The nucleus accumbens (NAc) core is critical in the control of motivated behaviors. The muscarinic acetylcholine receptors (mAChRs) modulating the excitatory inputs into the NAc core have been reported to impact such behaviors. Recent studies suggest that ventral and dorsal regions of the NAc core seem to be innervated by distinct popula tions of glutamatergic projection neurons. To further examine mAChRs modulation of these glutamatergic inputs to the NAc core, we employed intracellular recordings in rat NAc coronal slice preparation to characterize: 1) the effects of muscarine, an mAChRs agonist, on membrane properties of the NAc core neurons; 2) depolarizing synaptic potentials (DPSP) elicited by ventral and dorsal focal electrical stimuli; and 3) paired-pulse response with paired-pulse stimulation. Here we report that the paired-pulse ratio (PPR) elicited by dorsal stimuli was grea ter than that elicited by ventral stimuli. Bath application of muscarine (1-30 μ M) decreased both ventral and dorsal DPSP in a concentration-dependent manner, with no effect on electrophysiological properties of NAc core neurons. Muscarine at 30 μ M also elicited larger depression of dorsal DPSP than ventral DPSP. Moreover, muscarine increased the PPR of both dorsal and ventral DPSP. These data indicate that the glutamatergic afferent fibers traversing the dorsal and ventral NAc are separate, and that differential decrease of distinct afferent excitatory neurotransmission onto NAc core neurons may be mediated by presynaptic mechanisms., X. The effects of NaCl stress on the growth and photosynthetic characters of Ulmus pumila L. seedlings were investigated under sand culture condition. With increasing NaCl concentration, main stem height, branch number, leaf number, and leaf area declined, while Na+ content and the Na+/K+ ratio in both expanded and expanding leaves increased. Na+ content was significantly higher in expanded leaves than in those just expanding. Chlorophyll (Chl) a and Chl b contents declined as NaCl concentration increased. The net photosynthetic rate, intercellular CO2 concentration, stomatal conductance, and transpiration rate also declined, but stomatal limitation value increased as NaCl concentration increased. Both the maximal quantum yield of PSII photochemistry and the effective quantum yield of PSII photochemistry declined as NaCl concentration rose. These results suggest that the accumulation of Na+ in already expanded leaves might reduce damage to the expanding leaves and help U. pumila endure high salinity. The effect of drought stress on energy dissipation and antioxidant enzyme system in two sweet sorghum inbred lines (M-81E and Roma) was investigated. Results showed that the germination indicator increased more in M-81E than that in Roma under rehydration. Under drought stress, both the maximal photochemical efficiency of PSII (Fv/Fm) and oxidoreductive activity (ΔI/I0) of Roma decreased more than those in M-81E. Relative to Fv/Fm, the ΔI/I0 decreased markedly, which indicated that PSI was more sensitive to drought stress than PSII. Increases in the reduction state of QA (1-qp), nonphotochemical quenching (NPQ) and minimal fluorescence yield of the dark-adapted state (F0) were greater in Roma than those in M-81E; meanwhile, the H2O2 content was lower in M-81E than that in Roma. Our results suggested that the photoinhibition might be related to the accumulation of reactive oxygen species (ROS). The antioxidant enzyme system and energy dissipation of M-81E could respectively increase drought tolerance by eliminating ROS and excess energy more efficiently than that of Roma., Y. Y. Guo, S. S. The negative affective state of opiate abstinence plays an important role in craving and relapse to compulsive drug use. The dopamine system participates in the reward effects of opiate use and the aversive effect of opiate abstinence. The amygdala is an essential neural substrate for associative learning of emotion. To establish a model of conditioned place aversion (CPA) in morphine-treated rats, we used different visual and tactual cues as conditioned stimuli (CS) within a conditioning apparatus. An injection of naloxone served as the unconditioned stimulus (US). The 6-hydroxydopamine (6-OHDA) lesion technique was used to investigate the effects of the dopaminergic system of the central nucleus of the amygdala (CeA) on naloxone-induced CPA. Rats were rendered physically dependent via administration of increasing doses of morphine delivered via intraperitoneal injection. Doses increased by 20 % each day for 14 days, starting from an initial dose of 6 mg/kg. All rats also received a low dose of naloxone (0.1 mg/kg) by injection 4 hours after morphine treatment on days 11 and 13 to induce CPA in a biased twocompartment conditioned place apparatus. Morphine-dependent rats with sham lesions were found to develop significant CPA after naloxone treatment. Bilateral 6-OHDA lesions of the CeA impaired the acquisition of CPA but had no effect on locomotor activity. In transgenic (TG) tomato (Lycopersicon esculentum Mill.) overexpressed ω-3 fatty acid desaturase gene (LeFAD7) was identified, which was controlled by the cauliflower mosaic virus 35S promoter and induced increased contents of unsaturated fatty acids in thylakoid membrane. Under chilling stress at low irradiance (4 °C, 100 µmol m-2 s-1) TG plants with higher linolenic acids (18: 3) content maintained a higher O2 evolution rate, oxidizable P700 content, and maximal photochemical efficiency (Fv/Fm) than wild type (WT) plants. Low temperature treatment for 6 h resulted in extensive changes of chloroplast ultrastructure: in WT plants most chloroplasts became circular, the number of amyloids increased, appressed granum stacks were dissolved, grana disappeared, and the number of grana decreased, while only a few grana were found in leaves of TG plants. To investigate the photoprotection of energy dissipation and water-water cycle, a C3 euhalophytic herb, Suaeda salsa L., was exposed either to chilling temperature (4°C) accompanied by moderate irradiance (600 μmol m-2 s-1) (CM) and/or to chilling temperature (4°C) accompanied by low irradiance (100 μmol m-2 s-1) (CL). During chilling stress, both the maximal photochemical efficiency of PSII (Fv/Fm) and the oxidizable P700 decreased in S. salsa leaves either under CM or CL, which indicated the severe photoinhibition. Relative to Fv/Fm, the oxidizable P700 decreased markedly under CL, which indicated that PSI was more sensitive to CL treatment than PSII. Initial fluorescence, number of closed PSII centers, and nonphotochemical quenching increased under CM, but more markedly under CL in S. salsa leaves. Activity of superoxide dismutase and ascorbate peroxidase was higher under CM than that under CL. The production of reactive oxygen species (ROS) decreased first and then increased under both treatments, but the content of O2.- and H2O2 was higher under CL than that under CM after 12 h of chilling stress. These results suggested that photoinhibition in S. salsa might be related to the accumulation of reactive oxygen species (ROS) induced by excess energy. Over-expression of chloroplastic glycerol-3-phosphate acyltransferase gene (LeGPAT) increased unsaturated fatty acid contents in phosphatidylglycerol (PG) of thylakoid membrane in tomato. The effect of this increase on the xanthophyll cycle and chloroplast antioxidant enzymes was examined by comparing wild type (WT) tomato with the transgenic (TG) lines at chilling temperature (4 °C) under low irradiance (100 µmol m-2 s-1). Net photosynthetic rate and the maximal photochemical efficiency of photosystem (PS) 2 (Fv/Fm) in TG plants decreased more slowly during chilling stress and Fv/Fm recovered faster than that in WT plants under optimal conditions. The oxidizable P700 in both WT and TG plants decreased during chilling stress under low irradiance, but recovered faster in TG plants than in the WT ones. During chilling stress, non-photochemical quenching (NPQ) and the de-epoxidized ratio of xanthophyll cycle in WT plants were lower than those of TG tomatoes. The higher activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in TG plants resulted in the reduction of O2-. and H2O2 contents during chilling stress.
