The chromosome complements of thirteen species of the planthopper family Dictyopharidae are described and illustrated. For each species, the structure of testes and, on occasion, ovaries is additionally outlined in terms of the number of seminal follicles and ovarioles. The data presented cover the tribes Nersiini, Scoloptini and Dictyopharini of the subfamily Dictyopharinae and the tribes Ranissini, Almanini, and Orgeriini of the Orgeriinae. The data on the tribes Nersiini and Orgeriini are provided for the first time. Males of Hyalodictyon taurinum and Trimedia cf. viridata (Nersiini) have 2n = 26 + X; Scolops viridis, S. sulcipes, and S. abnormis (Scoloptini) 2n = 36 + X; Callodictya krueperi (Dictyopharini) 2n = 26 + X; Ranissus edirneus and Schizorgerius scytha (Ranissini) 2n = 26 + X. Males of Almana longipes and Bursinia cf. genei (Almanini) have 2n = 26 + X and 2n = 24 + XY, respectively. The latter chromosome complement was not recorded previously for the tribe Almanini. Males of Orgerius ventosus and Deserta cf. bipunctata (Orgeriini) have 2n = 26 + X. The testes of males of A. longipes and B. cf. genei each have 4 seminal follicles, which is characteristic of the tribe Almanini. Males of all other species have 6 follicles per testis. When the ovaries of a species were also studied, the number of ovarioles was coincident with that of seminal follicles. For comparison, Capocles podlipaevi (2n = 24 + X and 6 follicles per testis in males) from the Fulgoridae, the sister family to Dictyopharidae, was also studied. We supplemented all the data obtained with our earlier observations on Dictyopharidae. The chromosomal complement of 2n = 28 + X or that of 2n = 26 + X and 6 follicles per testis are suggested to be the ancestral condition among Dictyopharidae, from which taxa with various chromosome numbers and testes each with 4 follicles have differentiated.
Physico-chemical properties and carbohydrate-binding specificity of hemagglutination activity (HA) were compared in tissue lysates and haemolymph of unfed and bloodied females of five sandfly species. Sandfly gut lectins were found to be heat-labile, sensitive to dithiotreitol treatment, freezing/thawing procedures and were affected by divalent cations. The pH optimum of HA ranged between 7.0-7.5. Specificity of gut HA of all species studied was directed towards aminosugars and some glycoconjugates, mainly lipopolysaccharide from Escherichia coli K-235, heparin and fetuin. Gut HA of Phlebotomus papatasi (Scopoli, 1786) was strongly inhibited by lipophosphoglycan (LPG) from Leishmania major promastigotes. In females, that took blood, the HA was higher but the carbohydrate-binding specificity remained the same; this suggests that the same lectin molecule was present, at different levels, both in unfed and fed flies. High HA was found in ovaries of fed females of Lutzomyia longipalpis (Lutz et Nieva, 1912), P. papatasi and P. duhoscqi Neveu-Lemaire, 1906. In P. papatasi and P. duboscqi the HA was present also in the haemolymph and head lysates of both fed and unfed females. Carbohydrate-binding specificity of HA present in these tissues was similar with the gut lectin.
Episyrphus balteatus only matures eggs after emergence. Ovaries develop in 4 stages. In the absence of oviposition sites, females refrain from ovipositing and their ovaries progressively fill the abdomen and then egg resorption occurs. The potential fecundity, which is expressed by the ovariole number, the reproductive biomass and the abdomen volume, scales isometrically with the size of females. Egg size is much less variable and does not rise proportionally to body size. In laboratory conditions, females of E. balteatus might lay between 2,000 and 4,500 eggs during their life-time at a rate of 1 to 2 eggs per ovariole per day. Both life-time fecundity and rate of egg production are directly related to the size of females. The potential and realized fecundities are likely to be limited by the availability of food resources during larval and adult life, respectively.
Aquaporins (AQPs) are water channel proteins responsible for water homeostasis and important for proper functioning of all body systems, including reproductive structures. This study was designed to determine their localization and quantitative changes in the pig ovary during different stages of the estrous cycle and early pregnancy. The expression of AQP 1, 5 and 9 proteins was determined by immunocytochemistry and Western blot analyses. AQP1 was found in the plasma membranes of capillary endothelium, AQP5 - in the plasma membranes of granulosa cells of developing follicles and flattened follicle cells of the primordial follicles, and AQP9 - in granulosa cells of the developing follicles. In the cyclic pigs, the expression of AQP1 and 5 proteins was the highest on Days 18-20, but did not change significantly between Days 2-4, 10-12 and 14-16 of the cycle. In pregnant pigs (Days 14-16 and 30-32), the expression of AQP1 and 5 did not change and was similar to that observed during Days 10-12 and 14-16. In turn, AQP9 expression did not change between all studied periods. In conclusion, studied AQP are localized in different cells populations, the endothelial and granulosa cells, and AQP1 and 5 seem to be crucial for follicular development in pigs., A. Skowronska, P. The organization and development of ovaries in representatives of two families (Putoidae and Monophlebidae) of scale insects are described. Developing ovaries of Puto albicans McKenzie, 1967 and Crypticerya morrilli (Cockerell, 1914) consist of numerous clusters of cystocytes that are arranged in the form of rosettes. At the end of the last nymphal instar these clusters start to protrude from the interior of the ovary into the body cavity and the ovarioles begin to be formed. The ovary of a young female is composed of about 200 spherical telotrophic ovarioles devoid of terminal filaments. The ovarioles of C. morrilli contain 8 germ cells (7 trophocytes and a single oocyte). From 25 to 45 germ cells (23-43 trophocytes and 2 or 3 oocytes) occur in the ovarioles of P. albicans. An ovariole of an adult female is subdivided into a trophic chamber (tropharium), vitellarium and ovariolar stalk (pedicel). At each stage of development, the ovaries are accompanied by large cells (termed bacteriocytes) that contain endosymbiotic microorganisms. The organization of the ovary in P. albicans is more similar to that in archaeococcoid scale insects than in neococcoid taxa. In contrast, the number of germ cells per ovariole in C. morrilli is not typical of other archaeococcoids, but resembles the derived condition seen in other iceryine taxa. The classification and phylogeny of scale insects are discussed in the light of these results.
The aim of our in vitro studies was to understand the role of leptin in controlling proliferation, apoptosis, and protein kinase A (PKA) in human ovarian cells. We analyzed the in vitro effects of leptin (0, 1, 10 or 100 ng/ml) on the accumulation of proliferation-related peptides (PCNA, cyclin B1), apoptosis-associated peptide (Bax) and the intracellular signaling molecule PKA in cultured human granulosa cells using immunocytochemistry and Western immunoblotting. It was observed that leptin stimulated in a dose-dependent manner the accumulation of PCNA (at doses 1-100 ng/ml), cyclin B1 (at doses 10 or 100 ng/ml), Bax (at doses 10 or 100 ng/ml) and PKA (at doses 1-100 ng/ml) in cultured human ovarian cells. The aim of the present study was to examine the role of nutritional status, the metabolic hormone ghrelin and their interrelationships in the control of chicken hormones involved in the regulation of reproduction. For this purpose, we identified the effect of food deprivation, administration of ghrelin 1-18 and their combination on plasma levels of testosterone (T), estradiol (E), arginine-vasotocin (AVT) and growth hormone (GH) as well as the release of these hormo nes by isolated and cultured ovarian fragments. It was observed that food deprivation reduces plasma T and E and increases plasma AVT and GH levels. Food restriction also reduced the amount of E produced by isolated ovaries, but it did not affect the ovarian secretion of T and AVT. No ovarian GH secretion was detected. Ghrelin administered to ad libitum fed chickens did not affect plasma T and E levels, but it did increase plasma GH and AVT concentrations. Moreover, it partially prevented the effect of food deprivation on plasma E and AVT levels, but not on T or GH levels. Ghrelin administration to control birds promoted ovarian T, but not E or AVT release and reduced T and no other hormonal outputs in birds subjected to food restriction. Our results (1) confirmed the ovarian origin of the main plasma T and E and the extra-ovarian origin of the main blood AVT and GH; (2) showed that food deprivation-induced suppression of reproduction may be caused by suppression of T and E and the promotion of AVT and GH re lease; (3) suggest the involvement of ghrelin in control chicken E, AVT and GH output; and (4) indicates that ghrelin can either mimic or modify the effect of the intake of low calories on chicken plasma and ovarian hormones, i.e. it can mediate the effect of metabolic state on hormones involved in the control of reproduction., A. Protein kinases, transcription factors and other apoptosis- and proliferation-related proteins can regulate reproduction, but their involvement in sexual maturation remains to be elucidated. The general aim of the in vivo and in vitro experiments with porcine ovarian granulosa cells was to identify possible intracellular regulators of female sexual maturation. For this purpose, proliferation (expression of proliferating cell nuclear antigen - PCNA, mitogen-activated protein kinases - ERK 1,2 related MAPK and cyclin B1), apoptosis (expression of the apoptotic protein Bax and apoptosis regulator Bcl-2 protein), expression of some protein kinases (cAMP dependent protein kinase - PKA, cGMPdependent protein kinase - PKG, tyrosine kinase - TK) and cAMP responsive element binding protein 1 (CREB-1) was examined in granulosa cells isolated from ovaries of immature and mature gilts. Expression of PCNA, ERK1,2 related MAPK, cyclin B1, Bcl-2, Bax, PKA, CREB-1, TK and PKG in porcine granulosa cells were detected by immunocytochemistry. Sexual maturation was associated with significant increase in the expression of Bcl-2, Bax, PKA, CREB-1 and TK and with decrease in the expression of ERK1,2 related MAPK, cyclin B1 and PKG in granulosa cells. No significant difference in PCNA expression was noted. The present data obtained from in vitro study indicate that sexual maturation in females is influenced by puberty-related changes in porcine ovarian signaling substances: increase in Bcl-2, Bax, PKA, CREB-1, TK and decrease in ERK1,2 related MAPK, cyclin B1 and PKG. T-2 toxin and its metabolite HT-2 toxin are one of the most toxic mycotoxins of type A-trichothecenes, which are produced mainly by Fusarium species. Therefore, study of Fusarium toxins T-2 toxin and HT-2 toxin is an essential issue because they could also play role in failures of reproductive functions as well as endocrine system of domestic animals. Assessment of the effect of A-trichothecene mycotoxin HT-2 toxin alone or combined with insulin-like growth factor (IGF-I), leptin and ghrelin on estradiol secretion by rabbit ovarian fragments in vitro was done. Rabbit ovarian fragments were incubated without (control group) or with HT-2 toxin, or its combinations with IGF-I, leptin and ghrelin at various concentrations for 24 h. Secretion of 17β-estradiol was determined by ELISA. Firstly, HT-2 toxin at the doses 10 and 100 ng.ml-1, but not at 1 ng.ml-1 decreased 17β-estradiol secretion by ovarian fragments. Secondly, 17β-estradiol secretion was not affected by HT-2 toxin exposure combined with growth factor IGF-I, metabolic hormones leptin and ghrelin. In conclusion, HT-2 toxin has potent direct dose-dependent effects on ovarian steroidogenesis in rabbits. Numerous hypotheses have been proposed about the pathogenesis of the polycystic ovarian syndrome (PCOS). However, hormonal control of persistent follicles has not be enestablished. The objective of the present study was to compare the follicular structure and hormonal profiles of rats treated with the adrenocor ticotrophic hormone (ACTH) with two experimental models of PCOS. ACTH-treated animals were compared with those exposed to continuous light, those treated with estradiol valerate, and with control (in proestrous and diestrous). Serum hormone levels, histomorphometrical changes, and immunoexpression of vimentin, cytokeratins, cadherins, and proliferating cell nuclear antigen (PCNA) were examined. Treatment with ACTH resulted in an elevation of corticosterone secretion with LH reduction but without changes in ovarian morphology. Although stress (or ACTH) stimulation may be only one of pathophysiological mechanisms involved in follicular cystathogenesis in other species, we do not have important evidence to suppose that this would happen in rats., C.
