This paper describes the first study of the diatom assemblages in caves in the Czech Republic. The study focused on subaeric habitats: rock faces within caves, walls at cave entrances and “lampflora” assemblages, in three cave systems, Mladeč, Javoříčko and Zbrašov, all in central Moravia. The morphological and cytological variability, ecology and life strategies of diatoms were studied in fresh samples, in Naphrax preparations, in cultures grown on agar plates and in monoclonal cultures. A total of 22 diatom species was identified, mostly aerophytic species and tolerant of low light intensities. Luticola and Diadesmis species complexes are discussed. Taxa D. gallica and L. paramutica var. binodis are new for the Czech Republic. Sexual reproduction was observed in L. mutica and Orthoseira roeseana.
Monosodium glutamate (MSG), the sodium salt of glutamate, is commonly used as a flavor enhancer in modern nutrition. Recent studies have shown th e existence of glutamate receptors on lymphocytes, thymoc ytes and thymic stromal cells. In this study, we evaluated the in vitro effect of different MSG concentr ations on rat thymocyte apoptosis and expression of two apoptosis-related proteins, Bcl-2 and Bax. Rat thymocytes, obtained from male Wistar rats, were exposed to increasing concentrations of MSG (ranging from 1 mM to 100 mM) for 24 h. Apoptosis was detected using the Annexin V-FITC/PI apoptosis detection kit and cells were analyzed using a flow cytometer. Expression of Bcl-2 and Bax proteins were determined with flow cytometry using respective monoclonal antibodies. Exposure to MSG resulted in a dose-dependent decrease in cell survival (as determined by trypan blue exclusion method). Annexin V- FITC/PI also confirmed that MSG incr eased, in a dose-dependent manner, ap optotic cell death in rat thymocyte cultures. MSG treatment induced downregulation of Bcl-2 protein, while Bax protein levels were not significantly changed. Our data showed that MSG significantly modulates thymocyte apoptosis rate in cultures. The temporal profile of Bcl-2 and Bax expression after MSG treatment suggests that downregulation of Bcl-2 protein and the resulting change of Bcl-2/Bax protein ratio may be an important event in thymocyte apoptosis triggered by MSG., V. The ultrastructural cytology and reproduction of Amphiacantha longa Caullery et Mesnil, 1914 is described. Mcrogonial reproduction was not observed. The sporogony follows two lines: free disporoblastic. and enveloped, polysporoblastic, involving sporoblast mother cells. The enveloped sporogony is endogenous in spore sacs of sporont origin, daughter cells are formed by vacuolation. Probably all stages have coupled nuclei. Both free and enveloped spores are equipped with an extrusion apparatus composed of a flat polar sac, a straight polar filament of manubrium type, and a posterior globular appendix. Manubrium and appendix are enclosed in a membraneous coat. Circular elements of coat material occur in the proximity of the extrusion apparatus. The membraneous coat and the surface layer of the manubrium penetrate the polar sac. The extrusion apparatus is located at the wide pole of the spore, the nuclei at the narrower pole. Hosts are gregarines of the genus Lecudina Mingazzini, living in the gut of the polychaete Lumbrinereis fragilis (O. F. Muller). The cytology and reproduction are discussed and compared to other genera of metchnikovellideans, to the chytridiopsid genera, and to microsporidia expressing the typical cytology for the group. Metchnikovellideans and chytridiopsids exhibit cytological and reproductive similarities. The species is redescribed, the diagnosis of the genus Amphiacantha Caullery et Mesnil, 1914 is emended, and the new family Amphiacanthidae, comprising the genera Amphiacantha and Amphiamblys Caullery et Mesnil, 1914, is established.
