The general population is potentially exposed to many chemicals that can affect the endocrine system. These substances are called endocrine disruptors (EDs), and among them bisphenol A (BPA) is one of the most widely used and well studied. Nonetheless, there are still no data on simultaneous measurements of various EDs along with steroids directly in the seminal fluid, where deleterious effects of EDs on spermatogenesis and steroidogenesis are assumed. We determined levels of BPA and 3 estrogens using LC-MS/MS in the plasma and seminal plasma of 174 men with different degrees of infertility. These men were divided according their spermiogram values into 4 groups: (1) healthy men, and (2) slightly, (3) moderate, and (4) severely infertile men. Estradiol levels differed across the groups and body fluids. Slightly infertile men have significantly higher BPA plasma and seminal plasma levels in comparison with healthy men (p<0.05 and p<0.01, respectively). Furthermore, seminal BPA, but not plasma BPA, was negatively associated with sperm concentration and total sperm count (-0.27; p<0.001 and -0.24; p<0.01, respectively). These findings point to the importance of seminal plasma in BPA research. Overall, a disruption of estrogen metabolism was observed together with a weak but significant impact of BPA on sperm count and concentration., J. Vitku, L. Sosvorova, T. Chlupacova, R. Hampl, M. The present study examined the hypothesis that the extension of noxious effect of methamphetamine (MA) on maternal behavior and postnatal development on the pups may differ in dependence with time of application. Female rats were injected with MA (5 mg/kg) or saline during first (embryonic day (ED) 1-11) or second (ED 12-22) half of gestation. Our results demonstrated that MA exposure on ED 12-22 led to decreased birth weight and weight gained during lactation period relative to rats treated on ED 1-11. Both sexes treated prenatally with MA on ED 1-11 opened eyes earlier compared to animals treated on ED 12-22. As a matter of sensorimotor development application of MA on ED 1-11 impaired the righting reflex, while MA exposure on ED 12-22 impaired the performance of beam balance test in male rats. There were no differences in maternal behavior. Therefore, it seems that MA exposure in the first half of the gestation impaired the early sensorimotor development that is under control of the brain stem, while the MA exposure in the second half of gestation affected the beam balance performance that is dependent on the function of the cerebellum., M. Type I diabetes mellitus (DM1) is a complex disease with adverse effects on organs and tissues despite compensation by insulin treatment. The goal of our study was to study how kidney diseases change 31P MR parameters of muscle metabolism in DM1 patients with respect to gender. 51 DM1 patients (19 m/14 f without and 13 m/5 f with nephropathy) and 26 (14 m/12 f) healthy volunteers were examined using 31P magnetic resonance spectroscopy at 3T tomograph at rest, and during and after a calf muscle exercise. The exercise consisted of a six-minute plantar flexion using a pedal ergometer followed by a six-minute recovery. It is reflected by reduced relative β-ATP and increased Pi and phosphodiester signals to phosphocreatine (PCr) at rest and prolongation of the PCr recovery time after the exercise. Measurement on healthy volunteers indicated differences between males and females in pH at the rest and after the exercise only. These differences between patients groups were not significant. We have proven that nephropathy affects the metabolism in diabetic patients and our results confirm significant difference between patients with and without nephropathy. Gender differences in pH were observed only between male and female healthy volunteers., P. Sedivy, M. Dezortova, M. Drobny, Z. Vlasakova, V. Herynek, M. Hajek., and Obsahuje bibliografii
The effects of continuous soil hypoxia on stomatal conductance (g^ and net photosynthetic rate (P^) ů* seedlings of Taxodium disíichum, Quercus lyrata, and Q. falcata var. pagodaefolia were studied under controlled environment. Soil oxygen deficiency induced significant stomatal closure and reduction of in oak species within 1-3 d. This response pattem continued resulting in average daily values of g^ reduced 85 and 40 % within 14 d in g. falcata and Q. lyrata as compared to control plants, respectively. Ehiring the same period Pn reduced 96 and 71 % in 0. falcata and Q. lyrata as compared to control plants, respectively. In T. distichum, however, gg and Pff were reduced 18 and 33 % by day 8. Significant recoveiy of gg and was noted in T. distichum. By day 14, gg had recovered to 91 % and to 92 % of control plants. In oak species, however, remained significantly lower than in control plants without any apparent recovery. The regain of photosynthetic activity and stomatal functioning in flood-tolerant species appears to be an important flood- tolerance characteristic allowing these species to fimction under flooded soil.
Lianas perform better than co-occurring trees in secondary forests or disturbed areas. Lianas and trees differ strikingly in water use strategy, which may result in a significant difference in photosynthetic light use between both growth forms. However, the difference in the photosynthetic efficiency and light energy dissipation between these two growth forms is poorly understood. Moreover, photorespiration is an important mechanism of photoprotection under conditions of high light. In this study, we used Bridelia stipularis (Linn.) Bl. (liana) and Strophioblachia fimbricalyx Boerl. (tree) in order to measure the response curves of the gas exchange and photosynthetic electron flow to the incident light gradients and intercellular CO2 concentration, as well as the hydraulic conductivity. We tested whether the photochemical efficiency and photorespiration differed between both growth forms. Our results clearly demonstrated that B. stipularis possessed a significantly higher stem and leaf specific hydraulic conductivity, total electron flow, and maximum rate of ribulose-1,5-bisphosphate regeneration compared to the sympatric tree S. fimbricalyx. Correspondingly, B. stipularis exhibited a significantly higher photochemical quenching coefficient and electron flow to photorespiration relative to S. fimbricalyx under saturating light levels. We suggested that photorespiration might play an important role in photoprotection for both species under high light, but particularly for B. stipularis. These findings could enrich our knowledge of the superior photosynthetic and growth performance of lianas over the co-occurring trees., S.-B. Zhang, J.-L. Zhang, K.-F. Cao., and Seznam literatury
a1_Previous studies have focused mainly on the accumulation of photosynthates and less on their distribution in sweet potato (Ipomoea batatas L.). In addition, the effect of photosynthate accumulation in root tubers on photosynthate distribution was not considered. Thus, a field experiment was carried out from May to October (2011 and 2012) to clarify the differences in photosynthate transport between high- and low-yielding sweet potato. This study mainly focused on the photosynthetic capacities of leaves, photosynthate distribution, and characteristics of photosynthate accumulation in root tubers. Results showed the high-yielding varieties displayed the higher fresh root tuber yield and the economic coefficient than the low-yielding varieties. They also showed greater net photosynthetic rate with a pronounced increase at the early and middle growth stages (8.9% and 11.4%, respectively). After the growth peak, the leaf area index (LAI) of the high-yielding varieties decreased with time and was maintained at 2~3 until harvest, whereas the LAI of the low-yielding varieties decreased slowly. The high-yielding varieties reached the 13C distribution rate ≥ 50% at the early (2011, 2012) and middle (2011) growth stages, whereas the low-yielding varieties reached it at the late (2011) or middle (2012) growth stages. At harvest, the 13C distribution rates in the branches and root tubers of the
high-yielding varieties were 6.0-20.3% and 73.7-91.2%, respectively, whereas those of the low-yielding varieties were 29.6-34.7% and 60.7-63.5%, respectively. The high-yielding varieties showed the remarkable initial potential in root tubers, which was much better than that of the low-yielding varieties. The high-yielding varieties also produced heavier root tubers and the higher number of root tubers per plant at the early bulking stage., a2_The root tubers also attained the greater content of soluble sugar and starch. The high-yielding varieties formed root tubers earlier, showed strong abilities to transport photosynthates into the root tubers, and exhibited a higher mean accumulation rate. These varieties could also reduce the photosynthate consumption in branch leaves and stems. Therefore, the high-yielding varieties established growth advantage for the root tubers earlier. It contributed to a reasonable distribution structure of photosynthates that led to the high root tuber yield. Based on our results, effective agricultural measures can be chosen to improve the root tuber yield of sweet potato., H. J. Liu, S. S. Chai, C. Y. Shi, C. J. Wang, G. B. Ren, Y. Jiang, C. C. The aim of this study was to investigate the reaction of the hypothalamo-pituitary-adrenocortical (HPA) system to various stressors (fasting, crowding, cold and heat) by measuring blood ACTH and corticosterone (CORT) concentration as well as the cholesterol (CHOL) content in the adrenals. To examine the effects of stress termination, the rats were returned and kept under control conditions for the same period as that of stress duration (supposed recovery period). According to our results HPA system was activated by all the stressors applied. Heat seems to be the strongest stressor since the exposure of animals to a high ambient temperature resulted in the greatest rise of plasma ACTH concentration as well as CORT synthesis and secretion. These values remained elevated after the stress termination i.e. after the rats had been returned to room temperature. Fasting seems to be the weakest stressor given because it causes the smallest increase in blood ACTH and CORT concentrations. Moreover, in refed rats the HPA function was fully recovered. In conclusion, the various stressors applied seem to induce a different response of the HPA system as judged by quantitative changes in ACTH and CORT release., J. Djordjević, G. Cvijić, V. Increased oxidative stress is indisputably an important mechanism of doxorubicin side effects, especially its cardiotoxicity. To prevent impairment of non-tumorous tissue and to improve the specificity in targeting the tumor tissue, new drug nanotransporters are developed. In many cases preclinical therapeutic advantage has been shown when compared with the administration of conventional drug solution. Three forms of doxorubicin - conventional (DOX), encapsulated in liposomes (lipoDOX) and in apoferritin (apoDOX) were applied to Wistar rats. After 24 h exposition, the plasma level of 4-hydroxy-2- nonenal (4-HNE) as a marker of lipoperoxidation and tissue gene expression of thioredoxin reductase 2 (TXNRD2) and aldehyde dehydrogenase 3A1 (ALDH3A1) as an important part of antioxidative system were determined. Only conventional DOX significantly increases the level of 4-HNE; encapsulated forms on the other hand show significant decrease in plasma levels of 4-HNE in comparison with DOX. They also cause significant decrease in gene expression of ALDH3A1 and TXNRD2 in liver as a main detoxification organ, and a mild influence on the expression of these enzymes in left heart ventricle as a potential target of toxicity. Thus, 4-HNE seems to be a good potential biomarker of oxidative stress induced by various forms of doxorubicin., M. Hlaváčová, J. Gumulec, T. Stračina, M. Fojtů, M. Raudenská, M. Masařík, M. Photosynthetic light curve, chlorophyll (Chl) content, Chl fluorescence parameters, malondialdehyde (MDA) content, phosphoenolpyruvate carboxylase (PEPC) activity and reactive oxygen metabolism were studied under drought stress in two autotetraploid rice lines and corresponding diploid rice lines. Net photosynthetic rate decreased dramatically, especially under severe drought stress and under high photosynthetic active radiation in diploid rice, while it declined less under the same conditions in autotetraploid lines. Compared with the corresponding diploid lines, the Chl content, maximum photochemical efficiency of photosystem (PS) II, and actual photochemical efficiency of PSII were reduced less in autotetraploid lines. PEPC activities were higher in autotetraploid rice lines. PEPC could alleviate inhibition of photosynthesis caused by drought stress. The chromosome-doubling enhanced rice photoinhibition tolerance under drought stress. The lower MDA content and superoxide anion production rate was found in the autotetraploid rice indicating low peroxidation level of cell membranes. At the same time, the superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities were higher in autotetraploid rice lines. SOD, POD, and CAT could effectively diminish the reactive oxygen species and reduced the membrane lipid peroxidation., P.-M. Yang, Q.-C. Huang, G.-Y. Qin, S.-P. Zhao, J.-G. We evaluated the effects of N G -nitro-L-arginine methylester (L-NAME) (50 mg/kg/day) and 7-nitroindazole (7NI) (10 mg/kg/day) administered from 10th-16th week of age either individually or together on cardiovascular system of Wistar rats and SHR. Systolic blood pressure (sBP) was measured weekly by the plethysmographic method. For morphological studies, the animals (n=10) were perfused with a fixative (120 mm Hg), and thoracic aorta and carotid and co ronary arteries were processed for electron microscopy. For functional investigation (n=10), aortic rings were used in an or gan bath. In Wistar rats, L-NAME evoked an increase of sBP; hype rtrophy of the heart and arterial walls; an increase in cross-sectional areas (CSA) of endothelial cells (EC), muscle cells (SMC), extracellular matrix (ECM), and a decrease in acetylcholin e-induced endothelial-dependent relaxation (EDR). 7NI evoked sBP-independent hypotrophy of the heart and arterial walls, a decrease in CSA of EC and SMC without affecting the CSA of ECM, and a mild decrease in acetylcholine-induced EDR. 7NI and L-NAME administered together evoked lower effect on BP and trophicity of the heart and all arteries, and a similar de crease in acetylcholine-induced EDR compared to L-NAME alone. In SHR, 7NI did not evoke any effect on the studied parameters., F. Kristek, M. Drobna, S. Cacanyiova., and Obsahuje bibliografii
