Ascorbic acid (Asc) is a major plant antioxidant. L-galactono-1,4-lactone dehydrogenase (GLDH) is an enzyme that catalyzes the last step of Asc biosynthesis in higher plants. Effects of endogenous Asc on resistance to high-temperature stress were studied by using GLDH-overexpressed (GO-2) and GLDH-suppressed transgenic rice (GI-2) as experimental materials. After high-temperature treatment, the maximal quantum yield of PSII was significantly lower in GI-2, and higher in GO-2 compared to wild type rice. The content of reactive oxygen species (ROS) was the highest in GI-2. The higher Asc content resulted in lower lipid peroxidation in GO-2. The contents of chlorophyll, soluble proteins, and Rubisco large and small subunit were positively correlated to the Asc content. These results show that the higher Asc content reduced the accumulation of ROS and maintained the function of rice leaves. Extracellular ATP (eATP) has been considered as an important extracellular compound to mediate several physiological processes in plant cells. We investigated the effects of eATP on chlorophyll (Chl) fluorescence characteristics of kidney bean (Phaseolus vulgaris) leaves. Treatment with exogenous ATP at 1 mM showed no significant effect on the maximal photochemical efficiency of PSII. However, the treatment significantly enhanced the values of the PSII operating efficiency (ΦPSII), rate of photosynthetic electron transport through PSII (ETR), and photochemical quenching (qP), while the values of the nonphotochemical quenching (qN) and quantum yield of regulated energy dissipation of PSII (YNPQ) significantly decreased. Our observations indicated that eATP stimulated the PSII photochemistry in kidney bean leaves. Similarly, the treatment with exogenous Ca2+ or H2O2 at 1 mM caused also the significant increase in ΦPSII, qP, and ETR and the significant decrease in qN and YNPQ. LaCl3 (an inhibitor of Ca2+ channels) and dimethylthiourea (a scavenger of H2O2) abolished the effects of exogenous ATP. The aim of this study was to investigate whether haemocytes of Galleria mellonella (Lepidoptera: Pyralidae) larvae produce reactive oxygen species (ROS) like human blood phagocytes. The production of ROS was measured first using luminol-enhanced chemiluminescence of un-stimulated and stimulated (four activators with different modes of action) haemolymph or isolated haemocytes. However, spontaneous and activated production of ROS remained at the background level. In subsequent experiments an ultrasensitive fluorescence method using Amplex Red reagent to detect hydrogen peroxide (H2O2) was used. After optimization, Amplex Red was successfully used for determining H2O2 production by both un-stimulated and stimulated haemocytes. To determine the affect of pH and ions on the measurement, several diluent solutions were tested. This revealed that Ca2+ and Mg2+ ions are less important for the reaction in insect than mammalian cells. Among the activators tested, phorbol myristate acetate (PMA) and calcium ionophore (Ca-I) had the best stimulatory effect on insect samples, while opsonised zymosan particles (OZP) was the best activator for human phagocytes. The aim of this study was to characterize the key physiological aspects of three sugarcane cultivars (RB92579, RB867515 and RB872552) under biological nitrogen fixation (BNF). Plants were generated in tubes containing aseptic substrates and these plants were transferred to pots containing washed sand, but watered with a mineral fertilizer, and inoculated with a mixture of five diazotrophic bacteria three times at seven-day intervals. Under BNF, all of the cultivars contained half of their total leaf nitrogen content and 50% less shoot dry mass. The leaves of plants under BNF showed approximately 65% less of the total protein content (TP). The
gas-exchange control plants had twice the CO2 assimilation rates than the BNF plants. The activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX) was increased in all cultivars under BNF when compared with the control; thus, the content of hydrogen peroxide (H2O2) was also increased in these plants. The results of this study indicate that after acclimatization, the inoculation of young plants from tissue culture with diazotrophic bacteria could supply approximately 50% of their nitrogen requirement., C. Plants are able to acclimate to their growth light environments by utilizing a number of short- and long-term mechanisms. One strategy is to prevent accumulation of excess reactive oxygen species that can lead to photoinhibition of photosynthesis. Ureides, generated from purine degradation, have been proposed as antioxidants and involved in certain abiotic stress responses. Eutrema salsugineum (Thellungiella salsuginea) is an extremophilic plant known to exhibit a high degree of tolerance to a variety of abiotic stresses that invariably generate reactive oxygen species. In the present study we have investigated the possible role of the ureide metabolic pathway during acclimation to growth irradiance and its conference of tolerance to photoinhibition in Eutrema. Ureide accumulation was greater under high light growth which also conferred tolerance to photoinhibition at low temperature as measured by the maximal quantum yield of PSII photochemistry. This may represent an adaptive plastic response contributing to the extreme tolerance exhibited by this plant. The effects of enhanced ultraviolet-B (UV-B, 0.4 W m-2) irradiance and nickel (Ni, 0.01, 0.10 and 1.00 mM; Ni0.01, Ni0.10, Ni1.00, respectively) treatment, singly and in combination, on growth, photosynthetic electron transport activity, the contents of reactive oxygen species (ROS), antioxidants, lipid peroxidation, and membrane leakage in soybean seedlings were evaluated. Ni0.10 and Ni1.00 and UV-B declined the growth and chlorophyll content, which were further reduced following combined exposure. Contrary to this, Ni0.01 stimulated growth, however, the effect together with UV-B was inhibitory. Carotenoids showed varied response to both the stresses. Simultaneous exposure of UV-B and Ni as well as UV-B alone reduced the activities of photosystems 1 and 2 (PS1 and PS2) and whole chain activity significantly, while Ni individually, besides strongly inhibiting PS2 and whole chain activity, stimulated the PS1 activity. Both the stresses, alone and together, enhanced the contents of superoxide radical (O2⋅-), hydrogen peroxide (H2O2), malondialdehyde (MDA), electrolyte leakage, and proline content, while ascorbate content declined over control. Individual treatments increased the activities of catalase (CAT), peroxidase, and superoxide dismutase (SOD), but Ni1.00 declined SOD activity significantly. Combined exposure exhibited similar response, however, CAT activity declined even more than in control. Compared to individual effects of UV-B and Ni, the simultaneous exposure resulted in strong inhibition of photosynthetic electron transport and excessive accumulation of ROS, thereby causing severe damage to soybean seedlings. and S. Phagocyte released reactive oxygen species are often discussed in connection with ischemic and reperfusion injuries to the myocardium. The kinetics of the accumulation and oxidative burst of human blood phagocytes was studied by chemiluminescence during open heart surgery in the myocardium of human patients. Direct evidence is presented for an accumulation of neutrophils along with their markedly increased metabolic activity (oxygen radical formation), especially following the reperfusion of the ischemic myocardium. Leukocyte numbers and activity remained significantly elevated even in the venous blood obtained 24 h after the operation.
In plants, hydrogen peroxide (H2O2) acts as a signalling molecule that facilitates various biochemical and physiological processes. H2O2 is a versatile molecule, involved in several cellular processes both under stress and stress-free conditions. In regulating plant metabolism under stress conditions, exogenous application of H2O2 also plays a pivotal role which is manifested in improved growth, photosynthetic capacity, and antioxidant protection. Abiotic stress is an inevitable environmental factor that extensively affects and reduces growth, quality, yield, and productivity of plants. Several signalling pathways involved in H2O2-mediated stress and defense responses have been extensively studied and there is ample scope of additional research that could further clarify the mechanism and modulating factors which regulate these pathways. Several recent studies bring evidence of cell death enhancement in photodynamic compound loaded cells by ultrasonic treatment. There are a number of hypotheses suggesting the mechanism of the harmful ultrasonic effect. One of them considers a process in the activation of photosensitizers by ultrasonic energy. Because the basis of the photodynamic damaging effect on cells consists in the production of reactive oxygen species (ROS), we focused our study on whether the ultrasound can increase ROS production within cancer cells. Particularly, we studied ROS formation in ultrasound pretreated breast adenocarcinoma cells during photodynamic therapy in the presence of chloroaluminum phthalocyanine disulfonate (ClAlPcS2). Production of ROS was investigated by the molecular probe CM-H2DCFDA. Our results show that ClAlPcS2 induces higher ROS production in the ultrasound pretreated cell lines at a concentration of 100 μM and light intensity of 2 mW/cm2. We also observed a dependence of ROS production on photosensitizer concentration and light dose. Climate change impacts environmental conditions that affect photosynthesis. This review examines the effect of combinations of elevated atmospheric CO2, long photoperiods, and/or unfavorable nitrogen supply. Under moderate stress, perturbed plant source-sink ratio and redox state can be rebalanced but may result in reduced foliar protein content in C3 plants and a higher carbon-to-nitrogen ratio of plant biomass. More severe environmental conditions can trigger pronounced photosynthetic downregulation and impair growth. We comprehensively evaluate available evidence that microbial partners may be able to support plant productivity under challenging environmental conditions by providing (1) nutrients, (2) an additional carbohydrate sink, and (3) regulators of plant metabolism, especially plant redox state. In evaluating the latter mechanism, we note parallels to metabolic control in photosymbioses and microbial regulation of human redox biology.
