This investigation addresses the interaction of insulin (INS) and glucocorticoid (GC) signaling in the hepatic regulation of tryptophan oxygenase (TO) enzyme activity in the rat. Male Wistar rats (200-250 g b.w) received an injection of the different doses of INS (10, 25, 50, 70 and 100 μg/200 g b.w., i.p.) and were used for experiments 3 h and 18 h after INS administration. This study shows that maximum of TO activity was found at dose of 50 μg of INS with peak increases observed at 3 h and 18 h after injection of INS, while INS had no effect on TO activity in adrenalectomized rats. The analysis of INS effects on glucocorticoid receptor-complex (GC/GR complex) stability shows that complexes from INS-treated rats are less stable than those from control animals. In addition, INS-stimulated stability of glucocorticoid receptor (GR) protein was significantly increased from the controls. Furthermore, the results show that GC/GR complexes from INS-treated rats could be activated and accumulated at higher rate in cell nuclei of control animals. Energy transfer of aromatic amino acids in photosystem 2 (PS2) core antenna complexes CP43 and CP47 was studied using absorption spectroscopy, fluorescence spectroscopy, and the 0.35 nm crystal structure of PS2 core complex. The energy of tyrosines (Tyrs) was not effectively transferred to tryptophans (Trps) in CP43 and CP47. The fluorescence emission spectrum of CP43 and CP47 by excitation at 280 nm should be a superposition of the Tyr and Trp fluorescence emission spectra. The aromatic amino acids in CP43 and CP47 could transfer their energy to chlorophyll (Chl) a molecules by the Dexter mechanism and the Föster mechanism, and the energy transfer efficiency in CP47 was much higher than that in CP43. In CP47 the Föster mechanism must be the dominant energy transfer mechanism between aromatic amino acids and Chl a molecules, whereas in CP43 the Dexter mechanism must be the dominant one. Terbium (Tb^'*") was ušed as a fluorescence probe in the study of calcium-binding sites on 33 kDa protein of photosystem 2. The fluorescence of Tb^^ was enhanced markedly when bound to the 33 kDa protein, and the non-radiative energy transfer between tryptophan (Trp) residue and Tb^+, bound to the calcium-binding sites on the 33 kDa protein, took plače. According to the Forster non-radiative energy transfer mechanism, the average distance between the bound Tb3+ and Trp residue was found to be 1.05 nm. The pH titration indicated that major groups in the 33 kDa protein, involved in Ca2+ ions binding, were the carboxylic side groups of the glutamic acid and/or aspartic acid.
The aim of the article is to examine side effects of increased dietary intake of amino acids, which are commonly used as a dietary supplement. In addition to toxicity, mutagenicity and carcinogenicity, attention is focused on renal and gastrointestinal tract functions, ammonia production, and consequences of a competition with other amino acids for a carrier at the cell membranes and enzymes responsible for their degradation. In alphabetic order are examined arginine, β-alanine, branchedchain amino acids, carnosine, citrulline, creatine, glutamine, histidine, β-hydroxy-β-methylbutyrate, leucine, and tryptophan. In the article is shown that enhanced intake of most amino acid supplements may not be risk-free and can cause a number of detrimental side effects. Further research is necessary to elucidate effects of high doses and long-term consumption of amino acid supplements on immune system, brain function, muscle protein balance, synthesis of toxic metabolites, and tumor growth and examine their suitability under certain circumstances. These include elderly, childhood, pregnancy, nursing a baby, and medical condition, such as diabetes and liver disease. Studies are also needed to examine adaptive response to a long-term intake of any substance and consequences of discontinuation of supplementation.
