Chrysanthemum plantlets were cultivated in vitro on media with 2.0, 0.3, or 0 % sucrose, or photoautotrophically without an organic carbon source but with supplementation of the culture vessel atmosphere with 2 % CO2. The photoautotrophically cultivated plantlets showed a better growth and multiplication, higher contents of chlorophyll (Chl) and carotenoids, higher Chl a/b ratio, net photosynthetic rate and ribulose-1,5-bisphosphate carboxylase/oxygenase and phosphoenolpyruvate carboxylase activities than plantlets grown on the medium with sucrose. and C. Alterations in photosynthetic capacity of primary leaves of wheat seedlings in response to ultraviolet-B (UV-B; 280-320 nm; 60 µmol m-2 s-1) exposure alone and in combination with photosynthetically active radiation (PAR; 400-800 nm; 200 µmol m-2 s-1) during different phases of leaf growth and development were assessed. UV-B exposure resulted in a phase-dependent differential loss in photosynthetic pigments, photochemical potential, photosystem 2 (PS2) quantum yield, and in vivo O2 evolution. UV-B exposure induced maximum damage to the photosynthetic apparatus during senescence phase of development. The damages were partially alleviated when UV-B exposure was accompanied by PAR. UV-B induced an enhancement in accumulation of flavonoids during all phases of development while it caused a decline in anthocyanin content during senescence. The differential changes in these parameters demonstrated the adaptation ability of leaves to UV-B stress during all phases of development and the ability was modified in UV-B+ PAR exposed samples. and M. In crowns of chestnut trees the absorption of radiant energy is not homogeneous; leaves from the south (S) side are the most irradiated, but leaves from the east (E) and west (W) sides receive around 70 % and those from north (N) face less than 20 % of the S irradiation. Compared to the S leaves, those from the N side were 10 % smaller, their stomata density was 14 % smaller, and their laminae were 21 % thinner. N leaves had 0.63 g(Chl) m-2, corresponding to 93 % of total chlorophyll (Chl) amount in leaves of S side. The ratios of Chl a/b were 2.9 and 3.1 and of Chl/carotenoids (Car) 5.2 and 4.8, respectively, in N and S leaves. Net photosynthetic rate (PN) was 3.9 µmol(CO2) m-2 s-1 in S leaves, in the E, W, and N leaves 81, 77, and 38 % of that value, respectively. Morning time (10:00 h) was the period of highest PN in the whole crown, followed by 13:00 h (85 % of S) and 16:00 h with 59 %. Below 500 µmol m-2 s-1 of photosynthetic photon flux density (PPFD), N leaves produced the highest PN, while at higher PPFD, the S leaves were most active. In addition, the fruits from S side were 10 % larger than those from the N side. and J. Gomes-Laranjo ... [et al.].
The acclimation depression of capacity of photon utilisation in photochemical reactions of photosystem 2 (PS2) can develop already after three months of cultivation of the Norway spruces (Picea abies [L.] Karst.) under elevated concentrations of CO2 (i.e., ambient, AC, + 350 µmol(CO2) mol-1 = EC) in glass domes with adjustable windows. To examine the role that duration of EC plays in acclimation response, we determined pigment contents, rate of photosynthesis, and parameters of chlorophyll a fluorescence for sun and shade needles after three seasons of EC exposure. We found responses of shaded and exposed needles to EC. Whereas the shaded needles still profited from the EC and revealed stimulated electron transport, for the exposed needles the stimulation of both electron transport activity and irradiance saturated rate of CO2 assimilation (PNmax) under EC already disappeared. No signs of the PS2 impairment were observed as judged from high values of potential quantum yield of PS2 photochemistry (FV/FM) and uniform kinetics of QA reoxidation for all variants. Therefore, the long-term acclimation of the sun-exposed needles to EC is not necessarily accompanied with the damage to the PS2 reaction centres. The eco-physiological significance of the reported differentiation between the responses of shaded and sun exposed needles to prolonged EC may be in changed contribution of the upper and lower crown layers to the production activity of the tree. Whereas for the AC spruces, PNmax of shaded needles was only less than 25 % compared to exposed ones, for the EC spruces the PNmax of shaded needles reached nearly 40 % of that estimated for the exposed ones. Thus, the lower shaded part of the crown may become an effective consumer of CO2. and J. Kalina ... [et al.].
Radish (Raphanus sativus L.) and carrot (Daucus carota L.), plants with underground storage organs grown in the field, were exposed to either ambient (UVA) or 20 % UV-B (UVE) enhanced solar radiation till their root yield stage. In radish, UVE produced a significant increase in shoot and root fresh mass (FM), increase in the contents of chlorophyll, carotenoids, flavonoids, and total proteins per unit FM, Hill reaction rate, and root yield. In contrast, carrot responded negatively to UVE showing a loss in the above parameters. and S. M. J. Nithia, N. Shanthi, G. Kulandaivelu.
We investigated the strategies of four co-occurring evergreen woody species Quercus ilex, Quercus coccifera, Pinus halepensis, and Juniperus phoenicea to cope with Mediterranean field conditions. For that purpose, stem water potential, gas exchange, chlorophyll (Chl) fluorescence, and Chl and carotenoid (Car) contents were examined. We recognized two stress periods along the year, winter with low precipitation and low temperatures that led to chronic photoinhibition, and summer, when drought coincided with high radiation, leading to an increase of dynamic photoinhibition and a decrease of pigment content. Summer photoprotection was related to non-photochemical energy dissipation, electron flow to alternative sinks other than photosynthesis, decrease of Chl content, and proportional increase of Car content. Water potential of trees with deep vertical roots (Q. coccifera, Q. ilex, and P. halepensis) mainly depended on precipitation, whereas water potential of trees with shallow roots (J. phoenicea) depended not only on precipitation but also on ambient temperature. and F. Dynamic changes of neoxanthin (NEO), violaxanthin (VIO), anteraxanthin (ANT), zeaxanthin (ZEA), chlorophyll (Chl) a, Chl b, α-carotene, β-carotene, and their behaviour under increasing duration of high irradiance (HI) were investigated in the soybean hypocotyl callus culture. The calli were induced on solid (1.1 % agar) MS medium (pH 5.8) supplemented with 4.52 μM 2,4-D, 2.32 μM kinetin, and 3 % sucrose. After 30 d of culture, the green calli were irradiated with "white light" (133W m-2) for 0, 3.5, and 24 h. HPLC profiles were separated on a C18 column. With increasing duration of HI, the content of total carotenoids (Cars) increased, but the ratio of Chl a+b/Cars decreased. With lengthening the duration of HI, there was induction of ZEA. Contents of ANT, α-carotene, and β-carotene remained nearly constant, but ratio of ZEA/Chl a+b increased with lengthening the HI duration. and D. M. Pandey, K. H. Kim, U. D. Yeo.
The present study was conducted to study the effect of 24-epibrassinolide (EBL) on changes of plant growth, net photosynthetic rate, carbonic anhydrase (E.C. 4.2.1.1) and nitrate reductase (E.C.1.6.6.1) activities in the leaves of Raphanus sativus L. under the influence of cadmium (Cd) stress. Cd reduced plant growth, photosynthetic pigment levels, net photosynthetic rate and the activities of carbonic anhydrase and nitrate reductase. However seed application of EBL reduced the toxic effect of Cd on plant growth, pigment content, photosynthesis and enzyme activities. The studies clearly demonstrated the ameliorating effect of 24-epibrassinolide in mitigating the toxicity of Cd in plants. and S. Anuradha, S. Seeta Ram Rao.
The effect of a short cold stress in combination with photoinhibition stress, similar to a low temperature and a high irradiance situation during early morning in the spring time, was examined on four maize cultivars common for Belgium, that differ in early vigour. After 1 h of 2 °C and 500 μmol(photon) m-2 s-1, quantum efficiency and maximum photosynthesis rate at saturating irradiance decreased on average by 11 and 8 %, respectively. For one cultivar, Magister, the decrease was the largest: by 23 and 10 %, respectively. For this cultivar it was combined with a decrease of the water vapour conductance after the stress. The decrease of Fv/F0 due to the cold/light stress was dependent on the cold tolerance (early vigour) of the cultivars. Fv/F0 changed with -45.5 and -40.2 % for the cultivars Ardiles and Banguy, respectively (cultivars with a less good early vigour) in comparison to -36.3 and -35.9 % for Fjord and Magister, which have a good early vigour. Also the ratio of total chlorophylls/total carotenoids changed in dependence on cold tolerance of the cultivars. For more cold tolerant cultivars, the relative amount of total carotenoids (x+c) was higher, indicating a higher protective state. Both the parameter Fv/F0 and the ratio of total chlorophylls to total carotenoids can be used to differentiate the cold tolerant cultivars from the cold non-tolerant ones. Fv/F0 has the advantage because its resolving power is larger and the measurement is less expensive than determination of the pigment ratio. and P. Lootens, J. van Waes, L. Carlier.
Gas exchange and chlorophyll (Chl) fluorescence were measured on young mature leaves of rose plants (Rosa hybrida cvs. First Red and Twingo) grown in two near-to-tight greenhouses, one under control ambient CO2 concentration, AC (355 µmol mol-1) and one under CO2 enrichment, EC (700 µmol mol-1), during four flushes from late June to early November. Supply of water and mineral elements was non-limiting while temperature was allowed to rise freely during daytime. Leaf diffusive conductance was not significantly reduced at EC but net photosynthetic rate increased by more than 100 %. Although the concentration of total non-structural saccharides was substantially higher in the leaves from the greenhouse with EC, ΦPS2 (quantum efficiency of radiation use) around noon was not significantly reduced at EC indicating that there was no down-regulation of electron transport. Moreover, CO2 enrichment did not cause any increase in the risk of photo-damage, as estimated by the 1 - qP parameter. Non-photochemical quenching was even higher in the greenhouse with EC during the two summer flushes, when temperature and photosynthetic photon flux density (PPFD) were the highest. Hence rose photosynthesis benefits strongly from high concentrations of atmospheric CO2 at both high and moderate temperatures and PPFD. and L. Urban ... [et al.].
