As part of a biodiversity study in northwestern Hungary, we conducted a parasitological survey of small mammals. In both common shrews (Sorex araneus Linnaeus) and pygmy shrews (Sorex minutus Linnaeus), we found myxospores of a species of Soricimyxum Prunescu, Prunescu, Pucek et Lom, 2007 (Myxosporea) and plasmodia in the bile ducts within the liver. Spores from both species of shrewswere morphologically and morphometrically indistinguishable, but differed in their SSU rRNA gene sequences by 3.3%. We identified spores and developmental stages from the common shrew as Soricimyxum fegati Prunescu, Prunescu, Pucek et Lom, 2007, based on morphometric data and DNA sequence similarity. Spores from the pygmy shrew were only 96.7% similar to S. fegati, hence we identified them as a novel myxosporean Soricimyxum minuti sp. n. Myxozoans (Cnidaria: Myxozoa) are almost exclusively endoparasites of aquatic vertebrates and invertebrates, with the notable exception being two species of Soricimyxum Prunescu, Prunescu, Pucek et Lom, 2007 described from terrestrial shrews (Soricidae) in central Europe. Myxospores of the two parasites are morphologically indistinguishable, but have SSU rDNA sequences that differ by about 4%. Herein, we report additional molecular and histology data from Soricimyxum fegati Prunescu, Prunescu, Pucek et Lom, 2007 from common shrew (Sorex araneus Linnaeus) from Hungary, and add a new geographic record for S. fegati in pygmy shrew (Sorex minutus Linnaeus) from Slovakia. A limited survey of shrews from the northern United States, Blarina brevicauda Say and Sorex sp. from New York, and Sorex spp. from Oregon, did not discover any infections, which is in stark contrast to the relatively high infection rates (up to 66%) in European shrew populations. We also provide a summary and discussion of literature records of species of Soricimyxum and a host survey. Given the lack of distinguishing morphological or morphometric characters between Soricimyxum spp., and the overlap in vertebrate hosts and geographic ranges, unambiguous identification of these closely related shrew parasites can presently only be achieved through sequence comparison of one or more variable SSU rDNA regions., Csaba Székely, Stephen D. Cadmium (Cd), an environmental and industrial pollutant, poses a potential threat and affects many systems in human and animals. Although several reports on Cd toxicity were presented, the acute effect of Cd on systemic and thrombotic events was not reported so far. Cd (2.284 mg/kg) or saline (control) was injected intraperitoneally (ip), and the systemic parameters were assessed in mice. Compared to control group, acute intraperitoneal injection of Cd, in mice showed significant quickening of platelet aggregation (P<0.001) leading to pial cerebral thrombosis. Likewise, Cd exposure caused a significant increase in white blood cell numbers (P<0.05) indicating the occurrence of systemic inflammation. Also, alanine aminotransferase (ALT) (P<0.05) and creatinine (P<0.01) levels were both significantly increased. Interestingly, the superoxide dismutase activity was significantly decreased in Cd treated group compared to control group (P<0.001), suggesting the occurrence of oxidative stress. Acute liver failure (ALF) is a clinical condition with very high mortality rate. Its pathophysiological background is still poorly understood, which necessitates a search for optimal experimental ALF models with features resembling those of the human disorder. Taking into consideration reproducibility of induction of ALF, adequate animal size, cost of animals, the required time gap between insult and death of animals (“therapeutic window”), potential risk to investigator and other aspects, administration of thioacetamide (TAA) in rats is currently most recommended. However, the fundamental details of this ALF model have not yet been evaluated. This prompted us to investigate, first, the course of ALF as induced by intraperitoneal TAA at doses increasing from 175 to 700 mg/kg BW per day. The animals’ survival rate, plasma alanine and aspartate aminotransferase activities, and bilirubin and ammonia levels were determined over the follow-up period. Second, we examined whether Wistar and Lewis rats exhibit any differences in the course of ALF induced by different TAA doses. We found that the optimal dose for ALF induction in rats is 350 mg.kg-1 i.p., given as a single injection. Wistar rats proved more susceptible to the development of TAA-induced ALF compared with Lewis rats. Collectively, our present findings provide a sound methodological background for experimental studies aimed at evaluation of pathophysiology and development of new approaches in the therapy of ALF., E. The aim of the current study was to clarify the effect of high sucrose diet (HSD) on bile formation (BF) in rats with hereditary hypertriglyceridemia (HHTg). Potentially positive effects were studied for boldine, a natural choleretic agent. Administration of HSD to HHTg rats led to increased triglyceride deposition in the liver. HSD reduced BF as a consequence of decreased biliary secretion of bile acids (BA) and glutathione. Responsible mechanism was down-regulation of hepatic transporters for BA and glutathione, Bsep and Mrp2, respectively. Moreover, gene expressions of transporters for other constituents of bile, namely Abcg5/8 for cholesterol, Abcb4 for phospholipids, and Oatp1a4 for xenobiotics, were also reduced by HSD. Boldine partially attenuated cholestatic effect of HSD by promotion of biliary secretion of BA through up-regulation of Bsep and Ntcp, and by increase in biliary secretion of glutathione as a consequence of its increased hepatic disposition. This study demonstrates mechanisms of impaired BF during nonalcoholic fatty liver disease induced by HSD. Altered function of responsible transporters suggests also potential for changes in kinetics of drugs, which may complicate pharmacotherapy in subjects with high intake of sucrose, and with fatty liver disease. Patients with obesity and type 2 diabetes often display high levels of the anti-diabetic factor fibroblast growth factor-21 (FGF21), suggesting that the overproduction of FGF21 may result from increased adiposity in an attempt by white adipose tissue (WAT) to counteract insulin resistance. However, the production of FGF21 diabetes in the absence of WAT has not been examined. In this study, we investigated the effects of lipodystrophy in A-ZIP F-1 mice on FGF21 production in relation to diabetes. A-ZIP F-1 mice displayed high FGF21 plasma levels resulting from enhanced FGF21 mRNA expression in the liver. Concomitant enhancement of FGF21 receptor (FGFR1) and glucose transporter 1 (GLUT-1) mRNA expression was observed in the muscles of A-ZIP F-1 mice. Furthermore, the activation of hypothalamic NPY and AgRP mRNA expression positively correlated with plasma levels of FGF21 but not active ghrelin. The effects of various stressors on insulin receptors in adipose, liver and skeletal muscle tissues were studied in rats exposed to acute or repeated stress. Adult male rats were exposed to immobilization (IMO) for 2.5 hours daily for 1, 7 and 42 days, or to hypokinesia (HK) for 1, 7 and 21 days. We determined the values of specific insulin binding (SIB) and insulin receptor binding capacity (IR) of plasma cell membranes from adipose, liver and muscle tissue (IMO groups), or insulin binding to isolated adipocytes and hepatocytes (HK groups). A significant decrease of SIB and IR was observed in rats exposed to acute stress (1x IMO) in muscle, adipose and liver tissues. However, in animals exposed to repeated stress (7x and 42x IMO), SIB and IR were diminished in the muscle tissue, whereas no significant changes were noted in the liver and adipose tissue. When tissue samples were collected 3-24 hours after exposure to IMO stress, no changes of SIB and IR were found in liver and adipose tissue, but insulin binding was lowered in skeletal muscles. In animals exposed to HK for one day, a decrease of SIB and IR was found in isolated adipocytes, but no changes in insulin binding were noted in the liver tissue. In rats exposed to HK for 7 and 21 days, values of IR were similar as in control group. Clinical reports suggest close interactions between stressors, particularly those of long duration, and liver diseases, such as hepatic inflammation, that is proposed to occur via reactive oxygen species. In the present study we have used 21-day social isolation of male Wistar rats as a model of chronic stress to investigate protein expression/activity of liver antioxidant enzymes: superoxide dismutases (SODs), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GLR), and protein expression of their upstream regulators: glucocorticoid receptor (GR) and nuclear factor kappa B (NFkB). We have also characterized these parameters in either naive or chronically stressed animals that were challenged by 30-min acute immobilization. We found that chronic isolation caused decrease in serum corticosterone (CORT) and blood glucose (GLU), increase in NFkB signaling, and disproportion between CuZnSOD, peroxidases (CAT, GPx) and GLR, thus promoting H2O2 accumulation and prooxidative state in liver. The overall results suggested that chronic stress exaggerated responsiveness to subsequent stressor at the level of CORT and GLU, and potentiated GLR response, but compromised the restoration of oxido-reductive balance due to irreversible alterations in MnSOD and GPx., J. The effect of adding 5 % powdered oyster mushroom (Pleurotus ostreatus) during 12 weeks on kinetic parameters of cholesterol metabolism was studied in male rats (Wistar, initial body weight 85 g) fed a semisynthetic diet containing 0.3 % of cholesterol. The plasma cholesterol decay curve (examined for the final 29 days of the experiment after a single dose of cholesterol-4- 14C) was evaluated by mathematical analysis using a two-pool model of plasma cholesterol metabolism. The oyster mushroom in the diet reduced the half-times of both exponentials resulting in lower calculated values (by 28 %) of total entry of cholesterol into the body cholesterol pool (absorption + endogenous synthesis) and lower sizes of both pools (with slower and faster cholesterol exchange). The rate of cholesterol exchange between the pools was enhanced and the rate of total clearance of cholesterol from the system (metabolic turnover rate of cholesterol, i.e. the rate of degradation and excretion of cholesterol from the organism) was enhanced by 50 %. (decrease by 25 %) that were induced by the cholesterol diet.
