Trehalose is not only an important disaccharide, but also a key stress resistance factor in the development of many organisms, including plants, bacteria, fungi, and insects. To study the potential function of trehalose in development and behaviour, cDNA for a trehalose-6-phosphate synthase from Catantops pinguis (CpiTPS) was cloned and sequenced. Results revealed that the CpiTPS cDNA sequence contains an open reading frame of 2430 nucleotides encoding a protein of 809 amino acids with a predicted molecular weight of 91.13 kDa and a pI value of 6.25. Northern blot and RT-PCR analyses showed that CpiTPS mRNA expression was high in the fat body and testes, ovaries, Malpighian tubules, brain, trachea, rectum, and posterior extensor of C. pinguis. Northern blotting revealed that CpiTPS mRNA was expressed in the fat body at different developmental stages and was present at a high level in first instar larvae and adults. Species of Acanthamoeba Volkonsky, 1931 are the commonest among free-living amoebae that are widespread in different water resources but with lacking phylogenetic data. This study aims at detecting molecular prevalence and genetic diversity of Acanthamoeba isolates in Kafrelsheikh Governorate, Egypt. Forty-eight water samples were collected from 12 swimming pools; four samples during each season over one year. Samples were filtered, cultivated on non-nutrient agar plates and examined microscopically. Polymerase chain reaction (PCR) and sequence analysis of positive samples targeting diagnostic fragment 3 (DF3) of the small subunit rRNA gene were done. Cultivation succeeded to detect 14 (29%) positive samples while PCR missed three positive samples. The obtained sequences were phylogenetically analysed. The phylogenetic tree was constructed for them with sequences of reference species from the NCBI database. The identified species were Acanthamoeba castellanii Douglas, 1930 (T4), A. astronyxis (Ray et Hayes, 1954) (T9) and A. hatchetti Sawyer, Visvesvara et Harke, 1977 (T11). The prevalence of species of Acanthamoeba was higher during summer and fall. Therefore, the control of the presence of Acanthmoeba spp. in swimming pools needs immediate, effective and practical measures to prevent and control infection with species of Acanthamoeba.
In the present studies, we have found a fragment of amino acid sequence, called TFT motif, both in light-dependent protochlorophyllide oxidoreductase (LPOR) and in the L subunit of dark-operative (light-independent) protochlorophyllide oxidoreductases (DPOR). Amino acid residues of this motif shared similar physicochemical properties in both types of the enzymes. In the present paper, physicochemical properties of amino acid residues of this common motif, its spatial arrangement and a possible physiological role are being discussed. The aim of this article is to present a specific method for the study of the life-course, which focuses on life-course trajectories as a whole through the use of sequence analysis. In the first part, two approaches for the quantitative analysis of the life-course are distinguished: an event-oriented perspective and a trajectory-based (holistic) perspective. The holistic perspective is based on sequence analysis and more specifically on optimal matching. The trajectory-based perspective does not focus on single life events, but on whole sequences of events. In the second part, using the Czech wave of the ISSP 2002 dataset, which includes partnership and family histories, this article presents several examples of the use of sequence analysis of family trajectories.
