Short-winged (brachypterous) and long-winged (macropterous) adult females of Pyrrhocoris apterus (L.) originating from a Mediterranean population (Israel) were analyzed for their adipokinetic responses. The adipokinetic response, expressed as an increase of haemolymph lipids after injection of adipokinetic hormone from Locusta migratoria (Lom-AKH-I), was assessed in relation to age and dose of the hormone. We demonstrate in this study that the adipokinetic responses induced by bug's corpora cardiaca extract and Lom-AKH-I were dose-dependent for both brachypterous and macropterous females. Significant differences between the morphs were recorded for doses >= 0.25 corpora cardiaca equivalent (P < 0.01) and doses >= 0.25 pmol Lom-AKH-I (P < 0.05). The haemolymph lipid elevations induced in both morphs by 2 pmols Lom-AKH-I are comparable with that induced by crude extract of one pair of the bug's own corpora cardiaca. The age-dependent test showed much higher adipokinetic responses (P < 0.01) in macropterous females (>= 5 days old) than in the brachypterous females of the same age, when treated with 2 pmols Lom-AKH-I. Starting from day 7, the concentration of haemolymph lipids was also considerably higher (2.5-5 times) in macropterous females than in the brachypterous ones. Hexamerins are hemocyanin-related haemolymph proteins that are widespread in insects and may accumulate to extraordinarily high concentrations in larval stages. Hexamerins were originally described as storage proteins that provide amino acids and energy for non-feeding periods. However, in recent years other specific functions like cuticle formation, transport of hormones and other organic compounds, or humoral immune defense have been proposed. During evolution, hexamerins diversified according to the divergence of the insect orders. Within the orders, there is a notable structural diversification of these proteins, which probably reflects specific functions. Gain-of-function (GOF) mutations in ion channels are rare events, which lead to increased agonist sensitivity or altered gating properties, and may render the channel constitutively active. Uncovering and following characterization of such mutants contribute substantially to the understanding of the molecular basis of ion channel functioning. Here we give an overview of some GOF mutants in polymodal ion channels specifically involved in transduction of painful stimuli - TRPV1 and TRPA1, which are scrutinized by scientists due to their important role in development of some pathological pain states. Remarkably, a substitution of single amino acid in the S4-S5 region of TRPA1 (N855S) has been recently associat ed with familial episodic pain syndrome. This mutation increases chemical sensitivity of TRPA1, but leaves the voltage sensitivit y unchanged. On the other hand, mutations in the analogous regi on of TRPV1 (R557K and G563S) severely affect all aspects of channel activation and lead to spontaneous activity. Comparison of the effects induced by mutations in homologous positions in different TRP receptors (or more generally in other distan tly related ion channels) may elucidate the gating mechanisms conserved during evolution., S. Insect peptidyl-dipeptidase A [angiotensin I - converting enzyme (ACE)] is a soluble single-domain peptidyl-dipeptidase that has many properties in common with the C-domain of mammalian somatic ACE and with the single-domain mammalian ACE. In agreement with a variety of insects, immunocytochemical studies reveal the presence of an ACE-like protein in Locusta migratoria. ACE-like immunoreactivity is present in neurosecretory cells of the pars intercerebralis. These cells have axons projecting into the nervus corporis cardiaci I and into the storage part of the corpus cardiacum, a neuroendocrine organ directly releasing into the aorta. This paper focuses on the roots of a functional theory of predication, which is represented primarly by Frege and Russell. After a brief presentation of the theory of Frege, I concentrate on the philosophical motivation of this theory. The example of the influence of F. H. Bradley on Russell’s conception of the categorical judgements shows a common epistemological position of both authors, which I recognize also in Frege. The point of the article is to find common grounds in Kant noetics, especially in his conception of synthetic judgement. Replacement of the problematic Kant’s notion of transcendental schema by the functional application subsequently allowed flourishing of the theory. In conclusion, I outline potential problems associated with the challenge of philosophical assumptions on which this theory is based., Příspěvek se zaměřuje na kořeny funkční teorie predikce, kterou reprezentují především Frege a Russell. Logical analysis of natural language (LANL) based on TIL defines meaning (Frege’s sense) in a procedural way, resulting in the following thesis: The meaning of an expression is independent of context. The meaning (in the case of non-indexical expressions concept) is thus an abstract procedure, which is explicated in TIL as (well-defined) construction. What does depend on context is the way in which the meaning has to be handled.
