2. Algesthesia after epileptic seizure

- The consequences of epileptic seizures related to postictal inhibition in early postictal period include postictal analgesia. We studied this phenomenon over 96 h following flurothyl-induced seizures in adult male Wistar rats. Nociception of control (no seizure) and seizured groups were tested using the plantar and von Frey hair tests. We determined latency of forepaw and hind paw reactions using plantar tests and the number of von Frey hairs reactions. Shortly after seizures, longer plantar test latencies were seen relative to the control group. Before the seizures the plantar test reaction times were significantly shorter in forepaws than in hind paws. The effect disappeared post-seizure and surprisingly, it also disappeared at the corresponding time in controls; it reappeared after 48 h in the seizure group and after 24 h in controls. Differences in the von Frey hairs test occurred at 5 and 60 min post-seizure, however, these differences could not be explained by limb anatomy; although, different thermal and mechanical nociception mechanisms could be significant. - Damage of molecules as a consequence of oxidative stress has been implicated in the pathogenes is of chronic diseases related to aging. Diet is a key environmental factor affecting the incidence of many chronic diseases. Antioxidant substances in diet enhance the DNA, lipid and protein protection by increasing the scavenging of free radicals. Products of oxidative damage of DNA (DNA strand breaks with oxidized purines or oxidized pyrimidines), lipids (conjugated dienes of fatty acids) and proteins (carbonyls) in relation to nutrition (vegetarian diet vs. non-vegetarian, traditional mixed diet) were measured in young women aged 20-30 years (46 vegetarians, 48 non-vegetarians) vs. older women aged 60-70 years (33 vegetarians, 34 non-vegetarians). In young subjects, no differences in values of oxidative damage as well as plasma values of antioxidative vitamins (C, β-carotene) were observed between vegetarian and non-vegetarian groups. In older vegetarian group significantly reduced values of DNA breaks with oxidized purines, DNA breaks with oxidized pyrimidines and lipid peroxidation and on the other hand, significantly increased plasma values of vitamin C and β-carotene were found compared to the respective non-vegetarian group. Significant age dependences of measured parameters (increase in all oxidative damage products and decrease in plasma vitamin concentrations in older women) were noted only in non-vegetarians. Vegetarian values of older women vs. young women were similar or non-significantly changed. 4. Evidence of the use of a horn yoke in the Middle La Tene period, and an analysis of animal finds from La Tene features in the Velké Zboží and Malé Zboží cadasters, central Bohemia

- During archaeological excavations of three La Tène features in 2001 and 2002 at the periphery of the Velké Zboží and Malé Zboží cadasters (Nymburk district), a total of 33 animal bones were obtained. Only domestic mammals were determined in the material: cattle, sheep/goat (of which only sheep is reliably documented), pig, horse and dog. A valuable find from feat. 8 (LT C2-D1) is a horn core from a domestic cattle (Bos taurus), which shows signs of pathology. The pathology comes in the form of a depression on the anterior surface, which leads to the narrowing of the horn. Such pathology is considered to be the result of long-term use of a forehead yoke. The find represents one of a few documented cases of the use of the yoke in prehistory in the territory of today’s Czech Republic. A hornless sheep is documented at another feature. and Při archeologickém výzkumu tří laténských objektů v letech 2001 a 2002 na hranici katastrů Velkého Zboží a Malého Zboží bylo získáno celkem 33 zvířecích kostí. V materiálu byly determinovány pouze domácí druhy savců: tur, ovce/koza (z nich spolehlivě doložena pouze ovce), prase, kůň a pes. Cenným nálezem z obj. 8 (LT C2-D1) je rohový výběžek tura domácího (Bos taurus), který vykazuje patologii. Patologie má charakter deprese na anteriorní ploše, což vede k zúžení rohu. Takováto patologie je pokládána za důsledek déletrvajícího používání nárožního jařma. Nález představuje jeden z mála dokumentovaných dokladů použití jařma v pravěku na území ČR. Z jiného objektu je doložena bezrohá ovce.

- Huntingtonova choroba (Huntington´s disease, HD) je smrtelné dědičné neurodegenerativní onemocnění s nástupem projevů až v dospělosti. Onemocnění je způsobeno expanzí cytozin-adenin-guanin (CAG) repetic v genu pro protein huntingtin (Htt), který je exprimován ve většině tkání. HD je charakteristická především rozsáhlou degenerací buněk centrální nervové soustavy, ale mutace má velký dopad i na další orgány a tkáně. Mechanismy těchto změn nejsou stále dostatečně popsány. Jednou z nezbytných součástí výzkumu HD jsou zvířecí modely., Huntington's disease (HD) is a fatal inherited neurodegenerative disease with onset of symptoms in adulthood. The disease is caused by the expansion of CAG repeats in the gene for the huntingtin protein, which is expressed in most tissues. HD is characterized by extensive degeneration of the cells of the central nervous system, but the mutation has a large impact on other organs and tissues too. - a1_The emergence of cryptosporidiosis, a zoonotic disease of the gastrointestinal and respiratory tract caused by Cryptosporidium Tyzzer, 1907, triggered numerous screening studies of various compounds for potential anti-cryptosporidial activity, the majority of which proved ineffective. Extracts of Indonesian plants, Piper betle and Diospyros sumatrana, were tested for potential anti-cryptosporidial activity using Mastomys coucha (Smith), experimentally inoculated with Cryptosporidium proliferans Kváč, Havrdová, Hlásková, Daňková, Kanděra, Ježková, Vítovec, Sak, Ortega, Xiao, Modrý, Chelladurai, Prantlová et McEvoy, 2016. None of the plant extracts tested showed significant activity against cryptosporidia; however, the results indicate that the following issues should be addressed in similar experimental studies. The monitoring of oocyst shedding during the entire experimental trial, supplemented with histological examination of affected gastric tissue at the time of treatment termination, revealed that similar studies are generally unreliable if evaluations of drug efficacy are based exclusively on oocyst shedding. Moreover, the reduction of oocyst shedding did not guarantee the eradication of cryptosporidia in treated individuals. For treatment trials performed on experimentally inoculated laboratory rodents, only animals in the advanced phase of cryptosporidiosis should be used for the correct interpretation of pathological alterations observed in affected tissue. All the solvents used (methanol, methanol-tetrahydrofuran and dimethylsulfoxid) were shown to be suitable for these studies, i.e. they did not exhibit negative effects on the subjects.
