2. Associations between cardiac fibrosis and permanent atrial fibrillation in advanced heart failure

- Atrial fibrosis is considered as the basis in the development of long-standing atrial fibrillation (AF). However, in advanced heart failure (HF), the independent role of fibrosis for AF development is less clear since HF itself leads to atrial scarring. Our study aimed to differentiate patients with AF from patients without AF in a population consisting of patients with advanced HF. Myocardial samples from the right atrial and the left ventricular wall were obtained during he art transplantation from the explanted hearts of 21 male patients with advanced HF. Long- standing AF was present in 10 of them and the remaining 11 patients served as sinus rhythm controls. Echocardiographic and hemodynamic measurements were recorded prior to heart transplantation. Collagen volume fraction (CVF), transforming growth factor-beta (TGF- β ), and connective tissue growth factor (CTGF) expression in myocardial specimens were assessed histologically and immunohistochemically. The groups were well matched according to age (51. 9±8.8 vs. 51.3±9.3 y) and co- morbidities. The AF group had high er blood pressure in the right atrium (13.6±7.7 vs. 6.0±5.0 mmHg; p=0.02), larger left atrium diameter (56.1±7.7 vs. 50±5.1 mm; p=0.043), higher left atrium wall stress (18.1±2.1 vs. 16.1±1.7 kdynes/m 2 ; p=0.04), and longer duration of HF (5.0±2.9 vs. 2.0±1.6 y, p=0.008). There were no significant differences in CVF (p=0.12), in CTGF (p=0.60), and in TGF- β expression (p=0.66) in the atrial myocardium between the two study groups. In conclusions, in advanced HF, atrial fibrosis expressed by CVF is invariably present regardless of occurrence of AF. - Analyzovali jsme stanoviště a rozšíření houby šupinovky Henningsovy (Pholiota henningsii) v Evropě, kde je známa z Holandska, Akvitánie, Pařížské pánve, Massiv Central, úpatí Pyrenejí a Alp ve Francii a Švýcarsku, třetihorních pánví v Rakousku a jižních Čechách, z Mazurských jezer v Polsku, ostrova Sjaelland v Dánsku a ostrovů poblíž Stockholmu ve Švédsku. Všude je velmi vzácná a v mnoha zemích je zařazena na červených seznamech ohrožených druhů hub. Roste na slatinách, přechodových rašeliništích i vrchovištích, jak živinami bohatých, tak chudých. Váže se na lokality a oblasti s dlouhodobou (tisíciletou) historií otevřených rašelinišť ležících v blízkosti vodních nádrží. V ČR byly tři historické lokality doplněny v roce 2012 o dvě nové. Dlouhodobá mozaika lesa a bezlesí se zdá být hlavní podmínkou pro přežívání tohoto druhu reliktních stanovišť., The occurrence of the rare fungus Pholiota henningsii has been analysed in Europe. It is known from the Netherlands, the Parisian and Aquitanian basins, the Massif Central, and the foothils of the Pyrenées and Alps in France, Switzerland and Austrian Terciary basins, with numerous mires, in southern Bohemia, the Mazury lakes in Poland, Sjaelland island in Denmark and the island region near Stockholm in Sweden. In many countries it is included in Red Lists of endangered species. The fungus occurs in spring fens, transitional mires and raised bogs, both nutrient rich and oligotrophic. It is confined to localities with long term (i.e. millennia-long) mire non-forest continuity, in the vicinity of open water bodies up to the present. In the Czech Republic three historical sites were completed by two new localities in 2012. A temperate and forest - treeless mire mosaic seems to be the conditional factor for the long-term persistence of this relict species., and Jan Holec, Tomáš Kučera.

- Sirtuin 1 (SIRT1) is involved in important biological processes such as energy metabolism and regulatory functions of the cell cycle, apoptosis, and inflammation. Our previous studies have shown hepatoprotective effect of polyphenolic compound resveratrol, which is also an activator of SIRT1. Therefore, the aim of our present study was to clarify the role of SIRT1 in process of hepatoprotection in animal model of drug-induced liver damage. Male Wistar rats were used for both in vivo and in vitro studies. Hepatotoxicity was induced by single dose of acetaminophen (APAP). Some rats and hepatocytes were treated by resveratrol or synthetic selective activator of sirtuin 1 (CAY10591). The degree of hepatotoxicity, the activity and expression of the SIRT1 were determined by biochemical, histological and molecular-biological assessments of gained samples (plasma, liver tissue, culture media and hepatocytes). Resveratrol and CAY attenuated APAP-induced hepatotoxicity in vivo and in vitro. Moreover, both drugs enhanced APAPreduced SIRT1 activity. Our results show that modulation of the SIRT1 activity plays a role in hepatoprotection. Synthetic activators of SIRT1 would help in understanding the role of SIRT1 and are therefore a major boost towards the search for specific treatment of liver disease., L. Wojnarová, N.
