We investigated the effects of repeated hyperthermic bouts on the heat shock response of heat s hock protein ( HSP ) 72 in skeletal muscle. Rats were assigned to control and hyperthermia groups which were exposed to heated water at 42 °C. The hyperthermia group was further divided into sub -groups: a single bout (H30) or four bouts of hyperthermia for 30 min (H30x4). There was an increase in HSP72 protein content of the H30 groups in both extensor digitorum longus (EDL) and soleus muscles. Moreover, HSP72 protein expression in H30x4 group was significantly higher than in H30 group in both EDL and soleus muscles. The HSP72 mRNA was markedly increased from control levels in the H30 and H30x4 group in both types of muscles . However, HSP72 mRNA of the H30x4 group was lower than that of the H30 group in soleus muscles. Heat shock response of HSP72 is activate d even after repeated bouts of hyperthermia, with a differential regulation between muscle types., J. Lee, K. Himori, D. Tatebayashi, M. Abe, T. Conventional radiotherapy with X- and γ-rays is one of the common and effective treatments of cancer. High energy hadrons, i.e., charged particles like protons and 12C ions, due to their specific physics and radiobiological advantages are increasingly used. In this study, effectiveness of different radiation types is evaluated on the radio-resistant human HTB140 melanoma cells. The cells were irradiated with γ-rays, the 62 MeV protons at the Bragg peak and in the middle of the spread-out Bragg peak (SOBP), as well as with the 62 MeV/u 12 C ions. The doses ranged from 2 to 24 Gy. Cell survival and proliferation were assessed 7 days after irradiation, whereas apoptosis was evaluated after 48 h. The acquired results confirmed the high radio-resistance of cells, showing better effectiveness of protons than γ-rays. The best efficiency was obtained with 12C ions due to higher linear energy transfer. All analyzed radiation qualities reduced cell proliferation. The highest proliferation was detected for 12C ions because of their large killing capacity followed by small induction of reparable lesions. This enabled unharmed cells to preserve proliferative activity. Irradiations with protons and 12C ions revealed similar moderate pro-apoptotic ability that is in agreement with the level of cellular radio-resistance., A. Ristić-Fira ... The kinetics of Ig-positive cell populations in carp tissues was followed during an infection with the gut dwelling coc-cidian Goussia carpelli Léger et Stankovich, 1921. In cell suspensions of the anterior and posterior sections of the intestine, the proportion of Ig-positive cells increased with the development of the coccidia and peaked during oocyst formation at day 15 post exposure. These results suggest a reaction of the local mucosal immune system. In cell suspensions of pronephros the proportion of Ig-positive cells increased as well, indicating that a systemic immune response was also induced against this intestinal cocci-dian parasite of carp.
In Indian agro-ecosystems Rhynocoris marginatus (F.) is one of the most abundant predatory arthropods and feeds on a wide range of insect pests. We investigated the responses of R. marginatus to six species of cotton pests: Spodoptera litura (F.), Sylepta derogata (F.), Pericallia ricini (F.), Mylabris indica (Thunberg), Mylabris pustulata (Thunberg) and Dysdercus cingulatus (F.), in terms of its predatory behaviour (approach and handling times), weight gain, macromolecular profile (content of carbohydrates, proteins, free aminoacids and lipids) and haemocytic profile. We also determined the predator's reliance on kairomones from different species of prey. Larvae of the species of Lepidoptera studied were approached and captured more quickly than adults of the two meloid coleopteran and one heteropteran pest and were more beneficial to the predator in terms of weight gain. Predators had a higher total protein content when reared on larvae of the three lepidopteran species, higher lipid content when reared on adults of the two meloid coleopteran species and a higher carbohydrate content when reared on adults of one heteropteran species. The number of haemocytes was greater in predators reared on larvae of the Lepidoptera studied, followed by those reared on adults of the heteropteran and lowest in those reared on adults of the two meloid coleopterans. Response to kairomones was strongest for S. litura followed by S. derogata and M. pustulata. We conclude that the Lepidoptera studied tended to be, for this predator, superior prey, with S. litura being especially beneficial and the prey species for which R. marginatus has the highest kairomonal preference. Moreover, we propose that R. marginatus may be useful as a biocontrol agent against lepidopteran cotton pests., Kitherian Sahayaraj, Subramanian Muthu Kumar, Annie Enkegaard., and Obsahuje bibliografii
This study aimed to compare the effects of three different resistance exercise models on the quadriceps muscle crosssectional area, as well as on mTOR phosphorylation and other pivotal molecules involved in the upstream regulation of mTOR. Twenty-four male Wistar rats were divided into untrained (control), endurance resistance training, strength resistance training, and hypertrophy resistance training (HRT) groups (n=6). After 12 weeks of training, the red portion of the quadriceps was removed for histological and Western blot analyses. The results showed that the quadriceps weight and cross-sectional areas in the exercised groups were higher than those of the untrained rats. However, the HRT group presented better results than the other two experimental groups. This same pattern was observed for mTOR phosphorylation and for the most pivotal molecules involved in the upstream control of mTOR (increase of PKB, 14-3-3, ERK, p38 MAPK, and 4E-BP1 phosphorylation, and reduction of tuberin, sestrin 2, REDD1, and phospho AMPK). In summary, our study showed that HRT leads to high levels of mTOR phosphorylation as well as of other proteins involved in the upstream regulation of mTOR., T. F. Luciano, S. O. Marques, B. L. We studied the psychophysiology of soluble intercellular adhesion molecule-1 (sICAM-1) in 25 apparently healthy middle-aged men who underwent an acute psychosocial stressor three times with one week apart. Measures of the biological stress response were obtained at week one and three. The magnitude of the sICAM-1 stress response showed no habituation between individual visits. At week one, cognitive stress appraisal independently predicted integrated sICAM-1 area under the curve (AUC) between rest, immediately post-stress, and 45 min and 105 min post-stress (β=0.67, p=0.012, ΔR2=0.41). Diastolic blood pressure AUC (β=-0.45, p=0.048, ΔR2=0.21) and heart rate AUC (β=0.44, p=0.055, ΔR2=0.21) were independent predictors of sICAM-1 AUC at week three. Adjustment for hemoconcentration yielded a decrease in sICAM-1 levels from rest to post-stress (p<0.001). Stress responsiveness of plasma sICAM-1 was predicted by stress perception and hemodynamic reactivity and affected by stress-hemoconcentration but unrelated to cortisol reactivity and not readily adapting to repeated stress., R. von Känel, D. We present the current state of complex circulatory dynamics model development based on Guyt on’s famous diagram. The aim is to provide an open-source model that will allow the simulation of a number of pathological conditions on a virtual patient including cardiac, respiratory, and kidney failure. The model will also simulate the therapeutic influence of various drugs, infusions of electrolytes, blood transfusion, etc. As a current result of implementation, we describe a co re model of human physiology targeting the systemic circulation, arterial pressure and body fluid regulation, including short- and long-term regulations. The model can be used for educational purposes and general reflection on physiological regulation in path ogenesis of various diseases., J. Kofránek, J. Rusz., and Obsahuje bibliografii
The results of geodetic GNSS measurements on the EYPA station (bult by INSU CNRS from France) in Corinth Gulf in Greece are analyzed. Data is analyzed in the time interval before and after the earthquakes, which occured in January 2010. Periprosthetic fractures are the third most common reason for revision total hip arthroplasty. Surgical treatment of periprosthetic fractures belongs to the most difficult procedures due to the extensive surgery, elderly polymorbid patients and the high frequency of other complications. The aim of this study was to evaluate the results of operatively treated periprosthetic femoral fractures after total hip arthroplasty. We evaluated 47 periprosthetic fractures in 40 patients (18 men and 22 women) operated on between January 2004 and December 2010. The mean follow-up period was 27 months (within a range of 12-45 months). For the clinical evaluation, we used modified Merle d'Aubigné scoring system. In group of Vancouver A fractures, 3 patients were treated with a mean score of 15.7 points (good result). We recorded a mean score of 14.2 points (fair result) in 6 patients with Vancouver B1 fractures, 12.4 points (fair result) in 24 patients with Vancouver B2 fractures and 12.7 points (fair result) in 7 patients with Vancouver B3 fractures. In group of Vancouver C fractures, we found a mean score of 16.2 points (good result) in 7 patients. Therapeutic algorithm based on the Vancouver classification system is, in our opinion, satisfactory. Accurate differentiation of B1 and B2 type of fractures is essential. Preoperative radiographic images may not be reliable. If in doubt, checking the stability of the prosthesis fixation during surgery should be performed. and M. Korbel, P. Sponer, T. Kucera, E. We present first results of the study of possible relations between the seismic activity and crustal fluids (groundwater an d carbon dioxide) in the area of the Hronov-Poříčí Fault Zone (HPFZ), situated on the NE margin of the Bohemian Massif. Local seismic monitoring and observations of groundwater levels in deep wells and concentrations of carbon-dioxide in the mineral spring at Třtice was started in 2005. Since then, more than 30 local seismic events were observed in the area of the HPFZ. The two strongest earthquakes with macroseismic effects were recorded on August 10, 2005 (M = 2.4) and October 25, 2005 (M = 3.3). Most of the epicentres were situated along the central part of the HPFZ. Only some weak events from February and March 2006 were concentrated along the SE termination of the HPFZ. Results of the hydrological monitoring show that water level fluctuations are affected mainly by the precipitation, snow-melt, air pressure changes, and tidal deformations of the Earth’s crust. The effects of seismo-tectonic activity were detected only in one out of five water wells, where we observed several step-like water level anomalies with amplitudes of 4 to 15 cm. Two of them preceded the August 10, 2005 and October 25, 2005 earthquakes. Three other anomalies seemed to originate independently of the seismic activity. We therefore suppose that they were induced by aseismic movements along the HPFZ. Contrary to the water level fluctuations, CO2 concentrations in the mineral spring seem to be dependent on water temperature; no evident seismic-induced changes have been observed yet., Vladimír Stejskal, Lumír Skalský and Ladislav Kašpárek., and Obsahuje bibliografické odkazy
