The impact of drought stress (DS) on eight Eurasian and North African genotypes of wild barley (Hordeum spontaneum) was evaluated by analysis of chlorophyll (Chl) a fluorescence fast induction curves using the JIP-test. Three-week-old, pot-grown plants were exposed to a DS treatment by withholding water for nine days. The genotype-specific impairment of the functionality of the photosynthetic electron transport chain was quantified using the relative decline of the performance indices (PIabs and PItot), two key parameters of the JIP-test. The genotypes showing the highest (HOR10164) and lowest (HOR10710) relative PIs under DS were subjected to additional experiments, including measurements of leaf gas exchange, water status, pigment content, key enzyme activity, and protein abundance. The genotypes showed a specific profile of DS-mediated inhibition of photosynthesis, associated with higher relative leaf water contents in HOR10164 at the end of the treatment. Whereas decreased photosynthetic rate in HOR10164 was mainly caused by stomatal closure, nonstomatal limitations (decreased Rubisco content and activity) were detected in HOR10710. The effect of drought stress (DS) on photosynthesis and photosynthesis-related enzyme activities was investigated in F. pringlei (C3), F. floridana (C3-C4), F. brownii (C4-like), and F. trinervia (C4) species. Stomatal closure was observed in all species, probably being the main cause for the decline in photosynthesis in the C3 species under ambient conditions. In vitro ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and stromal fructose 1,6-bisphosphatase (sFBP) activities were sufficient to interpret the net photosynthetic rates (PN), but, from the decreases in PN values under high CO2 (C a = 700 µmol mol- 1) it is concluded that a decrease in the in vivo rate of the RuBPCO reaction may be an additional limiting factor under DS in the C3 species. The observed decline in the photosynthesis capacity of the C3-C4 species is suggested to be associated both to in vivo decreases of RuBPCO activity and of the RuBP regeneration rate. The decline of the maximum PN observed in the C4-like species under DS was probably attributed to a decrease in maximum RuBPCO activity and/or to decrease of enzyme substrate (RuBP or PEP) regeneration rates. In the C4 species, the decline of both in vivo photosynthesis and photosynthetic capacity could be due to in vivo inhibition of the phosphoenolpyruvate carboxylase (PEPC) by a twofold increase of the malate concentration observed in mesophyll cell extracts from DS plants. and M. Five-year-old trees of deciduous Quercus robur L., evergreen Q. ilex L., and their semideciduous hybrid, Q. × turneri Willd. (var. pseudoturneri), growing in pots, were subjected to drought stress by withholding water for 18-22 days, until leaf water potentials decreased below -2 MPa. Gas-exchange rates, oxygen evolution, and modulated chlorophyll (Chl) fluorescence measurements revealed that by strong stomata closure and declining photosynthetic capacity down to approximately 50%, all three taxa responded with strongly reduced photosynthesis rates. In Q. robur, photochemical quenching of the drought-stressed plants was much lower than in nonstressed controls. The four Fabaceae species of different chilling tolerance (Pisum sativum L., Glycine max. Merr., Phaseolus vulgaris L. and Vigna unguiculata L.) were subjected to simulated bad weather periods at the beginning of the growth season. The treatment parameters were 14 d at constant 10 “C, 100 pmol m-2 s"*, 80 % relative humidity within a 12 h photoperiod. During chilling and subsequent recovery, photosynthesis, chlorophyll fluorescence and assimilate concentrations, as well as ribulose-1,5- bisphosphate carboxylase/oxygenase (RuBPCO) activity were followed. Despite the observation that no significant photoinhibition had been induced by the treatment, French beán and cowpea showed a strong decrease of photosynthetic capacity within the first week. Diuring chilling, soybean, French beán and cowpea virtually did not grow fiirther, but acciunulated large amounts of starch, in contrast to pea. Specific activity of RuBPCO (Mg2+- and HC03'-activated) was not altered significantly by the chilling treatment. Yet in French beán a generál protein breakdovm occurred in the second week. At the same time, a very strong decrease of qp was obtained in this species, indicating a breakdown of the electron transport. Pea and soybean were not negatively afifected by the long-term chilling.
Metabolite changes and senescence behaviour after mechanical phloem girdling were studied in leaf tissue of Quercus pubescens. Sugar accumulation is not only considered to be an important part of several developmental signalling pathways, but is also seen as one of the basic triggers for senescence induction, or at least an obligatory accessory phenomenon. Our survey showed that an accumulation of the soluble sugars, glucose and fructose, was not on its own obligatorily connected with the induction of leaf senescence, since no indication or even an onset of senescence could be observed during the course of the experiment. Instead, we observed an inhibition of leaf development with a decrease of photosynthesis and a slow-down of development in nearly all chlorophyll a fluorescence analysis parameters using the JIP-test. The responses to root oxygen deficiency conceming the photosynthesis, saccharide contents and minerál uptake háve been investigated in Carex species, which were different in their anoxia-tolerance. The net rate of photosynthesis (Fn) anoxia- sensitive C. extensa was not affected by root anaerobiosis. Despite short-term effects of root anaerobiosis on values in the anoxia-tolerant species C. remota and C. pseudocyperus, root oxygen supply did not influence P^, when the plants were allowed to acclimate. The saccharide contents of the anoxia-tolerant species were almost not affected by prolonged anaerobiosis. 40 days of root oxygen deficiency led to significant changes in the shoot saccharide composition of C. extensa: contents of glucose and fructose were increased 4-fold and sucrose content was significantly decreased. In the roots no sucrose could be detected, but the starch content was increased, and the glucose and fructose contents did not change when compared with aerobic conditions. Under anaerobic conditions, minerál uptake of the anoxia- sensitive species decreased by 50 %, while the minerál uptake of the anoxia-tolerant species was significantly increased or did not change at all, in comparison with the aerobic conditions. The growth reduction of C. extensa under anaerobic conditions was therefore primarily due to inability to také up an adequate amount of nutrients.
In this article, the effects of drought stress (DS) on gas exchange, chlorophyll (Chl) a fluorescence and Calvin cycle enzymes in Phaseolus vulgaris are evaluated. Three-week-old plants were exposed to DS by receiving only so much water every evening to ensure 30% field capacity water content overnight. After three days under these conditions, we observed that DS induced a decline of the CO2 assimilation. Gas-exchange data showed that the closure of stomata during DS did not lead to a concomitant decline in calculated intercellular CO2 concentration. Moreover, DS plants showed a reduction of the photochemical Chl fluorescence quenching, photosystem II quantum yield and electron transport rate and a higher pH gradient and more heat dissipation as compared to controls. The activity of Calvin cycle enzymes, Rubisco, sFBPase, and Ru5PK, decreased strongly in DS plants as compared to controls. Data analysis suggest that the decrease of CO2 assimilation under drought conditions is not related to a diminished capacity of the use of NADPH and ATP but probably to the decline of enzyme activity involved in RuBP regeneration (Ru5PK). and M. In search for new forestation tree species for future Central European climate conditions, Mediterranean evergreen oak taxa are investigated for their summer drought- and winter frost-hardiness. Here we report on the winter performance of the photosynthetic apparatus of Quercus × hispanica Lam. and its evergreen parental species Q. suber L. under extraordinary harsh winter conditions. Both taxa showed a strong decline of photosystem II (PSII) quantum efficiency (Fv/Fm) with a concomitant increase in the deepoxidation state (DES) of the xanthophyll pigments depending on (severe) frost events during winter, and these parameters significantly correlated with minimum air temperatures during periods of chronic photoinhibition at mid-winter, but not at the onset of winter in response to the first frost nights. Environmental conditions that promote photorespiration are considered to be a major driving force for the evolution of C4 species from C3 ancestors. The genus Flaveria contains C3 and C4 species as well as a variety of intermediate species. In this study, we compare the water-use efficiency of intermediate Flaveria species to that of C3 and C4 species. The results indicate that under both well-watered and a drought-stress condition, C3-C4 and C4-like intermediacy in Flaveria species improve water-use efficiency as compared to C3 species. and M. C.
