Chicken turtles, Deirochelys reticularia (Latreille in Sonnini et Latreille) (Testudines: Emydidae) from Alabama, USA were infected by Spirorchis collinsi Roberts et Bullard sp. n. and Spirorchis cf. scripta. The new species is most easily differentiated from its congeners by the combination of having caeca that extend far beyond the genitalia, intercaecal genitalia positioned in the middle portion of the body, a testicular column that nearly abuts the caecal bifurcation, a cirrus sac positioned between the testes and ovary, a massive Mehlis' gland, an elongate, longitudinal metraterm that extends anteriad beyond the level of the ovary, a pre-ovarian genital pore, and a prominent, intercaecal Manter's organ. The specimens of S. cf. scripta differed from the holotype and published descriptions of Spirorchis scripta Stunkard, 1923 by several subtle morphological features, perhaps comprising intraspecific variation, but collectively warranted a detailed description herein. Based on examinations of the aforementioned specimens plus the holotype, paratypes and vouchers of morphologically-similar congeners, Spirorchis MacCallum, 1918 is emended to include the presence of oral sucker spines, a pharynx, lateral oesophageal diverticula ('plicate organ') and a median oesophageal diverticulum ('oeseophageal pouch'). Phylogenetic analysis of the nuclear large subunit rDNA (28S) recovered S. collinsi sister to Spirorchis picta Stunkard, 1923, > 99% similarity between S. cf. scripta and S. scripta, and a monophyletic Spirorchis MacCallum, 1918. No blood fluke infection has been reported previously from these drainages, Alabama, or this turtle species. Wallinia mexicana sp. n. is described from the Mexican tetra, Astyanax mexicanus (De Filippi) (Characidae Weitzman), from two localities in northern Mexico. The new species can be distinguished from the two congeneric species, described from small-bodied characids in South and Central America, mainly by the posterior extent of the vitelline follicles (halfway between the posterior testis and the end of the caeca), by having a larger oesophagus, testes that are always oblique, and eye-spot remnants. The distinct status of the new species was confirmed by molecular data (28S rRNA gene sequences). Phylogenetic analysis suggests the new species is the sister species of W. chavarriae Choudhury, Hartvigsen et Brooks, 2002 described from characids in northwestern Costa Rica. Additionally, genetic divergence between these congeners reached 3.3%, a value higher than that observed for closely related species pairs of allocreadiids for that molecular marker. Combining a biotin-enrichment protocol and 454GS-FLX titanium pyrosequencing technology, we characterised 22 polymorphic microsatellite loci from the parasitic wasp, Habrobracon hebetor (Say) (Hymenoptera: Braconidae), a cosmopolitan species commonly used in biological control against a wide range of both major lepidopterous pests of stored products and field crops in different parts of the world. Three multiplex PCR sets were optimised and characterised across 46 H. hebetor specimens from two samples collected from millet fields in Niger. Two to 11 alleles were found per locus and observed heterozygosity ranged from 0.289 to 0.826. Polymorphism was detected in both samples with a similar level of observed heterozygosity (0.482 vs. 0.502) and number of alleles (4.1 vs. 3.6). Deviation from Hardy-Weinberg equilibrium was detected at the same five loci in both samples and five or seven more loci in each sample but was not associated with heterozygote deficiencies. Even though evidence for linkage disequilibrium was found between a few alleles, these new loci segregated independently. The variability of the 22 loci will enable estimates of genetic diversity and structure patterns, as well as gene flow between H. hebetor populations at different spatial scales. Here we describe the new trypanosomatid, Phytomonas borealis sp. n., from the midgut of the spiked shieldbugs, Picromerus bidens (Linnaeus), collected in two locations, Novgorod and Pskov Oblasts of Russia. The phylogenetic analyses, based on the 18S rRNA gene, demonstrated that this flagellate is a sister species to the secondary monoxenous Phytomonas nordicus Frolov et Malysheva, 1993, which was concurrently documented in the same host species in Pskov Oblast. Unlike P. nordicus, which can complete its development (including exit to haemolymph and penetration into salivary glands) in Picromerus bidens, the new species did not form any extraintestinal stages in the host. It also did not produce endomastigotes, indispensable for transmission in other Phytomonas spp. These observations, along with the fact that P. bidens overwinters at the egg stage, led us to the conclusion that the examined infections with P. borealis were non-specific. Strikingly, the flagellates from the Novgorod population contained prokaryotic endosymbionts, whereas the parasites from the second locality were endosymbiont-free. This is a first case documenting presence of intracellular symbiotic bacteria in Phytomonas spp. We suggest that this novel endosymbiotic association arose very recently and did not become obligate yet. Fishway design not only takes into account the swimming abilities of target fishes, but also considers the hydrodynamic characteristics within the fishway. In this study, the flow fields of one vertical-slot fishway (i.e. VSF), five T-shape fishways (i.e. TSF-1~TSF-5) and two H-shape fishways (i.e. HSF-1 and HSF-2) are numerically simulated by solving the three-dimensional Reynolds-averaged Navier-Stokes equations and the K-Omega-SST turbulence model. The numerical results clearly indicate that the hydrodynamic properties of HSF-2 are overall superior to the remaining seven cases, in terms of the time-averaged flow pattern, the time-averaged velocity magnitude, the depth-mean timeaveraged velocity magnitude along the vertical-slot section, the volume percentages of the time-averaged velocity magnitude less than some critical values, and the distribution of the time-averaged turbulent kinetic energy. Therefore, HSF-2 is more friendly for fishes with relatively smaller sizes and weaker swimming capacities to transfer upstream. The novel HSF-2 is firstly proposed in this paper, and it is naturally designed during the process of improving the flow regime. Furthermore, the generalizability of the superiority of HSF-2 over VSF and the original T-shape fishway (i.e. TSF-1) has been exhibited with the aid of the numerical results of four operating conditions (i.e. Q = 400 L/s, 600 L/s, 800 L/s and 1000 L/s).
As part of a biodiversity study in northwestern Hungary, we conducted a parasitological survey of small mammals. In both common shrews (Sorex araneus Linnaeus) and pygmy shrews (Sorex minutus Linnaeus), we found myxospores of a species of Soricimyxum Prunescu, Prunescu, Pucek et Lom, 2007 (Myxosporea) and plasmodia in the bile ducts within the liver. Spores from both species of shrewswere morphologically and morphometrically indistinguishable, but differed in their SSU rRNA gene sequences by 3.3%. We identified spores and developmental stages from the common shrew as Soricimyxum fegati Prunescu, Prunescu, Pucek et Lom, 2007, based on morphometric data and DNA sequence similarity. Spores from the pygmy shrew were only 96.7% similar to S. fegati, hence we identified them as a novel myxosporean Soricimyxum minuti sp. n. Nucleoside diphosphate kinases (NDPK) are key enzymes involved in the intracellular nucleotide maintenance in all living organisms, especially in trypanosomatids which are unable to synthesise purines de novo. Four putative NDPK isoforms were identified in the Trypanosoma cruzi Chagas, 1909 genome but only two of them were characterised so far. In this work, we studied a novel isoform from T. cruzi called TcNDPK3. This enzyme presents an atypical N-terminal extension similar to the DM10 domains. In T. cruzi, DM10 sequences targeted other NDPK isoform (TcNDPK2) to the cytoskeleton, but TcNDPK3 was localised in glycosomes despite lacking a typical peroxisomal targeting signal. In addition, TcNDPK3 was found only in the bloodstream trypomastigotes where glycolytic enzymes are very abundant. However, TcNDPK3 mRNA was also detected at lower levels in amastigotes suggesting regulation at protein and mRNA level. Finally, 33 TcNDPK3 gene orthologs were identified in the available kinetoplastid genomes. Objectives. The aim of this study was to explore possible relationships between the HEXACO personality traits (i.e., Extraversion, Emotionality, Honesty-Humility, Agreeableness, Conscientiousness, Openness to experience) and negative effects of psychotherapy. Sample and settings. A total of 148 respondents answered an online survey. Descriptive and multivariate regression analyses were performed. Hypotheses. Clients’ personality traits were expected to predict negative effects of psychotherapy. Results. Extraversion was the strongest negative predictor of negative effects, serving as a protective factor against worsening of symptoms, hopelessness, and stigma. Furthermore, Emotionality was related to lower perceived quality of psychotherapy, Conscientiousness was related to hopelessness, and Openness to experience served as a protector against dependency. Limitations. The findings are based on a onetime retrospective measurement and a convenience sample. The results must be interpreted with caution because the assumptions of linear regression were not fulfilled. and Cíle.Cílem této studie bylo prozkoumat možné vztahy mezi osobnostními rysy HEXACO (tj. Extraverze, Emocionalita, Poctivost-Pokora, Přívětivost, Svědomitost, Otevřenost vůči zku-šenosti) a negativními účinky psychoterapie.Výzkumný soubor a metody.V online průzkumu odpovídalo celkem 148 respondentů. Ve výzku-mu byly provedeny deskriptivní a vícerozměrné regresní analýzy. Hypotézy. Od osobnostních rysů klientů autoři očekávali, že predikují negativní účinky psy-choterapie. Výsledky. Extraverze byla nejsilnějším nega-tivním prediktorem negativních účinků psy-choterapie, sloužila jako ochranný faktor před zhoršením symptomů, beznadějí a stigmatizací. Faktor Emocionalita souvisel s nižší vnímanou kvalitou psychoterapie, Svědomitost souvisela s beznadějí a Otevřenost vůči zkušenosti slouži-la jako ochrana před závislostí na psychoterapii. Limity.Nálezy jsou založeny na jednorázovém retrospektivním měření a na limitujícím vzorku. Výsledky je nutné interpretovat opatrně, protože předpoklady lineární regrese nebyly zcela spl-něny.Závěr.Tato pilotní studie poskytla nové důkazy o roli faktorů na straně klienta při vysvětlování negativních účinků v psychoterapii. V datech vysvětlovaly osobnostní rysy HEXACO od 9 % do 23 % rozptylu u negativních účinků psycho-terapie. I když to znamená, že tyto osobnostní rysy hrají důležitou roli při vzniku negativních jevů, procenta vysvětlené variance (spolu se zkreslenými rezidui na provedených regres-ních analýzách) jasně ukázala, že existují další podstatné faktory, které je třeba pro vysvětlení těchto účinků objevit. Budoucí studie se mohou pokusit zahrnout kontextové, předterapeutické a terapeutické faktory, terapeutické procesy a důvody ukončení (Curran et al., 2019) spolu s osobnostními rysy klientů, aby určily jejich relativní vlivy na negativní účinky psychoterapie.
As apex predators with a regulating effect on interspecific competitors and prey demographics, monitoring of spotted hyaena (Crocuta crocuta) population trends can provide a reliable indicator of ecosystem health. However, the ability of current survey techniques to monitor carnivore densities effectively are increasingly questioned. This has led recent studies to advocate increased application of spatial capture-recapture (SCR) methods to estimate population density for large carnivores. We reviewed the literature regarding methods used to estimate population density for spotted hyaena since 2000. Our review found that SCR methods are underutilised for estimating spotted hyaena density, with only eight published studies (13% of articles assessed) using an SCR approach. Call-in surveys were the most frequently used method, featuring in 47% of studies. However, 63% of studies that used call-in surveys could not estimate a site-specific calibration index. The calibration index estimates the distance and rate at which the focal species responds to audio lures and, as response rates are impacted by site-specific ecological and environmental factors, studies that could not calibrate this index are likely inaccurate. Further application of SCR techniques will allow more robust estimation of spotted hyaena density, reducing uncertainty and potential overestimation that limit inference from existing survey methods.
Flies of the Colocasiomyia toshiokai species group depend exclusively on inflorescences/infructescences of the aroid tribe Homalomeneae. The taxonomy and reproductive biology of this group is reviewed on the basis of data and samples collected from Southeast Asia. The species boundaries are determined by combining morphological analyses and molecular species delimitation based on sequences of the mitochondrial COI (cytochrome c oxidase subunit I) gene. For the phylogenetic classification within this species group, a cladistic analysis of all the member species is conducted based on 29 parsimony-informative, morphological characters. As a result, six species are recognised within the toshiokai group, including one new species, viz. C. toshiokai, C. xanthogaster, C. nigricauda, C. erythrocephala, C. heterodonta and C. rostrata sp. n. Various host plants are utilised by these species in different combinations at different localities: Some host plants are monopolized by a single species, while others are shared by two or three species. C. xanthogaster and C. heterodonta cohabit on the same host plant in West Java, breeding on spatially different parts of the spadix. There is a close synchrony between flower-visiting behaviour of flies and flowering events of host plants, which indicate an intimate pollination mutualism.
