The photosynthetic gas-exchange has been assessed traditionally either as O2 evolution or CO2 consumption. In this study, we used a liquid-phase O2 electrode combined with CO2 optodes to examine simultaneously photosynthesis in intact leaves of mangrove Rhizophora mucronata. We verified suitable conditions for leaf photosynthetic rates by assessing pH levels and NaHCO3 concentrations and compared these to the gas-exchange method at various PAR levels. The photosynthetic rate in response to pH exhibited a similar pattern both for O2 evolution and CO2 consumption, and higher rates were associated with intermediate pH compared with low and high pH values. The net photosynthetic quotient (PQ) of R. mucronata leaves ranged from 1.04-1.28. The PQ values, which were never lesser than 1, suggested that photorespiration did not occur in R. mucronata leaves under aqueous conditions. The similar maximum photosynthetic rates suggested that all measurements had a high capacity to adjust the photosynthetic apparatus under a light saturation condition. Nitrogen (N) starvation resulted in degreening, inhibition of photosynthetic oxygen evolution and dark respiration, reduced survival, and increased age-specific mortality in both Chlorella fusca and Chlorella vulgaris. Analysis of in vivo chlorophyll (Chl) fluorescence induction kinetics revealed the presence of N-starvation-induced changes at the level of degreened thylakoids in both species. These changes included decreased yield of the photochemistry of photosystem 2 (PS2), and a declined photosynthetic efficiency. Synthesis of secondary carotenoids represented a biochemical change in carotenogenesis that had a photoprotective effect in degreened C. fusca. This inferred photoprotection was reflected in the delayed inhibition of oxygen evolution and improved survival of C. fusca under N-starvation. The effect was further elucidated by comparison with C. vulgaris which was not able to synthesize secondary carotenoids under the same conditions.
The pericarp of cereal crops is considered a photosynthetically active tissue. Although extensive studies have been performed on green leaves, the photosynthetic role of the pericarp in cereal caryopsis development has not been well investigated. In the present study, we investigated the anatomy, ultrastructure, chlorophyll (Chl) fluorescence, and oxygen evolution of the pericarp during caryopsis ontogenesis in field wheat (Triticum aestivum L.). The results showed that wheat pericarp cross-cells contained Chl; the grana stacks and thylakoid membranes in the cross-cells were more distinct in the pericarp than those in the flag leaves as shown by transmission electron microscopy. Chl fluorescence revealed that the photosynthetic efficiency, which was indicated by values of maximum efficiency of PSII photochemistry and effective PSII quantum yield, was lower in the pericarp compared to that of the flag leaf eight days after anthesis (DAA), whereas similar values were subsequently observed. The nonphotochemical quenching values were lower from 8-16 DAA but significantly increased in the pericarp from 24-32 DAA compared to the flag leaf. The oxygen evolution rate of the flag leaves was consistently higher than that of pericarp; notably, isolated pericarps released more oxygen than intact pericarps during caryopsis development. These results suggest that the pericarp plays a key role in caryopsis development by performing photosynthesis as well as by supplying oxygen to the endosperm and dissipating excessive energy during the

Nostoc muscorum cells showed metal-induced decrease in the relative growth, pigment contents, O2 evolution, and Hill activity in response to lead (Pb2+) and cadmium (Cd2+) treatment, which was further accentuated with increase in metal exposure time and metal concentration. I50 concentrations (50% growth inhibitory concentrations) of Pb2+ and Cd2+ for growth of N. muscorum were 55 and 21 μg mL-1, respectively. These results indicated that the cells of N. muscorum were more susceptible to Cd2+ in comparison to Pb2+. The O2 production was relatively more sensitive to both heavy metals (I50: 16 and 10 μg mL-1 of Pb2+ and Cd2+, respectively) than the Hill activity (I50: 61 and 39 μg mL-1 of Pb2+ and Cd2+, respectively). Further, measurement of Hill activity in the presence of metals and electron donors showed that inhibition sites of both Pb2+ and Cd2+ were located on the oxidizing site of PSII. The chlorophyll a (Chl a) and phycobilisome (PB) fluorescence emission spectra showed that energy transfer from Chl a and PB to PSII reaction center was more susceptible to Cd2+ than Pb2+., S. Membrane-bound bicarbonate is believed by some to act as an essential activator of photosystem 2 (PS2) electron transport. Formáte and other inhibitory monovalent anions act by removing bound-bicarbonate. This belief relies to a great extent on the observation that formáte (100 mM) pretreated thylakoids exhibit a non- proportionality between Hill activity (HAR) and chlorophyll (Chl) concentration when preirradiated with bright radiation in reaction mixture that contains only 5 mM formáte. The non-linearity was attributed to a supposed loosening of residual bicarbonate still present after formáte treatment and which would be more abundant at higher Chl concentrations. In repeating this experiment, we observed an increase in HAR at higher Chl concentrations in preirradiated, but also in non-preirradiated samples, the latter were simply left in the dark for 3 min before measurements were made. Therefore, preirradiation is not needed to restore some HAR in formáte pretreated samples; a 3 min wait in the electrode chamber at low formáte concentration is sufficient to partially relieve the formáte inhibition of PS2 activity. Moreover, HAR in samples preirradiated by weak radiation, or not preirradiated at all, was directly proportional to Chl concentration. We can attribute the increase in activity to a dissociation of bound formáte, not necessarily to the effect of residual bicarbonate. Non-linearity in HAR with Chl concentration was found only in high- irradiance pretreated samples. We can attribute this to a greater amount of photoinhibition occurring in the dilute samples, where the effective irradiance was greater. There is no need to postuláte the existence of residual bound bicarbonate to explain these results.
The effects of metabolisable sugars sucrose and glucose along with non-metabolisable isomers of sucrose palatinose and turanose were tested. Rate of oxygen evolution (P), electron transport rate (ETR), and photochemical quenching (qp) showed substantial decrease after 24 and 48 h by glucose and sucrose treatments, whereas there was no effect on all these parameters by the treatment with palatinose and turanose. Also the Fv/Fm ratio remained constant through the time of studies revealing that the maximal photochemical capacity of the cells was unchanged. Non-photochemical quenching (qN) showed a decrease compared to the control values by all the treatments. Lowering irradiance can delay the flower stalk, i.e., spike development, in order to schedule flowering time of Phalaenopsis; however, the effect on photosynthetic performance and spiking inhibition remains poorly understood. We compared light and shade treatments of Phalaenopsis aphrodite subsp. formosana in order to determine how limiting light affects day-night changes in the photosynthetic capacity of leaves and the carbon pool of leaves and stems resulting in delayed spiking. The low irradiance treatment [20 μmol(photon) m-2 s-1] for six weeks did not affect potential functions of photosynthetic apparatus estimated by chlorophyll a fluorescence analysis, but it significantly reduced the net CO2 uptake and O2 evolution rates, carbohydrate and organic acid concentrations, and amplitudes of CAM activity in new and fully expanded leaves of Phalaenopsis and delayed the spiking compared with the control kept at 150 μmol(photon) m-2 s-1. The shortened stem contained a remarkably high sucrose concentration, accounting for more than 80% of total soluble sugars for both treatments throughout the day. Moreover, the sucrose concentration was unaffected by the lowering of irradiance. Fluorescence spectroscopy at 77 K showed that the application of glucose lead to the depletion of phycobilisomes (PBS) and photosystems (PS) 2 and 1, and that PS2 was more sensitive to glucose than PS1. The application of sodium thiosulfate, an effective scavenger of reactive oxygen intermediates, counteracted the effects of glucose. Sodium thiosulfate effectively protected photosynthetic apparatus, PS2, PS1, and PBS against glucose-induced depletion. Sodium thiosulfate showed strong capability to inhibit the disappearance of chlorophyll induced by glucose. At a relatively low concentration of glucose, the application of sodium thiosulfate can even be helpful for the assembly of photosynthetic apparatus. Hence the reactive oxygen species might be involved in the depletion of the photosynthetic apparatus in the cyanobacterium Synechocystis sp. Ionic environment is important in regulating photosynthetic reactions. The roles of cations, Mn2+, Mg2+, Ca2+, Na+, and K+ as cofactors in electron transport, energy transfer, phosphorylation, and carbon assimilation are better known than the roles of anions, except for chloride and bicarbonate. Only a limited information exists on the roles and effects of nitri formate, sulphate, and phosphate. In this review, we evaluate and highlight the roles of some specific anions on electron transport as well as on excitation energy transfer processes in photosynthesis. Anions exert significant effects on thyla membrane conformation and membrane fluidity, possibly by redistributing the thylakoid membrane surface charges. The anion/cation induced phase transitions in the hydrophilic domains of the thylakoid membranes are probably responsible for the various structural and co-related functional changes under stress. Anions are also important in regulation of energy distribution between the two photosystems. Anions do not only divert more energy from photosystem (PS) 2 to PS1, but can also reverse the effect of cations on energy distribution in a valence-dependent manner. The efficiency in selective extraction of photosystem (PS) 2 oxygen evolving complexes was compared among seven detergents. These were applied to thylakoid membranes of the thermophilic cyanobacterium Synechococcus elongatus. Used were five non-ionic detergents with one ionic and one zwitterionic for comparison. To compare the suitability and efficiency of the detergents the following properties of the extracts were examined: maximum rate of oxygen evolution with various electron acceptors, the relative variable fluorescence (FV/FM), the contamination of the extract with photosystem (PS) 1, and the status of the electron acceptor side of PS2 reaction centre. None of the detergents yielded a highly selective extraction of the PS2 complexes (negligible contamination with PS1) which would simultaneously display a high photochemical activity and high structural intactness. Heptylthioglucoside and dodecylmaltoside yielded the nearest approximation to the optimum result.
