Seminal vesicle secretion is important for increasing the stability of sperm chromatin, inhibition of the immune activity in the female reproductive tract and so on. Metronidazole (MTZ), a drug used for treatment of infections caused by anaerobic bacteria and protozoa, may have negative effects on the genital gland including the seminal vesicles. Curcumin exhibits antioxidant as well as anti-inflammatory properties. The present study aims to evaluate the negative effects of MTZ on the seminal vesicle structure and ameliorative effects of curcumin using stereological methods. Thirty balb/c mice were divided into six groups. The control group was received distilled water. The second and the third received higher doses of MTZ (500 mg/kg body weight/day) and MTZ (500 mg/kg/day) + 100 mg/kg/day curcumin, respectively. The fourth and the fifth were treated with lower doses of MTZ (165 mg/kg body weight/day) and MTZ (165 mg/kg body weight/day) + curcumin (100 mg/kg body weight/day), respectively. The sixth group received 100 mg/kg body weight/day curcumin. All the administrations were done by oral gavages for 14 days. After 30 days, seminal vesicles were removed. Stereological study of the seminal vesicle structure revealed a significant reduction in gland and vesicular fluid volume in MTZ-treated (higher or lower doses) animals. Curcumin protected the reduction of both parameters in therapeutic-dose treated animals. Metronidazole treatment does not induce structural changes in the seminal gland; however, it can have a significant impact on its secretion ability. Zkoumali jsme účinky dekompresní kraniektomie u krysílio modelu „maligního" infarktu střední mozkové tepny (ACM). Celkem 55 samců krys Sprague-Dawiey o váze 290 až 340 g bylo rozděleno do těchto skupin: 1) simulovane operovaná skupina (skupina 1, n = 14, jako kontrolní); 2) skupina s okluzí ACM (OACM) (skupina 2, n = 26, zůstala neléčená po permanentní OACM); 3) skupina s dekompresní kraniektomií (skupina 3, n = 15, vhodná dekomprese do 1 hodiny po OACM). Po 48 hodinách bylo zjištěno, že dekompresní kraniektomie může znamenat jednoznačný prínos z hlediska přežití, lepších neurologických výsledků, jakož i menšího rozsahu infarktu po OACM u krys. Výrazně snižuje odumírání neuronů v kůře, ne však v hlubokých struktúrách, jako je hippocampus a striatum., We investigated the effects of decompressive craniectomy in a rat model of „malignant“ middle cerebral artery (MCA) infarction. A total of 55 male Sprague-Dawley rats weighing 290–340 g were allocated to the following groups: (1) Sham operated group (group 1, n = 14, as controls); (2) MCA occlusion (MCAO) group (group 2, n = 26, remained untreated after right permanently MCAO); (3) Decompressive craniectomy group (group 3, n = 15, received right decompression at 1 hour after MCAO). 48 hours later we found that decompressive craniectomy can offer a clear survival benefit, a better neurological score as well as a reduced infarct volume after MCAO in rats. Huntingtonova nemoc (HD) je neurodegenerativní porucha způsobená elongací CAG repetic v genu kódující protein huntingtin (Htt). U pacientů jsou v postižených tkáních přítomny vedle monomerní formy hlavně N‑koncové fragmenty, oligomery a polymery mutovaného huntingtinu (mtHtt), oproti tomu samotná monomerní forma mtHtt je exprimována v podstatě ve všech buňkách. Nejvíce postižené tkáně jsou bazální ganglia a mozková kůra. V této studii jsme analyzovali přítomnost N‑koncových fragmentů a oligomerů Htt v různých tkáních 24- a 36měsíčních transgenních (TgHD) miniprasat exprimujících N‑koncovou část lidského mutovaného huntingtinu a jejich zdravých sourozenců. Zjistili jsme, že mozková kůra a varlata na rozdíl od svalu a srdce TgHD miniprasat obsahují kromě monomerní formy i N‑koncové fragmenty a oligomerní smíry. Ve svalech z 36 měsíčních TgHD miniprasat však již začíná mírná fragmentace. Tato zjištění napodobují časnou progresi onemocnění u lidí, a proto miniprase poskytuje slibný model pro terapeutické testování HD. In patients, in addition to the monomeric form of huntingtin, N‑terminal fragments, oligomers, and polymers are present mostly in the affected tissues, even though the mutated huntingtin (mtHtt) is expressed basically in all cells. The most affected tissues are basal ganglia and cerebral cortex. In this study we analyzed the presence of N‑terminal fragments and oligomers of Htt in different tissues of 24 and 36 months old experimental animals. This was done in our large animal model of HD, which uses transgenic (TgHD) minipigs expressing N‑terminal part of human mtHtt. Among all the tissues tested, we found cortex and testes to contain N‑terminal fragments as well as oligomeric smears in TgHD minipigs compared to wild type siblings. On the other hand, we did not detect any fragments or oligomers in muscle and heart of TgHD minipigs, only starting fragmentation in muscles of 36 months old animals. Activated hepatic stellate cells (HSC) are a major source offibrous proteins in cirrhotic liver. Inducing or accelerating their apoptosis is a potential way of liver fibrosis treatment. Extracellular matrix (ECM) surrounding cells in tissue affects their differentiation, migration, proliferation and function. Type I collagen is the main ECM component in fibrotic liver. We have examined how this protein modifies apoptosis of normal rat HSC induced by gliotoxin, cycloheximide and cytochalasin D in vitro and spontaneous apoptosis of HSC isolated from CCl4-damaged liver. Background and objective: Investigation of the effects of MnTnHex-2-PyP on some markers of inflammation and lipid peroxidation in an asthma mice model. Methods: The experiment was carried out on 24 female mice C57Bl/6, divided into four groups: group 1, controls; group 2, injected with ovalbumin (OVA); group 3, treated with MnTnHex-2-PyP and group 4, treated with OVA and MnTnHex-2-PyP. The animals from groups 1 and 3 were injected i.p. on days 0 and 14 with a 100 μl phosphate-buffered saline (PBS), and those from groups 2 and 4 were injected with a 100 μl ovalbumin solution, containing 20 μg OVA. On days 24, 25 and 26 the mice from groups 1 and 2 were inhaled with PBS for 30 min, and those from groups 2 and 4 were given a 1% ovalbumin solution. One hour before inhalation, and 12 hours later the animals from groups 1 and 2 were injected i.p. with 100 μl PBS, and those from groups 3 and 4 received a 100 μl MnTnHex-2-Pyp solution in PBS сontaining 0.05mg/kg. Results: Ovalbumin alone (group 2) increased the total cell number, total protein content, the levels of IL-4, IL-5 and 8-isoprostane in bronchoalveolar lavage. Elevations were observed in IgE level in serum, and the malone dialdehyde (MDA) content in the lung homogenate. These markers were decreased significantly in group 4 as compared to the OVA group. INTRODUCTION: A previous study has employed shear-wave ultrasound elastographic imaging to assess corneal rigidity in an ex-vivo porcine eye model. This study employs the same modality in vivo in a rabbit eye model in order to assess lens, ciliary body and total ocular rigidity changes following the instillation of atropine and pilocarpine. METHODS: Ten non-pigmented female rabbits were examined. Measurements of the lens, ciliary body and total ocular rigidity as well as lens thickness and anterior chamber depth were taken with the Aixplorer system (SuperSonic Imagine, Aix-en-Provence, France) with the SuperLinear™ SL 15-4 transducer in both eyes at baseline as well as after pilocarpine and atropine instillation. The IOP was also measured with the TonoPen tonometer. RESULTS: Changes in rigidity in the examined areas following atropine instillation were statistically not significant. Ciliary body rigidity was significantly increased whereas lens and total ocular rigidity were significantly reduced following pilocarpine instillation. The decrease in lens rigidity following pilocarpine was significantly associated with the respective increase in ciliary body rigidity. CONCLUSIONS: Shear-wave ultrasound elastography can detect in vivo rigidity changes in the anterior segment of the rabbit eye model and may potentially be applied in human eyes, providing useful clinical information on conditions in which rigidity changes play an important role, such as glaucoma, pseudoexfoliation syndrome or presbyopia. and E. Užití mezenchymálních kmenových buněk (MSC) je jedním z experimentálních terapeutických přístupů k léčbě poranění míchy. MSC lze získat z kostní dřeně, tukové tkáně i z jiných periferních tkání dospělých jedinců. Výhodou je, v porovnání s jinými kmenovými buňkami, jejich dobrá dostupnost, snadná expanze a možnost autologního použití. MSC byly v posledních 15 letech studovány v experimentálním míšním poranění zejména u hlodavců se slibnými výsledky. Existuje několik mechanizmů, jakými působí MSC na míšní lézi. Především to je remyelinizace demyelinizovaných vláken, podpora pučení axonů (sprouting), angiogeneze, imunosupresivní efekt či sekrece neurotrofních faktorů, které mohou vést k funkčnímu zlepšení. Tyto slibné výsledky urychlily zahájení klinických studií u pacientů s poraněním míchy. Klinické studie fáze I/II ukázaly, že se jedná o bezpečnou metodu, avšak funkční průkaz vyžaduje další klinické studie. Ukazuje se, že MSC bude třeba kombinovat s jinými metodami, jako je přemostění léze, enzymatické štěpení jizvy a blokátory inhibičních faktorů. Tato práce přináší přehledný souhrn o aplikaci MSC u míšního poranění v experimentu i klinice., The use of mesenchymal stem cells (MSC) represents an experimental therapeutic modality in the treatment of spinal cord injury. MSC can be harvested from the bone marrow, fat tissue and other peripheral tissues from adult individuals. Compared to other types of stem cells, MSC are easy to access and expand and they can be used in autologous settings. Over the last 15 years, MSC have been widely studied in experimental spinal cord injury, especially in rodents, with promising results. MSC support remyelination of demyelinated axons, axonal sprouting, angiogenesis, have immunosuppressive effect and secrete neurotrophic factors that may led to functional improvement. These promising results led to launching of clinical studies in patients with spinal cord injury. Phase I/II clinical studies showed that the use of MSC represent a safe method. However, functional effect needs to be proved in further clinical studies. Data suggest that MSC will need to be combined with other methods, such as lesion bridging, scar tissue breakdown and blocking of inhibitory molecules. Huntingtonova nemoc (HN) je autozomálně dominantní neurodegenerativní onemocnění charakterizované poškozením volních a mimovolních pohybů, poruchami chování a zhoršením kognitivních funkcí. Společně s hlavními motorickými příznaky byly poruchy hlasu a řeči pozorovány u většiny pacientů s HN. Zvířecí prasečí model je často používán pro výzkum v preklinických studiích. I přes zjevné rozdíly v anatomii artikulačních orgánů mezi prasaty a lidmi lze očekávat stejné trendy u patofyziologických mechanizmů s ohledem na chrochtání i lidskou fonaci. Hlavním cílem této studie bylo proto navržení vhodného experimentu, který umožní získání dostatečně dlouhého záznamu chrochtání od co největšího počtu prasátek. Dalším cílem studie bylo zrealizovaní výsledné verze experimentu na celé databázi a vyhodnocení množství a kvality získaných nahrávek. Databáze použitá pro studii zahrnovala 17 HN transgenních miniprasátek a 16 zdravých sourozenců ze stejných vrhů. Testované varianty experimentu, provedené na části databáze zahrnující čtyři prasnice, byly rozděleni do čtyř podskupin: (a) pozitivní – krmení, (b) pozitivní – zvuková stimulace, (c) negativní – bránění v pohybu, (d) negativní – nepříjemné doteky. Hodnocení kvality získaných nahrávek bylo provedeno s pomocí audio softwaru, ve kterém bylo izolováno čisté prasečí chrochtání a všechny akustické artefakty vymazány. Nejlepších výsledků bylo dosaženo s použitím experimentu při kterém: (i) je záznamové zařízení upevněno na tělo prasátka, (ii) prasátko je ponecháno několik minut o samotě v místnosti, aby se uklidnilo a (iii) osoba vstoupí do místnosti a snaží se nabízet prasátku krmivo zatímco před ním couvá. V důsledku tohoto jednání prasátko následuje osobu s krmením, což je doprovázeno chrochtáním. Dostatečně dlouhé (20 chrochtnutí a více) a čisté nahrávky byly získány od 24 z 33 prasátek (73 %). Závěrem lze tedy říci, že experiment je proveditelný. Besides the main motor symptoms, voice and speech disorders have been documented in a large majority of patients with HD. The animal model of pigs is often used in preclinical studies. Although there are obvious differences in the anatomy of the articulation organs between pigs and humans, the same trends in pathophysiological mechanisms can be expected in both grunting and human phonation. The main aim of the study was therefore to design a suitable experiment that would allow for acquisition of a sufficiently long recording of grunting from as many pigs as possible. The second goal was to perform the final version of the experiment in all available pigs and to evaluate the amount and quality of the acquired recordings. The database consists of 17 HD transgenic minipigs and 16 healthy siblings. Tested variants of the experiment, performed on subgroup of four sows, were divided into four subgroups: (a) positive – feeding, (b) positive – sound stimulation, (c) negative – hindering in movement, (d) negative – unpleasant touch. The evaluation of the quality of the elicited recording was performed using audio software where pure pig grunting was selected and all acoustic artefacts deleted. The best results were reached using the experiment in which: (i) a recording device is put on the pig’s body, (ii) the pig is left alone for few minutes in the pen in order to calm down, and (iii) a person enters the room and tries to offer the pig food while walking backwards. As a result, the pig follows the person and grunts. Sufficiently long (20 single grunts or more) and clear recordings were received from 24 out of 33 pigs (73%). The realisation of the experiment is therefore possible. Skiaskopická kontrola se stala základní metodou při navigaci katétrů a provádění angiografií či měření krevních tlaků. Katétry byly zaváděny nejprve chirurgicky, od roku 1953 perkutánně po zavedení punkční techniky Seldingerem. Během šedesátých let se katétry staly chirurgickým nástrojem tím, že začaly být používány k dilataci tepen Dotterem od roku 1964. Transluminální angioplastika byla zlepšena zavedením vysokotlakých balónkových katétrů Grůntzigem v roce 1974. Embolizace provedená přes katétry k uzávěru arteriovenózního zkratu byla zavedena v roce 1960, k zástavě krvácení od roku 1972 a k selektivnímu uzávěru aneuryzmat od roku 1991., A brief history of catheterisation techniques is described. The history begun with first animal experiments in the 19th century and in beginning of 20th century. Fluoroscopy guidance has become basic method of following catheter navigation and performing angiograms or blood pressure measurements. Catheters were introduced surgically and then percutaneously after developing percutaneous approach by Seldinger in 1953. During 60. catheters became surgical instruments by using them for arterial dilatation by Dotter in 1964. Method of transluminal angioplasty has been improved by introduction of high-pressure balloon catheters by Grůntzig in 1974.
