The Czech BIORAF project employs the techniques of green chemistry for biomass utilization to high added-value products and energy sources. Refining it results in food supplements, fodders and fertilizers, new-generation biofuels and energy from the biomass of microbial, plant or animal origin. Metal stress was induced in maize (Zea mays L.) by the addition to the soil of a range of concentrations of either ethylene-diamine-tetra-acetate (EDTA) or citric acid (CA) as chelating agents. Measurements were taken using a recently-developed sensor capable of plant fluorescence detection at wavelengths of 762 and 688 nm. Atmospheric oxygen absorbs radiation at these wavelengths. As such, measured fluorescence can be attributed to the plants under observation. Red/far-red (690/760 nm, R/FR) chlorophyll (Chl) fluorescence ratios were measured before addition of the chelating agents and during the month following. Significant differences were seen in the fluorescence responses of those plants for which high concentrations [≥ 30 mmol kg-1(d.m. soil)] of EDTA were added to the pots compared to those for which CA or no chelating agent was added. The plants for which high concentrations of EDTA were added also exhibited higher tissue metal concentrations and demonstrated visible signs of stress. Before signs of visual stress became apparent, R/FR Chl fluorescence ratios for metal-stressed plants were significantly different to those observed for unstressed plants. The objective of this study was to investigate the relative salt tolerance of four eggplant cultivars (Solanum melongena L.) by studying chlorophyll (Chl) fluorescence parameters during the vegetative growth stage under increasing salinity levels. The plants were grown in pots filled with peat under controlled conditions and were subjected to the salt stress ranging from 0 (control), 20, 40, 80, and 160 mM NaCl for 25 days. The results showed that the increasing NaCl concentration affected hardly the maximum quantum yield of photosystem (PS) II. The quantum yield of PSII (ΦPSII) decreased significantly in ‘Adriatica’ and ‘Black Beauty’ under the salt stress. The photochemical quenching decreased in ‘Black Beauty’ and nonphotochemical quenching increased in ‘Adriatica’ under the salt stress. The Chl fluorescence parameters did not change significantly under the salt stress in ‘Bonica’ and ‘Galine’, revealing their tolerance to salinity. After 25 days of the salt stress, the plant growth was reduced in all cultivars, however, this decline was more pronounced in ‘Adriatica’ and ‘Black Beauty’. Additionally, a significant correlation between the biomass and ΦPSII was observed in ‘Adriatica’ and ‘Black Beauty’. Saline soils spread wildly in the world, therefore it is important to develop salt-tolerant crops. We carried out a pot study in order to determine effects of arbuscular mycorrhizal fungi (AMF) (Rhizophagus irregularis and Glomus versiforme) in black locust seedlings under salt (NaCl) stress. The results showed that AMF enhanced in seedlings their growth, photosynthetic ability, carbon content, and calorific value. Under salt stress, the biomass of the seedlings with R. irregularis or G. versiforme were greater by 151 and 100%, respectively, while a leaf area increased by 197 and 151%, respectively. The seedlings colonized by R. irregularis exhibited a higher chlorophyll content, net photosynthetic rate, intercellular CO₂ concentration, stomatal conductance, and transpiration rate than that of the nonmycorrhizal seedlings or those colonized by G. versiforme. Both R. irregularis and G. versiforme significantly enhanced a carbon content, calorific value, carbon, and energy accumulations of black locust under conditions of 0 or 1.5 g(NaCl) kg-1(growth substrate). To investigate the effects of atmospheric CO2 enrichment on physiology and autumnal leaf phenology, we exposed 3-year-old sugar maple (Acer saccharum Marsh.) seedlings to 800 (A8), 600 (A6), and 400 μL(CO2) L-1 (AA) in nine continuous stirred tank reactor (CSTR) chambers during the growing season of 2014. Leaf abscission timing, abscised leaf area percentages, leaf number, light-saturated net photosynthetic rate (PNmax), leaf area, accumulative growth rates, and biomass were determined and assessed. The results suggested the following: (1) no significant differences were found in the timing of leaf abscission in the three CO2-concentration treatments; (2) PNmax was continuously stimulated to the greatest extent in A8 at 319% and 160% in A6 until the end of the growing season, respectively; and (3) leaf number, leaf area, and accumulative height growth all significantly increased by elevated CO2, which led to a 323% increase in A8 biomass and 235% in A6 biomass after 156-d fumigation. In summary, the results suggest, the timing of leaf abscission of sugar maple in fall was not modified by CO2 enrichment, the increased carbon gain by elevated CO2 was mainly due to increased leaf area, more leaves, and the continuously enhanced high photosynthesis throughout the growing season instead of the leaf life span., L. Chloroplasts are commonly the site of the earliest abiotic injury visible in plant ultrastructure. In this study, six inbred lines of maize (Zea mays L.) were used to analyze changes in the ultrastructure of chloroplasts and related physiological parameters under conditions of drought stress simulated by 20% polyethylene glycol 6000 (-0.6 MPa) for two days. Chloroplasts of three maize lines proved to be more sensitive. They showed changes in the ultrastructure in response to drought, including damage of thylakoid membranes, an increase in the number and size of plastoglobuli, swelling of thylakoid membranes both stromal and granal, disorganization of the thylakoid membrane system, an obvious increase in the intrathylakoid space, and a decrease in the
length-to-width ratio and area of chloroplasts. In addition, the contents of malondialdehyde increased markedly in the sensitive lines. Contrary to the sensitive lines, stable structures and shapes of chloroplasts were observed in the drought-resistant lines; it could be considered as an advantage contributing to drought tolerance in the plants. In addition, the drought index of leaf fresh mass (LMDI) in the drought-sensitive lines was ≤ 0.5, which was also associated with a lower content of leaf chlorophyll. Fifty-d-old poplar (Populus deltoides L.) plants were irrigated with 50-200 mM NaCl. 100 and 200 mM NaCl significantly reduced net photosynthetic rate, chlorophyll and carotenoid contents, leaf area, dry matter accumulation, and harvest index (HI) in all tested poplar clones (Bahar, S7C15, and WSL22). The combined effects of UV-B irradiation and foliar treatment with selenium on two buckwheat species, common (Fagopyrum esculentum Moench) and tartary [Fagopyrum tataricum (L.) Gaertn.] buckwheat, that underwent different intensity of breeding, were examined. Plants grown outdoors under three levels of UV-B radiation were studied for 9 weeks, from sowing to ripening. At week 7 they were sprayed with solution containing 1 g(Se) m-3 that presumably mitigates UV-B stress. Morphological, physiological, and biochemical parameters of the plants were monitored. Elevated UV-B radiation, corresponding to a 17 % reduction of the ozone layer, induced synthesis of UV absorbing compounds. In both buckwheat species it also caused a reduction in amounts of chlorophyll a during the time of intensive growth, an effect, which was increased in tartary buckwheat in the presence of selenium. The respiratory potential, measured as terminal electron transport system activity, was lower in plants subjected to enhanced UV-B radiation during the time of intensive growth. The effective quantum yield of photosystem 2 was also reduced due to UV-B radiation in both buckwheat species and was mitigated by the addition of Se. We conducted a hydroponic experiment in order to study effects of the ammonium/nitrate ratio (0:15, 5:10, 7.5:7.5, and 10:5) on photosynthetic characteristics and biomass accumulation in Brassica chinensis under low light intensity and water stress. Results showed that net photosynthetic rate, transpiration rate, intrinsic water-use efficiency, stomatal conductance, intercellular CO2 concentration, effective quantum yield of PSII photochemistry, electron transport rate, and nonphotochemical quenching were lower in the treatment (low light intensity and water deficit) than those in the control, whereas stomatal limitation increased. Minimum fluorescence, maximal quantum yield of PSII photochemistry, and photochemical quenching were largely unchanged. Pigment contents first increased and then decreased as the ammonium/nitrate ratios were altered, with significant differences between treatment and control observed at all ratios except for 10:5. Biomass first increased slightly and then decreased both in treated and control plants. Radiation quality was an important environmental cue to stimulate seed germination in Acacia mangium. The photo-synthetic CO2 assimilation rate, dark respiration rate, total biomass, and relative growth rate of seedlings grown under monochromatic radiation were significantly lower than those of seedlings grown under full spectrum radiation.
