Differences were not observed between control and UV-treated plants in their contents of chlorophylls, carotenoids, and photosynthetic parameters. We conducted an experiment to assess the predictive capability of a leaf optical meter for determining leaf pigment status of Acer mono Maxim., A. ginnala Maxim., Quercus mongolica Fisch., and Cornus alba displaying a range of visually different leaf colors during senescence. Concentrations of chlorophyll (Chl) a, Chl b, and total Chl [i.e., Chl (a+b)] decreased while the concentration of carotenoids (Car) remained relatively static for all species as leaf development continued from maturity to senescence. C. alba exhibited the lowest average concentration of Chl (a+b), Chl a, and Car, but the highest relative anthocyanin concentration, while Q. mongolica exhibited the highest Chl (a+b), Chl b, and the lowest relative anthocyanin concentration. A. mono exhibited the highest Chl a and Car concentrations. The relationships between leaf pigments and the values measured by the optical meter generally followed an exponential function. The strongest relationships between leaf pigments and optical measurements were for A. mono, A. ginnala, and Q. mongolica (R2 ranged from 0.64 to 0.95), and the weakest relationships were for C. alba (R2 ranged from 0.13 to 0.67). Moreover, optical measurements were more strongly related to Chl a than to Chl b or Chl (a+b). Optical measurements were not related to Car or relative anthocyanin concentrations. We predicted that weak relationships between leaf pigments and optical measurements would occur under very low Chl concentrations or under very high anthocyanin concentrations; however, these factors could not explain the weak relationship between Chl and optical measurements observed in C. alba. Overall, our results indicated that an optical meter can accurately estimate leaf pigment concentrations during leaf senescence - a time when pigment concentrations are dynamically changing - but that the accuracy of the estimate varies across species., Future research should investigate how species-specific leaf traits may influence the accuracy of pigment estimates derived from optical meters., G. The source-sink relationship is one of major determinants of plant performance. The influence of reproductive sink demand on light-saturated photosynthesis (Pmax), dark respiration (RD), stomatal conductance (gs), intrinsic water-use efficiency (WUEi), contents of soluble sugar (SSC), nitrogen, carbon, and photosynthetic pigments was examined in blueberry (Vaccinium corymbosum L. cv. ‘Brigitta’) during the final stage of rapid fruit growth. Measurements were performed three times per day on developed, sun-exposed leaves of girdled shoots with 0.1, 1, and 10 fruit per leaf (0.1F:L, 1F:L, and 10F:L, respectively) and nongirdled shoots bearing one fruit per leaf (NG). Girdling and lower fruit amount induced lower Pmax, gs, N, and total chlorophyll (Chl) and higher WUEi, SSC, RD, Chl a/b ratio and carotenoids-to-chlorophylls ratio (Car/Chl) for the 1F:L and 0.1F:L treatments. The impact of girdling was counterbalanced by 10F:L, with NG and 10F:L having similar values. Variables other than Pmax, RD, gs, WUEi, and SSC were unaffected throughout the course of the day. Pmax and gs decreased during the course of the day, but gs decreased more than Pmax in the afternoon, while WUEi was increasing in almost all treatments. SSC increased from the morning until afternoon, whereas RD peaked at noon regardless of the treatment. Generally, Pmax was closely and negatively correlated to SSC, indicating that sugar-sensing mechanisms played an important role in regulation of blueberry leaf photosynthesis. With respect to treatments, Pmax and N content were positively related, while RD was not associated to substrate availability. The enhanced Car/Chl ratio showed a higher photoprotection under the lower sink demand. Changes in the source-sink relationship in 'Brigitta' blueberry led to a rearrangement of physiological and structural leaf traits which allowed adjusting the daily balance between carbon assimilation and absorbed light energy., E.
