The effects of drought on thylakoid acyl lipid composition, photosynthetic capacity (P max), and electrolyte lekage were evaluated in two-months-old peanut cultivars (57-422, 73-30, GC 8-35) growing in a glasshouse. For lipid studies, plants were submitted to three treatments by withholding irrigation: control (C), mild water stress (S1), and severe water stress (S2). Concerning membrane and photosynthetic capacity stability, drought was imposed by polyethylene glycol (PEG 600). In the cv. 73-30 a sharp decrease in the content of thylakoid acyl lipids was observed, already under S1 conditions, whereas cv. 57-422 was strongly affected only under S2. Cv. GC 8-35 had the lowest content of acyl lipids under control conditions, a significant increase under S1 conditions, and only under S2 a decrease occurred. Thus concerning lipid stability, cv. 73-30 was the most sensitive. Among lipid classes, phospholipids and galactolipids were similarly affected, as was MGDG relatively to DGDG. Water deficit imposed by PEG induced a higher increase in electrolyte leakage in cv. 73-30 than in the other cvs. A positive relationship between acyl lipid concentration and membrane integrity was found in all studied cvs. A positive association between acyl lipid concentration, membrane integrity, and P max was found in the cvs. 57-422 and 73-30. and J. We investigated whether polyethylene glycol-coated Fe3O4 nanoparticles (IONs), acute stress and their combination modifies vascular functions, nitric oxide synthase (NOS) activity, mean arterial pressure (MAP) as well as hepcidin and ferritin H gene expressions in Wistar-Kyoto rats. Rats were divided into control, ION-treated rats (1 mg Fe/kg i.v.), repeated acute air jet stressexposed rats and IONs-and-stress co-exposed rats. Maximal acetylcholine (ACh)-induced and sodium nitroprusside (SNP)-induced relaxations in the femoral arteries did not differ among the groups. IONs alone significantly elevated the Nω-nitroL-arginine methyl ester (L-NAME)-sensitive component of ACh-induced relaxation and reduced the sensitivity of vascular smooth muscle cells to SNP. IONs alone also elevated NOS activity in the brainstem and hypothalamus, reduced NOS activity in the kidneys and had no effect in the liver. Acute stress alone failed to affect vascular function and NOS activities in all the tissues investigated but it elevated ferritin H expression in the liver. In the ION-and-stress group, NOS activity was elevated in the kidneys and liver, but reduced in the brainstem and hypothalamus vs. IONs alone. IONs also accentuated air jet stress-induced MAP responses vs. stress alone. Interestingly, stress reduced ION-originated iron content in blood and liver while it was elevated in the kidneys. In conclusion, the results showed that 1) acute administration of IONs altered vascular function, increased L-NAME-sensitive component of ACh-induced relaxation and had tissue-dependent effects on NOS activity, 2) ION effects were considerably reduced by co-exposure to repeated acute stress, likely related to decrease of ION-originated iron in blood due to elevated decomposition and/or excretion.
Prosopis juliflora is a hardy plant tolerant to drought, salinity, extremes of soil pH, and heavy metal stress. We isolated and characterized a photosystem 2 (PS2) gene PsbR (Pj PsbR) and its promoter. Northern analysis for Pj PsbR in P. juliflora leaves under 25 % polyethylene glycol stress showed steady decrease in transcript level at 12, 24, and 48 h after stress application. Under 90 mM H2O2 stress, transcript level dropped drastically at 12 h, but increased again compared to the control at 24 h. A 1.7 kb fragment upstream the 5' UTR of this gene (putative promoter) was isolated and analyzed in silico. Several putative cis-acting DNA elements were identified in this sequence. and G. Suja, A. Six genotypes of taro (Colocasia esculenta L. Schott) were evaluated under in vitro and in vivo polyethylene glycol (PEG-6000)-mediated osmotic stress conditions. A significant variation in growth response was observed among the taro genotypes under in vitro-induced stress conditions. In vivo results indicated a significant effect of osmotic stress on photosynthetic parameters, such as net photosynthetic rate, transpiration rate, stomatal conductance, stomatal resistance, internal CO2 concentration, carboxylation efficiency, and transpiration efficiency on the tested genotypes at the tuberization stage. Lesser variations in photosynthesis and higher accumulation of proline, phenols, and antioxidative enzymes, namely, superoxide dismutase and guaiacol peroxidase, were associated with yield maintenance under osmotic stress conditions. The genotypes DP-89, IGCOL-4, and Ramhipur showed a higher degree of tolerance towards osmotic stress with a minimum variation in the studied parameters. Seedlings of Cyamopsis tetragonoloba were grown on Petri dishes either in water or water plus 3 % PEG-6000 to induce water stress. The senescing cotyledons experiencing the stress exhibited loss in contents of leaf proteins and chlorophyll (Chl) and a decline in oxygen evolution. The effect of PEG treatment was more pronounced at moderate (MI) than low (LI) irradiance. The stress-induced loss in the activity of superoxide dismutase and increase in the thylakoid lipid peroxidation accompanied a change in the physical status of the bilayer membrane as demonstrated by an enhancement of room temperature Chl a fluorescence polarization and decrease in energy transfer efficiency in pigment assembly. This resulted in a sustained decrease in photosystem 2 activity blocking channels of energy utilization.
