Winter wheat (Triticum aestivum L.) cultivars Yangmai 9 (water-logging tolerant) and Yumai 34 (water-logging sensitive) were subjected to water-logging (WL) from 7 d after anthesis to determine the responses of photosynthesis and anti-oxidative enzyme activities in flag leaf. At 15 d after treatment (DAT), net photosynthetic rate under WL was only 3.7 and 8.9 µmol(CO2) m-2 s-1 in Yumai 34 and Yangmai 9, respectively, which was much lower than in the control. Ratios of variable to maximum and variable to initial fluorescence, actual photosynthetic efficiency, and photochemical quenching were much lower, while initial fluorescence and non-photochemical quenching were much higher under WL than in control, indicating damage to photosystem 2. WL decreased activities of superoxide dismutase and catalase in both cultivars, and activity of peroxidase (POD) in Yumai 34, while POD activity in Yangmai 9 was mostly increased. Osteosarcoma (OS), a severe malignant bone tumour, usually occurs in adolescents and children and has a poor prognosis. Asiatic acid (AA), an active component isolated from Centella asiatica (L.) Urb., exhibits appreciable anti-oxidant and anti-tumour activities. So far, the effects and underlying mechanisms of AA against OS have not been clarified. Here, we explored the anti-tumour effects of AA against human OS and the involved mechanism mediating its actions. To evaluate effects of AA on the cell proliferation of human OS cells, cell viability and colony formation assays were performed. Flow cytometry was used to evaluate apoptosis in OS cells exposed to AA and mitochondrial membrane potential. Western blotting and RT-PCR were applied to determine expression of the relevant proteins and their mRNA levels. Our explorations showed that AA inhibits proliferation of human OS cells in a concentration- and time-dependent manner, and induces apoptosis of OS cells by the intrinsic (mitochondrial) pathway. Importantly, we found that inhibition of the AA-induced phosphorylation of JAK2/STAT3 signalling molecules and the decrease in MCL-1 contributed to the anti-tumour efficacy of AA. Collectively, our results suggest that AA could evoke mitochondrial- induced apoptosis in human OS cells by suppression of the JAK2/STAT3 pathway and MCL-1 expression. These results strongly demonstrate that AA could be a potential anti-tumour agent for OS treatment.
In order to understand better Cd resistance in soybean, Dongying wild soybean treated with different Cd concentrations were evaluated. The biomass, chlorophyll (Chl) content, leaf color, Chl a fluorescence parameters, photosynthesis parameters, and Cd contents were determined. Our results showed that when Cd concentration was ≤ 2 kg m-3, no significant decrease in biomass, photosynthetic parameters, and maximal photochemical efficiency of PSII was observed. This indicated that Dongying wild soybean resisted Cd toxic effects under such conditions. In addition, atomic absorption experiment results demonstrated that when Cd concentration was ≤ 0.5 kg m-3, the accumulation of Cd in wild soybean was lower in roots than that in shoots, while the accumulation of Cd was higher in roots than that in shoots when Cd concentration was ≥ 1 kg m-3. We compared the effects of salt-stresses (SS, 1: 1 molar ratio of NaCl to Na2SO4) and alkali-stresses (AS, 1: 1 molar ratio of NaHCO3 to Na2CO3) on the growth, photosynthesis, solute accumulation, and ion balance of barley seedlings, to elucidate the mechanism of AS (high-pH) damage to plants and the physiological adaptive mechanism of plants to AS. The effects of SS on the water content, root system activity, membrane permeability, and the content of photosynthetic pigments were much less than those of AS. However, AS damaged root function, photosynthetic pigments, and the membrane system, led to the severe reductions in water content, root system activity, content of photosynthetic pigments, and net photosynthetic rate, and a sharp increase in electrolyte leakage rate. Moreover, with salinity higher than 60 mM, Na+ content increased slowly under SS and sharply under AS. This indicates that high-pH caused by AS might interfere with control of Na+ uptake in roots and increase intracellular Na+ to a toxic level, which may be the main cause of some damage emerging under higher AS. Under SS, barley accumulated organic acids, Cl-, SO4 2-, and NO3 - to balance the massive influx of cations, the contribution of inorganic ions to ion balance was greater than that of organic acids. This study investigated whether each part of the heart is evenly innervated by the left or right vagus and observed the mechanism of compensatory recovery after unilateral cervical vagotomy. HR, BP, LVSP and ±dp/dt max all decreased one week after left vagotomy, whereas only BP and -dp/dt max decreased one week after right vagotomy. We stern blot analyses revealed that the expression of M2 receptors in the left atrium and left ventricle was upregulated after subacute (1 week) left/right vagotomy. However, significantly more cholinesterase-positive nerves in LV and RV were seen one week after unilateral vagotomy compared to the sham-operated group. In addition, baroreflex sensitivity was increased after subacute right vagotomy. The decreasing effects of ACh (0.5 μ g/kg) on LVSP and ±dp/dt max (but not on HR and BP) were facilitated by subacute unilateral vagotomy. Our present experiments indicate that 1) the working myocardium is innervated bilaterally by the vagus, 2) ventricular contractility is influenced more by denervation of the left than the right vagus and 3) up-regulation of M2 muscarinic receptors in the left heart, increase of cholinergic nerves, and high baroreflex sensitivity could be involved in the mechanism of compensatory hemodynamic recovery via contralateral vagus overactivity, thereby amplifying contralateral vagal activity and decreasing cardiac contractility., L. Competition plays an important role in the replacement of native species by alien plants. A greenhouse experiment was conducted to investigate whether the competition pattern of alien Robinia pseudoacacia L. and native Quercus acutissima Carr. is affected by soil sterilization. Physiological traits, such as gas-exchange parameters and chlorophyll (Chl) content, and growth traits, such as the biomass accumulation of the two species, were examined in natural soil or in soil sterilized with benomyl. The results show that native Q. acutissima inhibits the growth of R. pseudoacacia in natural soil. When the two plants coexisted and competed under sterilization treatment, R. pseudoacacia was less inhibited by Q. acutissima and the competition of R. pseudoacacia decreased the growth of Q. acutissima in terms of biomass, Chl a, Chl b, total Chl, and Chl a/b. These results suggest that soil sterilization benefits the growth of R. pseudoacacia and changes the competition pattern by the changed soil biota. Soil sterilization increased the biomass of root nodules, which ultimately benefits the growth of R. pseudoacacia and root nodule bacteria may be important in the dispersal and invasion process of nitrogen-fixing alien plants such as R. pseudoacacia., H. Effects of root treatment with 5-aminolevulinic acid (ALA) on leaf photosynthesis in strawberry (Fragaria ananassa Duch.) plants were investigated by rapid chlorophyll fluorescence and modulated 820 nm reflection using 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU) and methyl viologen (MV). Our results showed that ALA treatments increased the net photosynthetic rate and decreased the intercelluar CO2 concentration in strawberry leaves. Under DCMU treatment, trapping energy for QA reduction per PSII reaction center increased greatly, indicating DCMU inhibited electron transfer from QA−. The maximum photochemical efficiency of PSII (Fv/Fm) decreased under the DCMU treatment, while a higher Fv/Fm remained in the ALA-pretreated plants. Not only the parameters related to a photochemical phase, but also that one related to a heat phase remained lower after the ALA pretreatment, compared to the sole DCMU treatment. Two species with different resistances to alkaline pH, the glycophylic Triticum aestivum (wheat) and the halophilic Chloris virgata, were chosen as test organisms. The salt-alkaline (SA) mixed stress conditions with different buffer capacities (BC) but with the same salt molarities and pH were established by mixing neutral (NaCl, Na2SO4), and alkaline salts (NaHCO3 and Na2CO3) in various proportions. Growth, photosynthetic characteristics, and solute accumulation of the seedlings were monitored to test the validity of BC as a decisive index of alkali-stress (AS) intensity in SA mixed stress. At the same salinities and pHs, the relative growth rate, the content of photosynthetic pigments, and net photosynthetic rates of wheat and C. virgata decreased, while Na+ content and Na+/K+ ratios in shoots increased with increasing BC. Hence BC was a true measure of AS intensity at mixed SA stress and the alkali-resistance mechanism of plants was easy to interpret. A greenhouse experiment examined whether clonal integration improves photosynthesis of ramets of alligator weed [Alternanthera philoxeroides (Mart.) Griseb.], a widespread invasive clonal plant in China, in heterogeneous (He) nutrient habitats. The connected pairs of ramets experienced different nutrient levels [high homogeneous (Ho) nutrient, low Ho nutrient, and two He nutrient treatments]. Clonal integration significantly improved the net photosynthetic rate, stomatal conductance, transpiration rate, and minimal and maximal chlorophyll fluorescence of ramets of alligator weed in low nutrient condition. These characteristics may contribute to the success of the ramets of alligator weed in invading contrasting habitats. Regulatory volume decrease (RVD) is essential for the survival of animal cells. The aim of this study was to observe the RVD process in rat ventricular myocytes, and to determine if the KATP channels are involved in the RVD process in these cells. By using reverse transcriptase polymerase chain reaction and Western blot analysis, we demonstrated that there are two types of KATP channels expressed in rat ventricular myocytes: Kir6.1 and Kir6.2. When rat cardiac myocytes were exposed to hypotonic solution, cell volume increased significantly within 15 min and then gradually recovered. This typical RVD process could be inhibited by a Cl– channel blocker (0.5 mM 9-anthracene-carboxylic acid , 9-AC), a K+ channel blocker (5.0 mM CsCl) and a KATP channel blocker glibenclamide (10 μM). Electrophysiological results showed that hypotonic solution activated a whole-cell current, which had similar biophysical characteristics with KATP opener (pinacidil)-induced currents. This current could be blocked by glibenclamide.
