Acute respiratory distress syndrome (ARDS) is severe medicalcondition
occurring in critically ill patients and with mortality of 33-52 % is one of the leading causes of death in critically ill patients. To better understand pathophysiology of ARDS and to verify novel therapeutical approaches a reliable animal model is needed. Therefore we have developed modified
lavage model of ARDS in the pig. After premedication (ketamine and midazolam)35 healthy pigs were anesthetized (propofol, midazolam,
morphin, pipecuronium) and orotracheally intubated and ventilated. Primary
ARDS was induced by repeated cycles of lunglavage with a detergent Triton X100 diluted in saline (0.03 %) heated to 37 °C preceded by pre
-oxygenation with 100 % O2. Single cycle included two subsequent lavages
followed by detergent suction. Eachcyclewas followed by hemodynamic
andventilation stabilization for approx. 15 min, with eventualadministration of vasopressors according to an arterial bloodpressure. The lavage procedure
was repeated until the paO2/FiO2index after stabilization remained below 100 at PEEP 5 cm H2O. In 33 pigs we have achieved the desired degree
of severe ARDS(PaO2/FiO2<100). Typical number of lavages was 2-3 (min. 1,max.5). Hemodynamictolerance and the need for vasopressors
were strongly individual. In remainingtwo animalsan unmanageable hypotension developed. For other subjects theexperimental ARDS stability was good and allowed reliablemeasurement for more than 10 h. The
present model of theARDS is clinically relevant and thus it is suitable for further research of the pathophysiology and management of this serious
medical condition.
One of the most important problems faced in hydrology is the estimation of flood magnitudes and frequencies in ungauged basins. Hydrological regionalisation is used to transfer information from gauged watersheds to ungauged watersheds. However, to obtain reliable results, the watersheds involved must have a similar hydrological behaviour. In this study, two different clustering approaches are used and compared to identify the hydrologically homogeneous regions. Fuzzy C-Means algorithm (FCM), which is widely used for regionalisation studies, needs the calculation of cluster validity indices in order to determine the optimal number of clusters. Fuzzy Minimals algorithm (FM), which presents an advantage compared with others fuzzy clustering algorithms, does not need to know a priori the number of clusters, so cluster validity indices are not used. Regional homogeneity test based on L-moments approach is used to check homogeneity of regions identified by both cluster analysis approaches. The validation of the FM algorithm in deriving homogeneous regions for flood frequency analysis is illustrated through its application to data from the watersheds in Alto Genil (South Spain). According to the results, FM algorithm is recommended for identifying the hydrologically homogeneous regions for regional frequency analysis.
Ten ant species (Hymenoptera: Formicidae) from three subfamilies feed on honeydew excreted by nymphs of the leafhopper Balclutha punctata (Hemiptera: Cicadellidae). This relationship is facultative for the partners. Preliminary field observations suggest a mutualistic interaction that represents an intermediate stage in the spectrum of formicid-hemipteran mutualisms. Behavioural adaptations of the leafhopper (no escape reaction) and the ants (no predation of leafhoppers, protection from spiders) are evident, but characters of advanced facultative mutualism such as physical contact between the partners or ant-triggered excretion of honeydew were not observed.
Autologous vein grafts used as aortocoronary bypasses are often prone to intimal hyperplasia, which results in stenosis and occlusion of the vein. The aim of this study was to prevent intimal hyperplasia using a newly developed perivascular system with sustained release of sirolimus. This system of controlled drug release consists of a polyester mesh coated with a copolymer of L-lactic acid and ε -caprolactone that releases sirolimus. The mesh is intended for wrapping around the vein graft during surgery. The mesh releasing sirolimus was implanted in periadventitial position onto arteria carotis communis of rabbits, and neointimal hyperplasia was then assessed. We found that implanted sirolimus-releasing meshes reduced intima thickness by 47±10 % compared to a vein graft after 3 weeks. The pure polyester mesh decreased vein intima thickness by 35±9 %. Thus, our periadventitial system for controlled release of sirolimus prevented the develo pment of intimal hyperplasia in autologous vein grafts in vivo in rabbits. The parasitic caterpillars of Maculinea (Lepidoptera: Lycaenidae) need to be adopted and nursed by ants of the genus Myrmica (Hymenoptera: Formicidae). Each Maculinea species is locally associated with one or a few main and often several secondary host species. To determine whether the parasite-host associations bear marks of cophylogenetic constraints, we reconstructed phylogenies of Maculinea and Myrmica using DNA sequence data. We searched for evidence of cospeciation with a tree-independent (ParaFit) and tree-based (TreeFitter) method. This did not reveal any indication of phylogenetic host tracking in Maculinea. This agrees with earlier insights, which emphasise that as most of the potential host ant populations are never infested by Maculinea, the selective pressure of the butterflies on Myrmica is likely to be slight. Each Maculinea species also specialises on one or a few host plant species before adoption by ants. We suggest that Maculinea species have a substantial potential to accommodate evolutionarily to geographically changing ranges of potential Myrmica hosts, available at the oviposition sites of the butterflies. This note is concerned with a fragmentary gold covered disk brooch which comes from the hill-top settlement of Rubín u Podbořan, a site with evidence of occupation from the later prehistoric to post-Roman phases. Diabetic nephropathy (DN), the most serious complication of Type 1 diabetes (DM1), has a strong genetic component. Cyclooxygenase-2 (COX-2), an in ducible enzyme by a number of stimuli, has been implicated in pathophysiology of cardiovascular and renal disease, including DN. The allele -765C, of the -765G>C polymorphism (rs20417) in the COX-2 promoter has lower promoter activity compared with the -765G allele and protective effects in cardiovascular disease. This polymorphism was not investigated in patients with DM1 and nephropathy. The study was conducted in 779 Caucasian patients with DM1 and compared to a representative sample of healthy Czech population. The patients demonstr ated lower frequencies of the CC genotype (P=0.005). From th e DM1 cohort, 153 patients met the criteria for low risk of the development of DN (LRDN, duration of DM1>10 years, normoalbuminuria, normotension) and 139 patients had manifest DN. There were no differences in -765G>C polymorphisms between LRDN and DN patients. Moreover, the C/G allele frequenc ies did not also differ between the groups. In conclusion, patients with DM1 display lower freqencies of the protective CC genotype as compared to healthy subjects. The activity and number of protocerebral neurosecretory neurons of the dorsolateral group (L1, L2 and L2'), as well as the size of the corpora allata were investigated in 5th instar larvae of the gypsy moth (Lymantria dispar) from two populations (oak and locust-tree forests) fed one of two host-plants (oak is a suitable and locust-tree an unsuitable host-plant).
The monitoring of activity related cytological parameters and the number of protocerebral dorsolateral neurosecretory neurons revealed that differently adapted populations respond to nutritive stress differently. The activity of the L1 neurosecretory neurons in the protocerebra of the caterpillars is low in those from the locust-tree forest regardless of host-plant quality. The L2 neurosecretory neurons in the oak population become active when they were fed the unsuitable host-plant whereas their activity was high in locust-tree caterpillars regardless of the host-plant. A decrease in the number of neurosecretory neurons in a response to a novel food was noticed in both populations. The activity of the L2' neurosecretory neurons was similar in all caterpillars, but their number was increased in those from the locust-tree forest. The corpora allata of the locust-tree caterpillars were large whereas those of the oak forest caterpillars only increased in size when they were fed locust-tree leaves.
It is obvious that nutritive stress results in neurosecretory reorganization and changes in the titre of hormones that modulate the morphogenetic programme.
Chronic sojourn in hypoxic environment results in the structural remodeling of peripheral pulmonary arteries and pulmonary hypertension. We hypothesize that the pathogenesis of changes in pulmonary vascular structure is related to the increase of radical production induced by lung tissue hypoxia. Hypoxia primes alveolar macrophages to produce more hydrogen peroxide. Furthermore, the increased release of oxygen radicals by other hypoxic lung cells cannot be excluded. Several recent reports demonstrate the oxidant damage of lungs exposed to chronic hypoxia. The production of nitric oxide is high in animals with hypoxic pulmonary hypertension and the serum concentration of nitrotyrosine (radical product of nitric oxide and superoxide interaction) is also increased in chronically hypoxic rats. Antioxidants were shown to be effective in the prevention of hypoxia induced pulmonary hypertension. We suppose that the mechanism by which the radicals stimulate of the vascular remodeling is due to their effect on the metabolism of vascular wall matrix proteins. Non-enzymatic protein alterations and/or activation of collagenolytic matrix metalloproteinases may also participate. The presence of low-molecular weight cleavage products of matrix proteins stimulates the mesenchymal proliferation in the wall of distal pulmonary arteries. Thickened and less compliant peripheral pulmonary vasculature is then more resistant to the blood flow and the hypoxic pulmonary hypertension is developed., J. Herget, J. Wilhelm, J. Novotná, A.
