Ischemic preconditioning (IP) protects the heart against subsequent prolonged ischemia. Whether the β-adrenoceptor/adenylate cyclase pathway contributes to this cardioprotection is not yet fully known. Using enzyme catalytic cytochemistry we studied the adenylate cyclase activity and its distribution in the preconditioned rat heart. Adenylate cyclase activity was examined in Langendorff-perfused rat hearts subjected to the following conditions: control perfusion; 30 min regional ischemia; 5 min occlusion and 10 min reperfusion (IP); IP followed by ischemia. Ischemia-induced arrhythmias and the effect of ischemic preconditioning on the incidence of arrhythmias were analyzed. At the end of experiment the heart was shortly prefixed with glutaraldehyde. Tissue samples from the left ventricle were incubated in a medium containing the specific substate AMP-PNP for adenylate cyclase and then routinely processed for electron microscopy. Adenylate cyclase activity was cytochemically demonstrated in the sarcolemma and the junctional sarcoplasmic reliculum (JSR) in control hearts, while it was absent after test ischemia. The highest activity of the precipitate was observed after ischemic preconditioning. In the preconditioned hearts followed by test ischemia, adenylate cyclase activity in the precipitate was preserved in sarcolemma and even more in JSR. Protective effect of ischemic preconditioning was manifested by the suppression of severe arrhythmias. RNA optical density (concentration) measurements at the single cell level indicated that differentiation of lymphocytes is accompanied by a slightly decreased nucleolar RNA concentration in contrast to the cytoplasmic rim around the nucleus. On the other hand, the nucleolar size was markedly reduced and the cytoplasmic rim surrounding the nucleus was reduced only weakly. Concerning the calculated rough estimate of the RNA content, the differentiation induced its larger decrease in the nucleoli than in the cytoplasmic rim. These observations indicated that the nucleolar RNA concentration and RNA content together with the nucleolar morphology are more sensitive markers of the differentiation process than the RNA concentration and content in the cytoplasm. Thus, the nucleolar RNA transfer to the cytoplasm in advanced differentiation steps might still be going on regardless of the decreasing or inhibited nucleolar biosynthetic activity. In addition, the presence of ring-shaped nucleoli and micronucleoli characteristic of mature and terminal lymphocytes in some lymphocytic less differentiated steps, i.e., lymphoblasts and prolymphocytes, might indicate the premature differentiation state of such cells.
In the present paper, we describe the ultrastructure of the spermatozoon of the notocotylid Notocotylus noyeri (Joyeux, 1922) by means of transmission electron microscopy. The mature spermatozoon of N. noyeri exhibits the general pattern described in the majority of digeneans: two axonemes of the 9 + "1" pattern of the Trepaxonemata, nucleus, mitochondria, parallel cortical microtubules, spine-like bodies and ornamentation of the plasma membrane. The glycogenic nature of the electron-dense granules was evidenced applying the test of Thiéry. The ultrastructural features of the spermatozoon of N. noyeri present some differences in relation to those of the Pronocephalidea described until now, but confirm a general pattern for the Notocotylidae, namely a spermatozoon with two mitochondria and an anterior region with ornamentation of the plasma membrane associated with spine-like bodies. The posterior extremity of the spermatozoon exhibits only some microtubules after the disorganisation of the second axoneme. The present study confirms that some ultrastructural characters of the sperm cell such as the presence or absence of lateral expansions, the number of mitochondria and the morphology of both anterior and posterior spermatozoon extremities are useful for phylogenetic purposes within the Pronocephaloidea. Thus, unlike notocotylids, pronocephalids exhibit external ornamentation and a lateral expansion in the anterior spermatozoon region. Moreover, notocotylid spermatozoa present two mitochondria, whereas pronocephalid spermatozoa exhibit a single mitochondrion. Finally, pronocephalids are characterised by a type 2 posterior spermatozoon extremity, whereas notocotylids exhibit a type 3 posterior spermatozoon extremity., Papa Ibnou Ndiaye, Jordi Torres, Catarina Eira, Vladimir V.
