Temporal patterns and frequency spectra of the songs and stridulatory files of 14 species of the genus of the phaneropterid bush-crickets Isophya from Eastern Europe, Altai and the Caucasus are given. The sound signals of the species studied can be separated into three main types: (1) those consisting of two syllables (Isophya gracilis, I. kalishevskii, I. schneideri, I. caspica, Isophya sp. 1); (2) one syllable and series of clicks (I. modesta rossica, I. stepposa, I. taurica, I. brunneri, I. doneciana, I. altaica); (3) single repeating syllables of uniform shape and duration (I. pienensis, Isophya sp. 2 and possibly I. stysi). The central-marginal model is widely accepted in chromosomally polymorphic species of Drosophila. In fact, geographically and ecologically central populations of Drosophila show higher levels of polymorphism for paracentric inversions, whereas marginal populations tend to be monomorphic. This fact has been variously explained. Chromosomal polymorphisms in grasshoppers have also been attributed to show such geographical structuring, as in the case of the South-American grasshopper Dichroplus pratensis Bruner (Orthoptera: Acrididae). However, in three other cases involving Acrididae – Leptysma argentina Bruner, Trimerotropis pallidipennis (Burmeister) and Cornops aquaticum (Bruner), it is clear that chromosomal polymorphisms (sometimes with a wide extension over the Argentine area) do not conform to this pattern, and show instead clear correlations with environmental variables, especially minimum temperature, showing low or null frequencies of the rearrangements at one extreme of the environmental gradient and with high or fixed frequencies at the other. Furthermore, this correlation with temperature might also be true in the case of D. pratensis. These aforementioned examples emphasise the dangers of over-generalization when discussing chromosomal polymorphisms, and suggests that such polymorphisms should be considered very much in a case-specific manner in terms of the particular genetic system under study., Pablo C. We studied the diet of 10 species of grasshopper belonging to the family Pamphagidae over a period of 3 years at 6 localities in North Eastern Algeria. The species of plants consumed by the grasshoppers was determined by comparing slide mounted specimens of the pieces of plant epidermis in their faeces with those in a reference collection of identified plants collected from the same localities. The percentages of occurrence of the different species of plants in the faeces of the grasshoppers were not related to the abundance of the plants at the sites studied. All the grasshoppers were polyphagous but differed in the percentage of Poaceae in their diets. The diet of Tmethis and Ocneridia contained a higher percentage of Poaceae than the other species and are considered to be ambivores. The three species in the Pamphagus gr. djelfensis complex differ in their diets but all tend to avoid consuming Poaceae and are categorized as forbivores. We also compared the frequency of occurrence of Fabaceae in the faeces and in the field and O. volxemii is the only species that avoided consuming this plant family. The number of sensilla on the labrum was also studied in both sexes of each species. Once one corrects for differences in the size of the labrum, the forbivores have higher numbers of sensilla in groups A1, A2 and A3 (but not A10) than the ambivores. The ability of biocontrol agents to overcome the immune defense of pests is a crucial issue. This is the first study of lysozyme activity as an inducible humoral component of the defense of Schistocerca gregaria, which depends on the recognition of the elicitor molecules of pathogens and not on epidermal wounding or a spiking effect. The level of lysozyme activity in fat body, haemocytes and haemolymph plasma of naïve and immunologically challenged 5th instar S. gregaria was evaluated using the zone of inhibition test against Micrococcus lysodeikticus. Various Gram-positive and Gram-negative bacteria as well as peptidoglycans (PGN) and lipopolysacchrides (LPS) of bacterial cell walls induce and increase in the level of lysozyme activity. Escherichia coli induced an increase in the level of activity of lysozyme in the fat body, haemocytes and plasma, but not in mid gut epithelium, 6–12 h after an immunological challenge and then it decreased to the constitutive level after 72 h. The species of the Poecilimon heroicus-group occur around the Caucasus (from north-eastern Turkey to south-eastern Ukraine). We describe the diagnostic morphological characters of all these species and the male calling song of three of the four species. Based on this data the following phylogenetic relationship is derived (P. tschorochensis (P. tricuspis (P. heroicus, P. bifenestratus))). Within the genus Poecilimon, the species can be recognised by a relatively wide pronotum and large tegmina. In one species, Poecilimon tschorochensis Adelung, 1907 (type species of the monotypic genus Artvinia Karabag, 1962, syn. n.; P. rammei Miram, 1938, syn. n.), the tegmina are very large and the song has unusually low spectral components. This species produced di-syllabic echemes at intervals of about 10 s. In two other species of the group, P. heroicus and P. bifenestratus, the calling song of males consists of an uninterrupted dense sequence of long syllables (syllable duration around 0.5 s; ca. 1 syllable/s at 20°C). In these species the auditory spiracles are reduced in size in both sexes, and the females have extremely small tegmina and are unable to respond to the male song acoustically, which would be typical for Phaneropteridae. The change in communication from acoustically responding to mute females has not been previously documented within a group of closely related species.
Chromosomes of the males of five species of Odontura, belonging to the subgenera Odontura and Odonturella, were analyzed. Intensive evolution of the karyotype was recorded, both in terms of changes in the numbers of chromosomes (from 2n = 31 to 27) and the sex chromosome system (from X0 to neo-XY and X0 to neo-X1X2Y). Karyotype evolution was accompanied by tandem autosome fusions and interspecific autosomal and sex chromosome differentiation involving changes in the locations of nucleolar organizer regions, NORs, which were revealed by silver impregnation and confirmed by FISH using an 18S rDNA probe. O. (Odonturella) aspericauda is a polytypic species with X0 and neo-X1X2Y sex determination. The latter system is not common in tettigoniids. It possibly originated by a translocation of a distal segment of the original X chromosome onto a medium sized autosome, resulting in a shortened neo-X1 and a metacentric neo-Y. The remaining autosome homologue became the neo-X2 chromosome. A novel panel of 16 microsatellite markers, obtained by pyrosequencing of enriched genomic libraries, is reported for the flightless European bushcricket Ephippiger diurnus (Dufour) (Orthoptera: Tettigoniidae). Five multiplex and one simplex PCR protocols were optimized, and the polymorphism at the 16 loci was assessed in two natural populations from southern France. The mean allele number and (expected mean heterozygosity) were 8.94 (0.71) and 6.57 (0.70), respectively, in each population. Several loci were at Hardy-Weinberg disequilibrium (HWD), possibly due to the incidence of null alleles. The occurrence of null alleles has been previously reported for this species, and it is a common feature of microsatellite loci in Orthoptera. Cross-amplification tests demonstrated the transferability of some of these loci to other ephippigerine species. The microsatellite loci reported here substantially increase the number of available loci for this species and will afford an accurate picture of E. diurnus phylogeography, the genetic structure of its populations, and an improved understanding of the evolution of male song and other sexually-selected traits in this highly variable species., Yareli Esquer-Garrigos, Michael D. We report the karyotype characteristics including chromosome numbers of Saga campbelli campbelli, S. c. gracilis, and S. rammei using the following classical cytogenetic methods: C-banding, silver staining, and fluorochrome staining DAPI and CMA3. We also present FISH data showing the distribution of telomeric repeats and 18S rDNA on the chromosomes of these species and the results of similar studies cited in the literature on S. hellenica, S. natoliae, and S. rhodiensis. The five European Saga species exhibit a high rate of karyotype evolution. In addition to changes in chromosome number and morphology (by chromosomal inversion and/or chromosome fusion), interspecific autosomal differentiation involved changes in the distribution and quantity of constitutive heterochromatin and GC-rich regions, as well as the number and location of NORs. In the present study we focused on testing a hypothetical model of karyotype evolution in Saga, with particular reference to the cytogenetic mapping of rDNA and telomeric sequences. Variation in the distribution of rDNA and location of Ag-NORs are novel phylogenetic markers for the genus Saga.
Trehalose is not only an important disaccharide, but also a key stress resistance factor in the development of many organisms, including plants, bacteria, fungi, and insects. To study the potential function of trehalose in development and behaviour, cDNA for a trehalose-6-phosphate synthase from Catantops pinguis (CpiTPS) was cloned and sequenced. Results revealed that the CpiTPS cDNA sequence contains an open reading frame of 2430 nucleotides encoding a protein of 809 amino acids with a predicted molecular weight of 91.13 kDa and a pI value of 6.25. Northern blot and RT-PCR analyses showed that CpiTPS mRNA expression was high in the fat body and testes, ovaries, Malpighian tubules, brain, trachea, rectum, and posterior extensor of C. pinguis. Northern blotting revealed that CpiTPS mRNA was expressed in the fat body at different developmental stages and was present at a high level in first instar larvae and adults. Travinné ekosystémy jsou významnou součástí středoevropské krajiny. Polopřirozená travinná společenstva dnes tvoří v Evropě místa s vysokou biodiverzitou, jsou velmi náchylná k degradaci a k jejich efektivní ochraně je nutné znát vlivy různých typů managementu. Neodmyslitelnými obyvateli těchto ekosystémů jsou také zástupci rovnokřídlého hmyzu (Orthoptera), kteří zde hrají důležitou roli v potravní síti. Zároveň jsou vhodným indikátorem změn stanovištních podmínek, a to zejména struktury vegetace, čehož lze využít při zjišťování dopadů různých managementových opatření. Jedním z nejběžnějších a zároveň pro hmyz nejméně vhodných typů péče je dnes mechanizovaná seč. Nevhodná péče o travinné ekosystémy může ovlivňovat druhovou bohatost jak v měřítku lokálním tak i v měřítku krajiny. Snížení negativního dopadu seče ve prospěch biodiverzity lze docílit několika způsoby - např. zmírněnou intenzitou zásahu, a to posunutím nebo vynecháním první seče, nebo ponecháním neposečených ploch., Grasslands are an important part of the Central European landscape. Seminatural grasslands are considered to be biodiversity hotspots in Europe, with high conservation value. However, they are very unstable, and for their effective conservation it is necessary to know how different management systems affect the progression of the ecosystem. Orthopterans play an important role in the food web and they are also a good indicator of changes in habitat conditions, particularly in the vegetation structure, which can be used in determining the impact of different types of conservation management.
