Short-winged (brachypterous) and long-winged (macropterous) adult females of Pyrrhocoris apterus (L.) originating from a Mediterranean population (Israel) were analyzed for their adipokinetic responses. The adipokinetic response, expressed as an increase of haemolymph lipids after injection of adipokinetic hormone from Locusta migratoria (Lom-AKH-I), was assessed in relation to age and dose of the hormone. We demonstrate in this study that the adipokinetic responses induced by bug's corpora cardiaca extract and Lom-AKH-I were dose-dependent for both brachypterous and macropterous females. Significant differences between the morphs were recorded for doses >= 0.25 corpora cardiaca equivalent (P < 0.01) and doses >= 0.25 pmol Lom-AKH-I (P < 0.05). The haemolymph lipid elevations induced in both morphs by 2 pmols Lom-AKH-I are comparable with that induced by crude extract of one pair of the bug's own corpora cardiaca. The age-dependent test showed much higher adipokinetic responses (P < 0.01) in macropterous females (>= 5 days old) than in the brachypterous females of the same age, when treated with 2 pmols Lom-AKH-I. Starting from day 7, the concentration of haemolymph lipids was also considerably higher (2.5-5 times) in macropterous females than in the brachypterous ones. Effects of two adipokinetic hormones (Pyrap-AKH and Peram-CAH-II) on the presence of diacylglycerol (DG) molecular species and their fatty acid (FA) constituents in the haemolymph of the firebug Pyrrhocoris apterus were investigated using liquid chromatography (HPLC) and electrospray ionization mass spectrometry (ESI-MS). The results show that DGs with characteristic FAs are preferentially mobilized from the fat body (FB) by the action of both the AKHs produced by P. apterus. Both the macropterous and brachypterous morphs have similar DG and FA profiles. A difference in the action of the Pyrap-AKH and the Peram-CAH-II, however, results in distinct differences in the distribution of FAs in the macropterous morph. It seems that C16 to a slight extent and unsaturated C18 FAs mainly play a dominant role in the AKH based action, in particular linoleic acid (18:2), which represents 50-60% of the total DG mobilized. The metabolically active C16 and C18 FAs are preferentially absorbed from the linden seeds and accumulated in the FB. The relationships between AKH action and FA distribution in DGs in P. apterus, compared to other insect species are summarized and discussed in detail.
Genetic characteristics of the first three mutants found in P. apterus L.; white (w/w) 1965, yellow (y/y) 1966 and melanotic (m/m) 1973 have been described in detail. Exact Mendelian proportions of 1 : 1 and 3 : 1 in all standard test crosses and absence of sexual linkage revealed that each of these mutations was inherited as a single autosomal recessive gene. The dihybrid and trihybrid crosses showed that the w gene is epistatic over y. The absence of linkage shows that each of the described mutant genes is situated on a different chromosome. During 30 years of sustained rearings of P. apterus, the white (w/w) and yellow (y/y) mutants never originated de novo, whereas the melanotic (m/m) mutants originated independently from the macropterous strain three times. Triple recessive (w y m) white melanotic strain has been maintained and used for some genetic investigations for over 20 years.
The effects of body size on mating success and duration of copulation were investigated in Pyrrhocoris apterus. Under laboratory conditions relative mating success of small and large males was investigated in groups of 2 males (majority sex) : 1 female (minority sex). Large males were recorded significantly more often in copulation than small males when the female was large but not when the female was small. This was also the case when the group size was 24 males : 12 females or population density decreased by increasing the area of the experimental arena. In groups of 2females : 1 male, large females were recorded in copulation significantly more often than small females when the male was large but not when the male was small. Proportion of individuals of the minority sex that copulated was similar for males and females, regardless of body size. The average duration of copulation was similar for small and large males, but shorter for small than large females. The difference in the duration of copulation of small and large females was greater when it was with small rather than large males. The prolonged guarding of large females by small males may be explained by a trade-off between increasing the probability of inseminating an uneasily accessible high quality partner and copulating with more females.
Survival and Lt50 after exposures at constant low temperature were compared to the values obtained at alternating temperatures in two active (summer acclimated) temperate terrestrial arthropods. The experimental regimes used interruptions - daily transfers from the lower temperature to various higher temperatures for two hours or to one high temperature for Various durations. In both species the alternating conditions improved survival, implying reparation of the chill injury. In the collembolan Orchesella cincta, there was a maximum Lt50 when the higher exposure temperature was equal to the temperature of rearing (19°C). In the bug Pyrrhocoris apterus, Lt50 increased strongly with increasing higher temperature from 0 to 15°C, and was subsequently constant over the entire physiological range suitable for development (to 35°C). The cold hardiness of individuals from overwintering populations of a freeze susceptible bug Pyrrhocoris apterus from central and southern Europe differed significantly. Supercooling point (SCP) correlated well with both lethal temperature (LT50) and lethal time (Lt50), and is agood index of cold hardiness of adults during and after diapause. In January, diapause terminated, but cold hardiness was similar to that recorded in November; cold hardiness decreased slightly in March and markedly in May. Short exposure (less than a week) to higher temperatures before termination of diapause did not reduce the cold hardiness. Cold hardiness did not closely follow air temperatures.The Bulgarian bugs retained lower cold hardiness regardless of acclimation to harsh field conditions in the Czech Republic. The interpopulation difference is therefore a heritable character representing an adjustment to local climates.
In the present study the temporal changes in the volume of the corpus allatum in three experimental groups of adult males (macropterous, reproductive brachypterous and diapausing brachypterous) of the flightless bug Pyrrhocoris apterus were determined and related to the size of male accessory glands. The results revealed wing morph- and age-dependent differences in the corpus allatum volume in males of this bug. In 4-14 day old males, the volumes of the corpus allatum and accessory glands were largest in long-day reproductive brachypters, intermediate in long-day macropters, and smallest in short-day diapausing brachypters. The smaller corpus allatum in young macropterous males than in same aged reproductive brachypterous males was due to the spontaneous fasting of the former. Later, starting on day 18 after adult emergence, i.e. when macropterous males were feeding normally, there were no significant differences in the volumes of the corpus allatum between long-day brachypterous and macropterous males. On the other hand, the corpus allatum of 18-28 day old diapausing brachypterous males was significantly smaller than that of same aged long-day macropterous and reproductive brachypterous males. The sizes of the corpus allatum and accessory glands were significantly positively correlated in macropterous and diapausing brachypterous males. This is the first report of corpus allatum volume-dependent wing morph-related differences in the rate of accessory gland maturation in males of insects with a non-functional macropterism. The role of differential activity of the corpus allatum in the different life history strategies of males of the two wing morphs in this wing-polymorphic insect is discussed.
In the present study we tested whether the walking activity of macropterous females of the flightless wing-polymorphic bug Pyrrhocoris apterus (L.) can be stimulated by its native adipokinetic hormone Peram-CAH-II and the crustacean red pigment-concentrating hormone (Panbo-RPCH), and the effectiveness of the latter hormone in a lipid mobilization assay. Two different doses (10 or 40 pmol) of Peram-CAH-II or Panbo-RCPH were injected into 10-day-old macropterous females of P. apterus to evaluate their effects on the walking activity of treated females. The results obtained showed a significant stimulation of walking activity only with the lower dose (10 pmol) of either hormone Peram-CAH-II or Panbo-RPCH. On the contrary, the walking activity of the same-aged females of macropterous morph treated with the higher dose (40 pmol) of these hormones was decreased. The energy substrates mobilized in Panbo-RPCH-treated macropterous females were lipids. The question of whether the stimulation of locomotion by Panbo-RPCH is limited only to P. apterus or if it might also represent an important function of this hormone in other insects or even in crustaceans is discussed.
The effect of wing length (brachyptery and macroptery) on mating activity was investigated in adult males and females of a flightless wing-polymorphic insect, Pyrrhocoris apterus (L.). Mating activity of the brachypterous and macropterous adult bugs was assessed according to 14 different parameters. The competition tests revealed higher numbers of copulations and greater duration of mating activity in brachypterous than in macropterous males. Brachypterous males are between two to four times more successful in competition for females than their macropterous counterparts, depending on the wing morph and physiological status of the females. Decreased mating success of macropterous males is associated with the smaller size of their accessory glands. Lowered competitive ability for mates is a likely penalty associated with macroptery. Receptivity tests showed the highest tendency to mate in reproductive brachypterous females, lower in macropterous females and the least in diapausing brachypterous females. This is the first report of decreased mating propensity of macropterous morphs in insects with non-functional wing polymorphism. The association of lowered mating success with the higher dispersal activity of the macropterous morph in this bug and a trade-off between the ability to reproduce and to disperse in the flightless wing-polymorphic insects is discussed.
Three experimental groups of adult females of Pyrrhocoris apterus (reproductive brachypterous, diapausing brachypterous, and macropterous with non-diapause type of reproductive arrest) were analysed for their dose dependency of adipokinetic response, sensitivity of the fat body to Pya-AKH and locomotor activity during a 24 hour cycle. The greatest adipokinetic response (lipid elevation after hormonal treatment) was observed in macropterous, lower in diapausing brachypterous and the lowest in reproductive brachypterous females. Macropterous females also exhibited the highest activity of walking, while the lowest locomotor activity was found in diapausing brachypters. Diel changes of adipokinetic response were recorded in all three experimental groups, with maximal response occurring during the day at 6-7 h after the onset of light. On the contrary, the minimal adipokinetic response was found in the night and early after the onset of light. No significant differences in diel rhythms were observed between groups exposed to various day lengths. Diel changes in locomotor activity were found only in reproductive brachypterous females and females of macropterous morph, and showed a diurnal rhythm with only one peak of activity during a 24 hour cycle. In the latter, the rhythmic pattern of walking activity was positively correlated with diel changes of adipokinetic response. This indicates an important role of Pya-AKH in dispersal strategy of macropterous females of this heteropteran.
