Theronts of the ciliate Ichthyophthirius multifiliis (Fouquet) with up to 4 micronuclei were recorded in populations from two different parasite isolates, maintained as primary infections in juvenile carp through 2 and 10 cycles, respectively. The largest number of multimicronucleate forms occurred within the older isolate (10 cycles) following cyst incubation at 20"C. Tro-phonts were induced to emerge from the host epidermis following incubation of the fish in Eagles MEM for 10-15 min at 20"C. This provided for the first time a technique to recover trophonts in all stages of development; observations were made on the transition from theront to trophont, with respect to nuclear events, the organelle of Lieberklihn, and phagocytosis of host cells. On re-exposure of carp already carrying a two-day primary infection, recently entered trophonts were found in various stages of fusion with established parasites. Results are discussed with respect to conjugation, anisogamy and senescence.
Our experiment was carried out in order to explore effects of plant growth regulators (PGR; thidiazuron, paclobutrazol, and ascorbic acid) on physiological traits of wheat genotypes under water surplus and deficit conditions. Study revealed that relative water content, membrane stability index, chlorophyll content, photosynthetic rate (PN), and maximal quantum yield of PSII improved with PGRs application across the genotypes both under irrigation and water stress. The response of HD 2733 genotype was more positive toward PGRs treatment as compared to other genotypes under water stress. Higher PN and chlorophyll contents were observed in HD 2987 followed by C 306 genotype under water-stress conditions. Moreover, Rubisco small subunit (SSU) expression was lower in wheat genotypes under water stress as compared to irrigated conditions. Application of PGRs led to upregulation of SSU under water stress, while no significant change was found in Rubisco level and activity under irrigated condition in dependence on PGRs treatments. Yield-related traits showed also significant reduction under water-stress conditions, while application of PGRs enhanced the yield and its components. Leaf senescence is always associated with decline in photosynthesis, consequently a loss of cellular sugar. On the other hand, execution of senescence program needs energy and leaves, therefore, tend to collect sugars from other sources to sustain energy homeostasis. This sugar reprogramming induced by loss of sugar involves operation of a complex catabolic network. The exact molecular mechanism of induction and regulation of the network, however, is not fully resolved but the current literature available suggests sugar starvation as a signal for induction of several senescence-associated genes including the genes coding for the enzymes for degradation of cellular constituents and their conversion to respiratory sugars. The late expression of genes coding for the cell wall hydrolases and enhancement in the activity of these enzymes late during senescence are indicative of the cell wall polysaccharides as the last source of sugars to sustain energy homeostasis for execution of the senescence program., B. Field experiments were conducted under rain-fed conditions to study the growth and photosynthetic efficiency of transgenic Bt cotton hybrids during 2002-03 and 2003-04 seasons. Three Bt cotton hybrids (Bollgard 1) and their non-Bt (NBt) counterparts viz. MECH 12, MECH 162, and MECH 184 were grown along with a local hybrid NHH44. Growth parameters such as plant height, main-stem nodes, biomass accumulation, and physiological processes like stomatal conductance (gs), and rates of transpiration (E) and photosynthesis (PN) did not differ significantly between Bt and NBt hybrids up to 80 DAS (d after sowing). Squaring commenced at 50 DAS both in Bt and NBt. The loss of young fruiting forms by the entomological factors was three times less in Bt than NBt. As a consequence, Bt had more early formed bolls on the lower canopy which contributed to higher biomass and seed cotton yield. On the other hand, bolls distributed intermittently in NBt. Heavy boll load altered the growth and physiological processes, and as a result Bt had higher gs, E, and PN than NBt. Since developing bolls (sink) divert the saccharides and nutrients from other organs, Bt plants with heavy boll load senesced early and stopped the production of new squares and bolls. The content of cytokinins (CKs), the plant inhibitors of the final phase of plant development, senescence, is effectively controlled by irreversible degradation catalysed by cytokinin oxidase/dehydrogenase (CKX). In transgenic tobacco, denoted as AtCKX, with over-expressed CKX causing lowered CK content, we investigated changes in the time courses of chlorophyll (Chl) and xanthophyll (violaxanthin, antheraxanthin, zeaxanthin, neoxanthin, and lutein) contents. We also determined parameters of slow Chl fluorescence kinetics such as minimum Chl fluorescence yield in the darkadapted state F0, maximum quantum yield of PS2 photochemistry (Fv/Fm), maximum ratio of quantum yields of photochemical and concurrent non-photochemical processes in photosystem 2 (PS2), Fv/F0, non-photochemical quenching (NPQ), and effective quantum yield of photochemical energy conversion in PS2 (Φ2). We used three different developmental leaf stages, old, mature, and young, and compared this with time courses of these characteristics in leaves with natural CK levels. The parameters Fv/Fm, Fv/F0, and Φ2 were unchanged during ageing in AtCKX plants in contrast to control ones where a significant decrease in old leaves was found. In control plants F0 increased during ageing, but in the oldest leaf a considerable decrease was observed. This could indicate progressive damage to PS2 reaction centres and then detachment and rapid degradation of Chl. This is in agreement with time course of Chl content. NPQ decreased with age and was similar in both plant types. We observed a decline of xanthophyll contents in the oldest leaves in both plant types, but the contents were enhanced in AtCKX compared to control plants, especially of neoxanthin. The higher xanthophyll contents in the transgenic plants contribute to a better photoprotection and the fluorescence parameters indicated that photosynthetic apparatus was in better condition compared to control and it consequently postponed the onset of leaf senescence. and Z. In the dimorphic stag’s hom fem, Platycerium coronarium (Koenig ex Mueller) Desv., photosynthetic characters and chlorophyll (Chl) contents were deteiinined in both nést and pendulous fronds at different stages of ontogeny and at different positions along the longest length of each frond. Area-specific radiant energy- saturated net photosynthetic (P^) and dark respiration rates, dry mass-specific Chl content and quantum yield increased during frond development and decreased when senescence set in. Radiant energy-saturated and dry-mass specific Chl content were greatest in the youngest tissues of each frond. In addition to the functions ascribed to the nést (water and nutrient collection) and pendulous (reproduction) fronds, the results indicate the important roles of both frond types in providing Chemical energy to the growth and survival of the plant.
The rate of accumulation of total chlorophyll (Chl) and carotenoids (Car) of leaves grown under high irradiance, HI (30 and 45 W m-2) was faster than at moderate irradiance, MI (15 W m-2). However, the senescence phase started earlier in the samples and proceeded at a faster rate. Chl a/b and Chl (a+b)/Car values showed faster loss of Chl a (compared to Chl b) and Chl (a+b) (compared to Car) in HI leaves. Protein accumulation and loss were also similar to that of Chl (a+b) content. Increase in Chl fluorescence during the development phase may suggest a gradual change in thylakoid organisation, however, the temporal kinetics were different in HI and MI samples. Increase in fluorescence polarisation during senescence of HI leaves compared to the control (MI) suggests conversion of thylakoid membranes to gel phase. Chloroplasts prepared from HI seedlings showed higher rate of photochemical activities, however, the activity declined earlier and at faster rate compared to the control. and Rajendra K. An early senescence (es) mutant of rice Oryza sativa L. with progressing death of most of leaves before heading stage was identified in the field in Hainan province. After tillering stage, the brown striations were found in the base of green leaves randomly, and then expanded to whole leaves. No fungi, bacteria, and viruses were detected in the brown striations suggesting that it was a genetic mutant. The ultrastructure of leaf cells at the site of brown striations showed breakdown of chloroplast thylakoid membrane structures and other organelles, and condensation of the cytoplasm at severe senescence stage. Changes in various components of photosynthetic apparatus during the 6-d dark incubation at 25 °C of detached control and DCMU-treated Triticum aestivum L. leaves were examined. The rate of photosystem 2 (PS2) activity was decreased with increase of the time of dark incubation in control leaves. In contrast to this, DCMU-treated leaves demonstrated high stability by slowing down the inactivation processes. Diphenyl carbazide and NH2OH restored the PS2 activity more in control leaves than in DCMU-treated leaves. Mn2+ failed to restore the PS2 activity in both control and DCMU-treated samples. Similar results were obtained when Fv/Fm was evaluated by chlorophyll fluorescence measurements. The marked loss of PS2 activity in dark incubated control leaves was primarily due to the loss of D1, 33, and 23 kDa extrinsic polypeptides and 28-25 kDa LHCP2 polypeptides. and N. Nedunchezhian, K. In individual leaves, the photon-saturated photosynthetic activity (Psat, expressed on a dry mass basis) was closely related to the nitrogen content (Nc) as follows: Psat = Cf Nc + Psat0, where Cf and Psat0 are constants. On a whole plant basis, the relative growth rate (RGR) was closely related to Nc in canopy leaf as follows: RGR = DMf Nc + RGR0, where DMf and RGR0 are constants. However, the coefficients Cf and DMf were markedly different among plant species. To explain these differences, it is suggested that carbon assimilation (or dry matter production) is controlled by both the Nc in a leaf (or leaves) and by the net N translocation from leaves. This is supported by the finding that Psat is related to the rate of 35S-methionine translocation from leaves. We propose another estimation method for the net N translocation rate (NFR) from leaves: Nc, after full leafing, is expressed as a function of time: Nc = (Nc0 - Ncd) exp(-Nft) + Ncd, where Nf is a coefficient, t is the number of days after leaf emergence, Nc0 is the initial value of Nc, and Ncd is the Nc of the dead leaf. The NFR is then calculated as NFR = ΔNc/Δt = -Nf (Nc - Ncd). Thus Nf is the coefficient for the NFR per unit Nc. NFR is a good indicator of net N translocation from leaves because NFR is closely related to the rate of 35S-methionine translocation from leaves. Since Psat is related to the 14C-photosynthate translocation rate, Cf (or DMf) corresponds to the coefficient of saccharide translocation rate per unit amount of Nc. Cf (or DMf) is closely related to the Nf of individual leaves (or the Nf of canopy leaf). This indicates that C assimilation and C translocation from leaves are related to Nc and N translocation from leaves (net translocation of N). Cf and Nf are negatively correlated with leaf longevity, which is important because a high or low CO2 assimilation rate in leaves is accompanied by a correspondingly high or low N translocation in leaf, and the degree of N translocation in leaves decreases or increases leaf longevity. Thus, since a relatively high Psat (or RGR) is accompanied by a rapid Nc decrease in leaves, it is difficult to maintain a high Psat (or RGR) for a sustained time period. and M. Osaki, T. Shinano.
