The photosynthetic response of three Arachis hypogaea L. cultivars (57-422, 73-30, and GC 8-35) grown for two months was measured under water available conditions, severe water stress, and 24, 72, and 93 h following re-watering. At the end of the drying cycle, all the cultivars reached dehydration, relative water content (RWC) ranging between 40 and 50 %. During dehydration, leaf stomatal conductance (gs), transpiration rate (E), and net photosynthetic rate (PN) decreased more in cvs. 57-422 and GC 8-35 than in 73-30. Instantaneous water use efficiency (WUEi) and photosynthetic capacity (Pmax) decreased mostly in cv. GC 8-35. Except in cv. GC 8-35, the activity of photosystem 1 (PS1) was only slightly affected. PS2 and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) were the main targets of water stress. After re-watering, cvs. 73-30 and GC 8-35 rapidly regained gs, E, and PN activities. Twenty-four hours after re-watering, the electron transport rates and RuBPCO activity strongly increased. PN and Pmax fully recovered later. Photoinhibition of photosynthesis was investigated in grapevine (Vitis vinifera L.) exposed to 2 or 4h of high irradiance (HI) (1 700-1 800 μmol m-2 s-1) leaves under field conditions at different sampling time in a day. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) and photosynthetic electron transport measurements. When the photochemical efficiency of photosystem 2 (PS2), Fv/Fm, markedly declined, F0 increased in both 2 (HI2) and 4 h (HI4) HI leaves sampled at midday. When various photosynthetic activities were followed on isolated thylakoids, HI4 leaves showed significantly higher inhibition of whole chain and PS2 activity than the HI2 leaves sampled at midday. Later, the leaves reached maximum PS2 efficiencies similar to those observed early in the morning during sampling at evening. The artificial exogenous electron donor Mn2+ failed to restore PS2 activity in both variants of leaves, while DPC and NH2OH significantly restored PS2 activity in HI4 midday leaf samples. Quantification of the PS2 reaction centre protein D1 and 33 kDa protein of water splitting complex following midday exposure of leaves showed pronounced differences between HI2 and HI4 leaves. The marked loss of PS2 activity noticed in midday samples was mainly due to the marked loss of D1 protein in HI2, while in HI4 it was mainly 33-kDa protein. and M. Bertamini, N. Nedunchezhian.

A smaller inhibition of PS1 activity was also observed in both leaf types. Later, the leaves reached maximum PS2 efficiencies similar to those observed in the morning during sampling at evening. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the PS2 activity in both leaf types at midday. Thus CN enhanced inactivation on the acceptor side of PS2 in grapevine leaves. The degree of photoinhibition of sun and shade grown leaves of grapevine was determined by means of the ratio of variable to maximum chlorophyll (Chl) fluorescence (Fv/Fm) and electron transport measurements. The potential efficiency of photosystem 2 (PS2), Fv/Fm, markedly declined under high irradiance (HI) in shade leaves with less than 10 % of F0 level. In contrast, Fv/Fm ratio declined with about 20 % increase of F0 level in sun leaves. In isolated thylakoids, the rate of whole chain and PS2 activity in HI shade and sun leaves was decreased by about 60 and 40 %, respectively. A smaller inhibition of photosystem 1 (PS1) activity was also observed in both leaf types. In the subsequent dark incubation, fast recovery was observed in both leaf types that reached maximum PS2 efficiencies similar to non-photoinhibited control leaves. The artificial exogenous electron donors DPC, NH2OH, and Mn2+ failed to restore the HI-induced loss of PS2 activity in sun leaves, while DPC and NH2OH were significantly restored in shade leaves. Hence HI in shade leaves inactivates on the donor side of PS2 whereas it does at the acceptor side in sun leaves, respectively. Quantification of the PS2 reaction centre protein D1 and the 33 kDa protein of water splitting complex following HI-treatment of leaves showed pronounced differences between shade and sun leaves. Seedlings of tropical leguminous tree Samanea saman (Jacq.) Merrill were exposed for 7 d to acidic mist (AM, induced by H2SO4) of pH 5.6, 4.0, and 2.0. AM significantly reduced seedling growth (root and shoot length, leaf density, leaf area, fresh and dry mass accumulation) and photosynthetic activities. In thylakoids isolated from leaves treated at pH 4.0 and 2.0 a decrease in the activities of photosystem (PS) 2 and whole chain electron transport was observed, but PS1 activity did not change. When the seedlings were subsequently sprayed with triacontanol (TRIA), the AM effect was partially or completely reversed indicating that TRIA can protect from AM effects. The artificial electron donors, di-phenylcarbazide (DPC) and hydroxylamine (NH2OH), markedly restored the loss of PS2 activity in AM (pH 2.0) treated leaves. Regulation mechanism of excitation energy transfer between phycobilisomes (PBS) and the photosynthetic reaction centres was studied by the state transition techniques in PBS-thylakoid membrane complexes. DCMU, betaine, and N-ethylmaleimide were applied to search for the details of energy transfer properties based on the steady fluorescence measurement and individual deconvolution spectra at state 2 or state 1. The closure of photosystem (PS) 2 did not influence on fluorescence yields of PS1, i.e., energy could not spill to PS1 from PS2. When the energy transfer pathway from PBS to PS1 was disturbed, the relative fluorescence yield of PS2 was almost the same as that of PS2 in complexes without treatment. If PBSs were fixed by betaine, the state transition process was restrained. Hence PBS may detach from PS2 and become associated to PS1 at state 2. Our results contradict the proposed "spill-over" or "PBS detachment" models and support the mobile "PBS model". and Ye Li ... [et al.].
Intact cells of Synechococcus elongatus were treated with different concentrations (0.1 and 1.0 mM = Cd0.1, Cd1.0) of CdCl2 for 24 h. Cd0.1 treatment stimulated growth of the cell culture and chlorophyll (Chl) a concentration in the culture. Cd1.0 inhibited both the above mentioned parameters. The oxygen evolving activity of intact cells (H2O → BQ) as well as of isolated thylakoid membranes, TM (H2O → DCPIP; H2O → PBQ + FeCy) decreased after 24 h of Cd1.0 cultivation to 7 %. Photosystem 1 (PS1) activity was less sensitive to the effect of Cd2+ than PS2 activity. CdCl2 concentration in cultivation media after 24 h of cultivation proved that the cyanobacterium cells take up these ions to a large extent from the cultivation medium. After 24 h of the Cd1.0 treatment only 12 % of the amount of Cd2+ originally added to the cultivation medium was found. The ratio of external-antenna pigments, phycocyanin, and allophycocyanin to Chl increased approximately twofold with growing Cd2+ concentration in the cultivation medium. Over-expression of chloroplastic glycerol-3-phosphate acyltransferase gene (LeGPAT) increased unsaturated fatty acid contents in phosphatidylglycerol (PG) of thylakoid membrane in tomato. The effect of this increase on the xanthophyll cycle and chloroplast antioxidant enzymes was examined by comparing wild type (WT) tomato with the transgenic (TG) lines at chilling temperature (4 °C) under low irradiance (100 µmol m-2 s-1). Net photosynthetic rate and the maximal photochemical efficiency of photosystem (PS) 2 (Fv/Fm) in TG plants decreased more slowly during chilling stress and Fv/Fm recovered faster than that in WT plants under optimal conditions. The oxidizable P700 in both WT and TG plants decreased during chilling stress under low irradiance, but recovered faster in TG plants than in the WT ones. During chilling stress, non-photochemical quenching (NPQ) and the de-epoxidized ratio of xanthophyll cycle in WT plants were lower than those of TG tomatoes. The higher activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in TG plants resulted in the reduction of O2-. and H2O2 contents during chilling stress. Hence the increase in content of unsaturated fatty acids in PG by the over-expression of LeGPAT could alleviate photoinhibition of PS2 and PS1 by improving the de-epoxidized ratio of xanthophyll cycle and activities of SOD and APX in chloroplast. and N. The effect of heat stress (35 to 50 °C) on photosynthesis was investigated in heat tolerant (N 22) and heat sensitive (IR 8) cultivars of rice {Oryza sativa L.). The net photosynthetic rate showed greater thermal stability in N 22 than in IR 8. The relative dechne of the rate of whole chain electron transport and photosystem 2 (PS2) activity was more pronounced in IR 8 than N 22. In both cultivars photosystem (PSI) activity was stimulated by thermal treatment. Chlorophyll (Chl) a fluorescence transient arising ffom PS2 showed inhibition in both cultivars at 45 and 50 °C. Maximum fluorescence decreased more in IR 8 than in N 22 by high temperature treatment.
