Triiodothyronine administration before partial hepatectomy increased the activity of mitochondrial glycerophosphate cytochrome c reductase. The enzyme activity was further activated after partial hepatectomy during the regenerative process. Our findings showed that: a) the increase of glycerophosphate cytochrome c reductase induced by triiodothyronine was further potentiated by the regeneration process, b) the high activity of the glycerophosphate shuttle was maintained after partial hepatectomy during the period, when most of the liver tissue had again been recovered., H. The effect of propylthiouracil (PTU) on the growth activity of intact liver and liver regenerating after partial (65-70 %) hepatectomy (PH) was studied in rats. FEU (Propycil, Kali-Chcmie, FRG) was dissolved in drinking water (1 g PTU per litre) and this was given to the rats, as their sole source of fluids, three days before PI I and then up to the end of the experiment. In rats given FPU, marked inhibition of liver DNA synthesis and the mitotic activity of hepatocytes was found after PH. This effect was potentiated to some extent by partial inanition of the experimental animals given FITJ, as demonstrated in a paired feeding test in control rats. PTU inhibition of DNA synthesis in intact and regenerating liver also took effect in thyroidectomized rats, even with substitution (thyroid hormone) therapy. The experiments demonstrated that the effect of propylthiouracil on DNA synthesis in the liver is mediated primarily by way of its direct effect on the liver.
Beta-hydroxy-beta-methylbutyrate (HMB) is a leucine metabolite with protein anabolic effects. Since HMB is synthesized in the liver, unique effects of exogenous HMB intake may be hypothesized in subjects with liver disease, in which muscle wasting is frequently found. We studied effects of HMB on the liver and soleus (SOL) and extensor digitorum longus (EDL) muscles in partially-hepatectomized (PH) rats. HMB or saline was infused using osmotic pumps to PH or sham-operated rats for 7 days. We found lower body weight and protein content in EDL of PH rats treated with saline than in sham-operated animals. These effects were insignificant in HMB treated animals. In blood plasma of PH rats treated with HMB we found lower concentrations of creatinine and higher concentrations of urea and branched-chain amino acids (BCAA; valine, leucine, and isoleucine) than in PH animals treated with saline. HMB increased BCAA concentrations in SOL and EDL of PH animals and decreased proteolysis in EDL of both sham-operated and PH animals. In the livers of PH rats treated with HMB we found higher DNA content, DNA fragmentation, and BCAA concentrations than in saline-treated animals. The results indicate that HMB affects metabolism of BCAA and has positive influence on protein balance in muscles. Further studies are needed to clarify the effect of HMB on liver regeneration., M. Liver silicosis was induced in rats by an intravenous injection of silica particles. The presence of silicotic granulomas in the liver lowered the initial response of hepatocytes to partial (37 %) hepatectomy and slowed down the decline in the number of dividing cells 6 days after the stimulus, but did not affect significantly the total number of mitotic cells.
The effect of aging (2-14 months) and total body irradiation (5.7 Gy of gamma radiation) on liver regeneration was investigated in rats 30 h after partial hcpatcctomy. Exposure of rats to irradiation 30 min before partial hepatectomy caused latent injury in the remaining liver cells. During the course of liver regeneration this became manifested as a delay in increasing the nucleic acid concentration and content and liver weight and, furthermore, as inhibition of the increase in the mitotic index and ccllularity and pronounced increase in the frequency of chromosome aberrations in the postmetaphase. The pattern of age-related changes during liver regeneratin was the same as that after irradiation, so that the differences between irradiated and nonirradiated animals became smaller with age.
The recovery of total DNA content and recovery of total cytochrome c oxidase activity in the rat liver after partial hepatectomy is accelerated by triiodothyronine applied in three doses, two before and one immediately after liver resection. Triiodothyronine-treated animals already have higher cytochrome c oxidase activity before resection. The recovery of the tissue oxidative capacity after partial hepatectomy is more rapid in triiodothyronine-treated animals. These data indicate that hormonal activation of the liver regeneration process is involved.
Decades of liver regeneration studies still left the termination phase least elucidated. However regeneration ending mechanisms are clinicaly relevant. We aimed to analyse the timing and transcriptional control of the latest phase of liver regeneration, both controversial. Male Wistar rats were subjected to 2/3 partial hepatectomy with recovery lasting from 1 to 14 days. Time-series microarray data were assessed by innovative combination of hierarchical clustering and principal component analysis and validated by real-time RT-PCR. Hierarchical clustering and principal component analysis in agreement distinguished three temporal phases of liver regeneration. We found 359 genes specifically altered during late phase regeneration. Gene enrichment analysis and manual review of microarray data suggested five pathways worth further study: PPAR signalling pathway; lipid metabolism; complement, coagulation and fibrinolytic cascades; ECM remodelling and xenobiotic biotransformation. Microarray findings pertinent for termination phase were substantiated by real-time RT-PCR. In conclusion, transcriptional profiling mapped late phase of liver regeneration beyond 5th day of recovery and revealed 5 pathways specifically acting at this time. Inclusion of longer post-surgery intervals brought improved coverage of regeneration time dynamics and is advisable for further works.
