The decay of tyrosine cation radical was found to be biphasic at 253 K. The fast phase corresponds to the YZ* component while the slow phase corresponds to the tyrosine D radical (YD*) component. At 253 K, the t1/2 value was ∼28.6 s for the fast phase and ∼190.7 s for the slow phase. The fast phase is attributed to the recombination of charges between YZ* and QA-. The activation energy for the reaction of YZ with QA- between 253 and 293 K was 48 kJ mol-1 in Cl--depleted photosystem 2 (PS2) membranes. Both the decay rate and the amplitude of the PAR-induced signal of YZ* were affected by addition of chloride anion. Change in the decay rate and the amplitude of the PAR-induced signal of YZ* was observed when other anions like Br-, I-, F, HCO3-, NO3-, PO43- were substituted in the Cl--depleted PS2. and A. Elevated temperature inhibited the accumulation of chlorophyll and photosynthetic proteins, and the development of photochemical activity, however, carotenoids continued to accumulate. Signal transduction pathway involved in protochlorophyllide oxidoreductase was unaffected by elevated temperature of 38°C. Two-dimensional gel electrophoresis of stroma proteins showed similar patterns in the dark-grown seedlings and seedlings irradiated at elevated temperature, although some low molecular mass proteins accumulated at 38°C. In contrast, seedlings irradiated at 25°C showed complex pattern of proteins. Field studies of gas exchange and chlorophyll fluorescence of three desert shrub species, Hedysarum fruticosum var. mongolicum, Artemisia ordosia, and Salix pasmmophylla, showed different patterns under different leaf temperature (T1) and incident photosynthetic photon flux density (PPFD). H. fruticosum var. mongolicum and A. ordosia exhibited higher PN and gs than S. pasmmophylla, especially under very high T1 (>46 °C) and high PPFD (>2 100 µmol m-2 s-1) in hot summer. The decreases of PN with the diurnal course were due mainly to stomata closure. However, PN of S. pasmmophylla was seriously depressed by very high temperature from midday to evening as shown by the negative PN in hot summer, whereas none of such depression was found in spring. Maximal spring photochemical efficiency of photosystem 2 (PS2), i.e, Fv/Fm, was the lowest at 16:00, indicating the injury of PS2 by heat at this stage. In hot summer again, all the three shrubs underwent pronounced midday depression of PN and gs, while in spring they showed a one-peak response. And the first peak appeared 2 h earlier in hot summer than in spring for all the three shrubs. It was the high temperature that led to the different patterns of gas exchange and the serious depression of PN in S. pasmmophylla. H. fruticosum var. mongolicum and A. ordosia were much more tolerant to heat and high irradiance than S. pasmmophylla, which fixed most of CO2 at the fast growing stage in spring. The acclimation depression of capacity of photon utilisation in photochemical reactions of photosystem 2 (PS2) can develop already after three months of cultivation of the Norway spruces (Picea abies [L.] Karst.) under elevated concentrations of CO2 (i.e., ambient, AC, + 350 µmol(CO2) mol-1 = EC) in glass domes with adjustable windows. To examine the role that duration of EC plays in acclimation response, we determined pigment contents, rate of photosynthesis, and parameters of chlorophyll a fluorescence for sun and shade needles after three seasons of EC exposure. We found responses of shaded and exposed needles to EC. Whereas the shaded needles still profited from the EC and revealed stimulated electron transport, for the exposed needles the stimulation of both electron transport activity and irradiance saturated rate of CO2 assimilation (PNmax) under EC already disappeared. No signs of the PS2 impairment were observed as judged from high values of potential quantum yield of PS2 photochemistry (FV/FM) and uniform kinetics of QA reoxidation for all variants. Therefore, the long-term acclimation of the sun-exposed needles to EC is not necessarily accompanied with the damage to the PS2 reaction centres. The eco-physiological significance of the reported differentiation between the responses of shaded and sun exposed needles to prolonged EC may be in changed contribution of the upper and lower crown layers to the production activity of the tree. Whereas for the AC spruces, PNmax of shaded needles was only less than 25 % compared to exposed ones, for the EC spruces the PNmax of shaded needles reached nearly 40 % of that estimated for the exposed ones. Seasonal and daily variations in chlorophyll (Chl) fluorescence were studied in two representative species of Mediterranean grasslands, Tuberaria guttata (an annual) and Chamaemelum nobile (a perennial), in order to assess physiological responses to climatically induced stresses during the growing season. The photochemical efficiency of photosystem (PS) 2 in dark-adapted leaves was measured by the Chl fluorescence ratio Fv/Fp. This ratio decreased progressively from December to July, as the effects of increasing solar radiation and summer drought became more severe. The seasonal decline was observed particularly as a depression of morning and midday values, when photoinhibition was more evident. In both species, the extent of this diurnal depression increased with midday irradiance throughout winter and spring. After sunset, there was complete recovery to optimum values. Towards the end of the life cycle, increased irradiance did not affect the midday decline further but Fv/Fp measurements in the morning and evening never regained their optimum values, indicating the accumulation of photodamage in the reaction centres of PS2. The half-rise time of Fp (T1/2), used to estimate the size of the plastoquinone pool, showed little daily variation in C. nobile throughout the most important part of its seasonal cycle. However, towards the end of its life cycle (June and July) T1/2 values ranged from ca. 200 ms before sunrise to near zero at midday on the same day. The annual species, T. guttata, showed similar disregulation in energy transmission rate both at the seedling stage and at end of its life cycle. Thus seedlings and reproductive plants in particular are sensitive to environmental conditions (extremes of temperature and drought) and cannot maintain consistent electron flow throughout the day. and L. The photochemical activity of native Central Siberian Scots pine trees (Pinus sylvestris L) was estimated from the middle of February to the middle of March 2001. We measured chlorophyll (Chl) fluorescence in attached intact needles from trees located approx. 30 km west of the Yenisey river (60°44'N, 89°09'E) near the village of Zotino. In this period, the air temperature varied between -39 °C and +7 °C. At temperatures below -10 °C, P. sylvestris needles did not exhibit any variable Chl fluorescence during the daylight period. During the night, however, the effective quantum yield of photosystem 2 (PS2) photochemistry, Φ2 [Φ2 = (Fm' - Ft)/Fm'), increased from values near zero to values between 0.05 and 0.20 depending on the needle temperature and sample investigated. The increase started soon after dusk and lasted for 3-6 h depending on the temperature. A faster increase of Φ2 was found for temperatures around -16 °C, and lower rates occurred at lower temperatures. Irrespective of the temperature, Φ2 decreased rapidly to near zero values at dawn, when the photosynthetic photon flux density increased to about 1-5 µmol m-2 s-1, and remained near zero throughout the day. At temperatures higher than -10 °C, the diurnal decrease and the nocturnal increase of Φ2 were less distinct or disappeared completely. Hence the winter-adapted Scots pine maintains some photochemical activity of PS2 even at extremely cold temperatures. The capacity of photochemical reactions below -10 °C is, however, very limited and PS2 photochemistry is saturated by an extremely low irradiance (less than 5 µmol m-2 s-1). and P. Šiffel, J. Membrane-bound bicarbonate is believed by some to act as an essential activator of photosystem 2 (PS2) electron transport. Formáte and other inhibitory monovalent anions act by removing bound-bicarbonate. This belief relies to a great extent on the observation that formáte (100 mM) pretreated thylakoids exhibit a non- proportionality between Hill activity (HAR) and chlorophyll (Chl) concentration when preirradiated with bright radiation in reaction mixture that contains only 5 mM formáte. The non-linearity was attributed to a supposed loosening of residual bicarbonate still present after formáte treatment and which would be more abundant at higher Chl concentrations. In repeating this experiment, we observed an increase in HAR at higher Chl concentrations in preirradiated, but also in non-preirradiated samples, the latter were simply left in the dark for 3 min before measurements were made. Therefore, preirradiation is not needed to restore some HAR in formáte pretreated samples; a 3 min wait in the electrode chamber at low formáte concentration is sufficient to partially relieve the formáte inhibition of PS2 activity. Moreover, HAR in samples preirradiated by weak radiation, or not preirradiated at all, was directly proportional to Chl concentration. We can attribute the increase in activity to a dissociation of bound formáte, not necessarily to the effect of residual bicarbonate. Non-linearity in HAR with Chl concentration was found only in high- irradiance pretreated samples. We can attribute this to a greater amount of photoinhibition occurring in the dilute samples, where the effective irradiance was greater. There is no need to postuláte the existence of residual bound bicarbonate to explain these results.
We investigated the strategies of four co-occurring evergreen woody species Quercus ilex, Quercus coccifera, Pinus halepensis, and Juniperus phoenicea to cope with Mediterranean field conditions. For that purpose, stem water potential, gas exchange, chlorophyll (Chl) fluorescence, and Chl and carotenoid (Car) contents were examined. We recognized two stress periods along the year, winter with low precipitation and low temperatures that led to chronic photoinhibition, and summer, when drought coincided with high radiation, leading to an increase of dynamic photoinhibition and a decrease of pigment content. Summer photoprotection was related to non-photochemical energy dissipation, electron flow to alternative sinks other than photosynthesis, decrease of Chl content, and proportional increase of Car content. Water potential of trees with deep vertical roots (Q. coccifera, Q. ilex, and P. halepensis) mainly depended on precipitation, whereas water potential of trees with shallow roots (J. phoenicea) depended not only on precipitation but also on ambient temperature. and F. Maize (Zea mays) seedlings were exposed for 6 h to strong irradiance (1 000 μmol m-1 s-1 of PPFD) at 5, 12, 17, or 25 °C, followed by an exposure to the darkness for 6 h at 22 °C. Leaf chlorophyll fluorescence, net photosynthetic rate (PN), and the amount of superoxide radicals (O2-⋅) in relation to chilling-induced photoinhibition were investigated. During the photophase, a good correlation (r=-0.879) was observed between ΦPS2 (relative quantum efficiency of PS2 electron transport) and the amount of O2-⋅. Treatment with exogenous O2-⋅ reduced the PN and ΦPS2 as the chilling stress did, that was inhibited by specific scavenger of O2-⋅. Hence chilling-induced photoinhibition might be due to the production of O2-⋅. In contrast, in the dark period, PN and ΦPS2 of the seedlings treated with the exogenous O2-⋅ were enhanced, but they were inhibited by the specific scavenger of O2-⋅, showing the photoprotective role of O2-⋅ in the recovery phase. Furthermore, in terms of the effect of exogenous O2-⋅ on the xanthophyll cycle, the O2-⋅ production suggested a promotion effect for the de-epoxidation of violaxanthin during the photophase, the epoxidation of zeaxanthin at the dark stage, and the increase of the xanthophyll pool both in the photophase and dark phase, resulting in an enhancement of the ability of non-photochemical quenching to avoid or alleviate the damage to photosynthetic apparatus. and D. Three-month-old mulberry (Morus alba L.) cultivars (drought tolerant S13 and drought sensitive S54) were subjected to water stress for 15 d. Water stress decreased the leaf water potential, net photosynthetic rate (PN), and stomatal conductance (gs) in both the cultivars. However, the magnitude of decline was comparatively greater in the sensitive cultivar (S54). Intercellular CO2 concentration (Ci) was unaltered during mild stress, but significantly increased at severe stress in both cultivars. The photosystem 2 activity significantly declined only at a severe stress in both cultivars. The Ci/gs ratio representing the mesophyll efficiency was greater in the tolerant cultivar S13. Involvement of stomatal and/or non-stomatal components in declining PN depended on the severity and duration of stress.
