Currently it remains difficult to obtain robust microsatellite markers for Lepidoptera. In an attempt to overcome the problems associated with developing microsatellite markers for this insect order we combined (i) biotin-enrichment protocol, (ii) next generation pyrosequencing (through 454 GS-FLX Titanium technology) and (iii) the use of individuals collected from eight geographically distant European populations representing three subspecies of Euphydryas aurinia. Out of 96 stringently designed primer pairs, 12 polymorphic microsatellite loci amplified without obvious evidence of null alleles in eight individuals from different subspecies. Between five and seven of these loci showed full within population applicability and three revealed to be robust and transferable between populations and sub-species, providing a first step towards the development of a valuable and robust tool for studying conservation issues and evolution in E. aurinia populations. Nevertheless, as in most studies dealing with Lepidoptera microsatellites, null alleles were detected in most of the developed markers. The Marsh fritillary (Euphydryas aurinia) (Lepidoptera: Nymphalidae) has declined across Europe, including the Czech Republic. Current conservation strategies rely on prevention of habitat loss and degradation, and increase in habitat quality and connectivity via promoting traditional grassland management. The population structure and adult demography parameters of a single population was investigated for eight years (single system), and of all the known Czech populations (multiple populations) for a single year, using mark-recapture. There was substantial variation in the patterns of adult demography, both among years in the single system and among the multiple populations in a single year. In the single system, the date of the first flight of an adult varied by 18 days over the 8 years and total annual numbers varied with a coefficient of variation of 0.40 (females fluctuating more than males). The average density was ca 80 adults/ha. The population size displayed density-dependence, i.e. decreased following years with high adult numbers, with an equilibrium density of 90 individuals/ha. The average density of the multiple populations was ca 120 individuals/ha. The estimated total population for the Czech Republic was 25,000 individuals (17,000 males / 8,000 females) in 2007, which does not indicate an imminent threat of extinction. We studied a population of the regionally endangered marsh fritillary butterfly Euphydryas aurinia inhabiting a system of loosely connected dry calcareous grasslands in sub-Mediterranean Slovenia. Our goal was to set the basis for a long-term monitoring of this butterfly in four meadows using mark-release-recapture (MRR). We determined its demographic parameters, dispersal, behaviour and utilization of nectar plants in different quality patches. Total population size was estimated to be approximately 347 males (95% confidence interval: 262-432) and 326 females (95% confidence interval: 250-402), with an unbiased sex ratio. The average lifespans were 6.3 and 8.6 days, respectively. Daily population sizes followed a parabola with marked protandry. Both sexes were relatively highly mobile with both occasionally moving over half a kilometre. The spatial distribution of animals seemed to be associated with patch size, host plant densities and nectar sources, resulting in much higher population densities in the largest patch. Adult behaviour differed between the sexes, with females resting more and flying less than males. Behaviour also changed during daytime and with the progression of the season. Adults were confirmed to be opportunistic feeders, since as many as ten nectar sources were detected. We conclude that demographic parameters differ greatly among regions and habitats, thus conservation aims should be planned accordingly. In Mediterranean habitats, the specialist butterfly Euphydryas aurinia oviposits on Lonicera implexa. Previous work has shown that ovipositing females select and lay a higher number of egg clusters on certain plants. In this paper the results of a field study aimed at assessing whether females use plant size and/or plant or leaf greenness (i.e., chlorophyll concentrations) as cues for oviposition are described. Size of plants did not appear to be an important factor in determining host plant selection, probably because even small plants provide enough resources for the young larvae to reach the diapausing stage and because last instar larvae, the most likely to face resource depletion, can move great distances in search of food. Measurements of both spectral reflectance and chlorophyll concentration of plants failed to reveal differences between host and non-host plants. On the other hand, reflectance and chlorophyll concentration of leaves were found to be important in oviposition choice as egg clusters were generally located on the greenest leaves with the highest chlorophyll contents. This suggests that females use visual cues to select the leaves that will provide optimal growth opportunities for newly hatched larvae. Although there was some indication that plants receiving a greater number of egg clusters also had more leaves of high chlorophyll content, multiple egg batches on single plants could also be a consequence of females being attracted by the presence of conspecific egg clusters.
In 1996, an old forest pasture grazed from the 1960s to 1988 was restored by coppicing, fencing and grazing by cattle to protect a local population of the endangered butterfly Euphydryas aurinia. An adjoining ungrazed meadow provided a control. In the first years, the butterfly became almost extinct due to the nearly complete consumption of the host plant of the larva, Succisa pratensis, by cattle. The butterfly population quickly recovered when the grazing pressure was lowered. Thus, the intensity of management should be adjusted by continuous monitoring of the target species. In the 2000s, the annual population was about 50 butterflies, but marked fluctuations took place, probably caused by natural factors. Grazing benefited the meadow flora and improved the habitat of butterflies in general. Extensive forest grazing clearly has the potential for enhancing biodiversity. The value of the experiment is, however, limited because only a single pair of meadows was available for comparison. In the future, it will be even more difficult to arrange a similar experiment due to the great decline in the numbers of traditional meadows and forest grazing in SE Finland.
