Low temperature during the vegetative stage affects rice (Oryza sativa L.) seed-setting rate in Heilongjiang province at Northeast China. However, little is known about changes of the photosynthetic rate and physiological response in contrasting rice cultivars during chilling periods. In this study, two rice cultivars with different chilling tolerance were treated with 15°C from June 27 to July 7. The chilling-susceptive cultivar, Longjing11 (LJ11), showed a significant decrease in a ripening rate and seed-setting rate after being treated for four days, whilst chilling-tolerant cultivar, Kongyu131 (KY131), was only slightly affected after 4-d treatment. The photosynthetic activities, chlorophyll contents, and antioxidative enzyme activities in LJ11 decreased significantly along with the chilling treatment. The decrease in ß-carotene contents might play a role as it could cause direct photooxidation of chlorophylls and lead to the inhibition of the photosynthetic apparatus. In the meantime, no significant damage was found in leaves of KY131 from June 27 to July 11. In conclusion, the chilling-tolerance mechanism of rice is tightly related to the photosynthetic rate, metabolism of reactive oxygen species, and scavenging system in the vegetative stage., L.-Z. Hyperbaric oxygen (HBO) therapy, i.e. breathing pure oxygen
under increased environmental pressures serves as a treatment
for diverse medical conditions. However, elevated oxygen
concentration can be detrimental to central nervous system or
lungs. Our study aimed to evaluate the effects of repeated
exposure to HBO on mitochondrial respiration assessed by highresolution respirometry (HRR), cell viability estimated by
PrestoBlue® reaction, morphology analyzed by routine phase
contrast and fluorescent microscopy, and superoxide dismutase
(SOD) and citrate synthase (CS) activities using human lung
fibroblasts. The cells were exposed to HBO for 2 h per day for
5 consecutive days. One day after the last exposure, HBO cells
displayed significantly smaller area and perimeter, compromised
viability and elevated SOD activity. No changes were detected in
CS activity or quality of mitochondrial network. HRR revealed
impaired mitochondrial oxygen consumption manifested by
increased leak respiration, decreased activity of complex II and
compromised ATP-related oxygen consumption when fatty acids
were oxidized. Our findings document that in conditions
mimicking chronic intermittent exposure to HBO, lung fibroblasts
suffer from compromised mitochondrial respiration linked to
complex II and impaired cellular growth in spite of increased
antioxidant defense. Underlying mechanism of this HBO-induced
mitochondrial dysfunction should be further explored.
Over-expression of chloroplastic glycerol-3-phosphate acyltransferase gene (LeGPAT) increased unsaturated fatty acid contents in phosphatidylglycerol (PG) of thylakoid membrane in tomato. The effect of this increase on the xanthophyll cycle and chloroplast antioxidant enzymes was examined by comparing wild type (WT) tomato with the transgenic (TG) lines at chilling temperature (4 °C) under low irradiance (100 µmol m-2 s-1). Net photosynthetic rate and the maximal photochemical efficiency of photosystem (PS) 2 (Fv/Fm) in TG plants decreased more slowly during chilling stress and Fv/Fm recovered faster than that in WT plants under optimal conditions. The oxidizable P700 in both WT and TG plants decreased during chilling stress under low irradiance, but recovered faster in TG plants than in the WT ones. During chilling stress, non-photochemical quenching (NPQ) and the de-epoxidized ratio of xanthophyll cycle in WT plants were lower than those of TG tomatoes. The higher activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in TG plants resulted in the reduction of O2-. and H2O2 contents during chilling stress. Hence the increase in content of unsaturated fatty acids in PG by the over-expression of LeGPAT could alleviate photoinhibition of PS2 and PS1 by improving the de-epoxidized ratio of xanthophyll cycle and activities of SOD and APX in chloroplast. and N. Physiological responses of two wheat (Triticum aestivum L.) genotypes (salt-tolerant DK961 and salt-sensitive JN17) to increased salt concentrations (50, 100, 150 mM NaCl: NaCl50, NaCl100, NaCl150) were studied. Photosynthetic capacity, irradiance response curves, contents of soluble sugars, proteins, and chlorophyll (Chl), K+/Na+ ratio, and activities of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase) in flag leaves were measured on 7 d after anthesis. In control (NaCl0) plants, non-significant (p>0.05) differences were found in gas exchange and saturation irradiance (SI) between salt-tolerant (ST) and salt-sensitive (SS) wheat genotypes. However, we found higher soluble sugar and protein contents, K+/Na+ ratio, and antioxidant enzyme activities, but lower Chl content and yield in ST wheat. Salinity stresses remarkably increased soluble sugar and protein contents and the antioxidant activities, but decreased K+/Na+ ratio, Chl contents, SI, photosynthetic capacities, and yield, the extent being considerably larger in JN17 than DK961. Although the soluble sugar and protein contents and the antioxidant activities of JN17 elevated more evidently under salt stresses, those variables never reached the high levels of DK961. The antioxidant enzyme activities of SS wheat increased in NaCl50 and NaCl100, but decreased rapidly when the NaCl concentration reached 150 mM. Thus the ST wheat could maintain higher grain yield than the SS one by remaining higher osmoregulation and antioxidative abilities, which led to higher photosynthetic capacity. The gas exchange, parameters of chlorophyll fluorescence, contents of pigments, and activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), as well as lipid peroxidation were investigated in two field-grown coffee species, Coffea arabica and C. liberica, exposed to drought and re-hydration. Drought caused a more pronounced inhibition of net photosynthetic rate in C. liberica compared to C. arabica. The de-epoxidation of xanthophyll cycle pigments at midday estimated by leaf reflectance was much higher in C. arabica than in C. liberica, but no significant change was found in response to drought. Under moderate drought, the activities of SOD and APX increased significantly only in C. arabica. The maximum photochemical efficiency of photosystem 2, PS2 (Fv/Fm) at predawn did not change and there was no lipid peroxidation during this time. Under severe drought Fv/Fm decreased and initial fluorescence (F0) increased for both species, and SOD activity increased, APX activity remained relatively high, and malondialdehyde (MDA) accumulated in C. arabica, while APX decreased in C. liberica. The photosynthetic apparatus of C. arabica was completely recovered after 5 d of re-irrigation as indicated by the restoration of Fv/Fm to the control values. A lack of recovery upon rewatering of C. liberica indicated irreversible damage to PS2. Hence compared to C. liberica, C. arabica possesses a higher desiccation-induced antioxidative protection and higher portion of the total pigment pool used in photoprotection, which might aid alleviating photoinhibitory damage during desiccation and photosynthesis recovery when favourable conditions are restored. and Z.-Q. The study of ischemia/reperfusion injury included 25 patients in the acute phase of myocardial infarction (19 perfused, 6 remained non-reperfused as evaluated according to the time course of creatine kinase and CK-MB isoenzyme activity) and a control group (21 blood donors). Plasma level of malondialdehyde was followed as a marker of oxidative stress. Shortly after reperfusion (within 90 min), a transient increase of malondialdehyde concentration was detected. The return to the baseline level was achieved 6 h after the onset of therapy. The activity of a free radical scavenger enzyme, plasma glutathione peroxidase (GPx), reached its maximum 90 min after the onset of treatment and returned to the initial value after 18 h. The specificity of the GPx response was confirmed by comparing with both non-reperfused patients and the control group, where no significant increase was detected. The erythrocyte Cu,Zn-superoxide dismutase (SOD) did not exhibit significant changes during the interval studied in perfused patients, probably due to the stability of erythrocyte metabolism. In non-reperfused patients, a decrease of SOD was found during prolonged hypoxia. a1_Different parameters that vary during leaf development may be affected by light intensity. To study the influence of different light intensities on primary leaf senescence, sunflower (Helianthus annuus L.) plants were grown for 50 days under two photon flux density (PFD) conditions, namely high irradiance (HI) at 350 μmol(photon) m-2 s-1 and low irradiance (LI) at 125 μmol(photon) m-2 s-1. Plants grown under HI exhibited greater specific leaf mass referred to dry mass, leaf area and soluble protein at the beginning of the leaf development. This might have resulted from the increased CO2 fixation rate observed in HI plants, during early development of primary leaves. Chlorophyll a and b contents in HI plants were lower than in LI plants in young leaves. By contrast, the carotenoid content was significantly higher in HI plants. Glucose concentration increased with the leaf age in both treatments (HI and LI), while the starch content decreased sharply in HI plants, but only slightly in LI plants. Glucose contents were higher in HI plants than in LI plants; the differences were statistically significant (p<0.05) mainly at the beginning of the leaf senescence. On the other hand, starch contents were higher in HI plants than in LI plants, throughout the whole leaf development period. Nitrate reductase (NR) activity decreased with leaf ageing in both treatments. However, the NR activation state was higher during early leaf development and decreased more markedly in senescent leaves in plants grown under HI. GS activity also decreased during sunflower leaf ageing under both PFD conditions, but HI plants showed higher GS activities than LI plants. Aminating and deaminating activities of glutamate dehydrogenase (GDH) peaked at 50 days (senescent leaves). The activities of cytosol superoxide dismutase (CuZnSOD) and mitochondrial superoxide dismutase (MnSOD) were measured in subcellular fractions of homogenates prepared from the brain, thymus and liver of ovariectomized (OVX) female rats, non-treated or treated 24 h prior to sacrifice with a single s.c. dose of 0.1 ml olive oil. In the brain, neither MnSOD nor CuZnSOD were affected by olive oil, whereas in the thymus the olive oil injection elevated CuZnSOD and did not affect MnSOD activity. At the same time, the activity of CuZnSOD was reduced and that of MnSOD was elevated in the liver following oil treatment. These results suggest that olive oil has modulatory effects on the expression of CuZnSOD and MnSOD activity in the liver and of CuZnSOD in the thymus of female rats.
The goldfish (Carassius auratus gibelio Bloch.) were exposed to cadmium in the concentration of 20 mg Cd/1 water under aquarium conditions for 1, 4, 7 and 15 days. After exposure to cadmium, the activities of superoxide dismutase (SOD) and catalase (CAT) were significantly decreased. At the same time, the liver ascorbic acid (AsA) content was increased.
The excessive production of nitric oxide (NO) and the subsequent increase of local oxidative stress is suggested as one of the pathophysiological mechanisms of streptozotocin-induced diabetes. It was reported that the administration of NO synthase inhibitors partially attenuated the development of streptozotocin-induced diabetes and reduced hyperglycaemia. Here we have studied the influence of methylene blue, which combines the properties of NO synthase inhibitor with antioxidant effects. The experiments were performed on male rats divided into four groups: control, diabetic (single dose of 70 mg of streptozotocin/kg i.p.), methylene blue (50 mg/kg in the food) and diabetic simultaneously fed with methylene blue. After 45 days the experiments were discontinued by decapitation. Serum glycaemia, glycated haemoglobin and oxidative stress parameters (plasma malondialdehyde concentration and erythrocyte superoxide dismutase activity) were significantly higher in the diabetic group. Simultaneous methylene blue administration partially reduced glycaemia and glycated haemoglobin, but did not decrease oxidative stress. We conclude that NO synthase inhibitor methylene blue partially attenuates the development of streptozotocin-induced diabetes in male rats, but does not reduce the development of oxidative stress in the diabetic group.
