Four peptides with allatostatic activity were isolated from brains of the Mediterranean field cricket, Gryllus bimaculatus. Three of them (Grb-AST A3: AGMYSFGL-NH2; Grb-AST A4: SRPFGFGL-NH2; Grb-AST A5: GPDHRFAFGL-NH2) belong to the wide-spread family of Y/FXFGL/I-amide peptides, the fourth (Grb-AST B5: AWDQLRPGW-NH2) is a member of the W2W9 - amide family of neuropeptides. All of these peptides are potent inhibitors of juvenile hormone (JH) biosynthesis by cricket corpora allata in vitro, causing 50% inhibition of JH biosynthesis at 0.4-3 × 10-8 M. The two peptides Grb-AST A5 and Grb-AST B5 have virtually the same potency and efficacy in inhibiting JH biosynthesis in vitro. No synergistic effect of the two peptide families with respect to the inhibition of JH biosynthesis could be observed. Peptides of both families decrease the accumulation of methylfarnesoate, the direct precursor of JH, within CA that have been incubated in farnesol-rich medium. Monosodium glutamate (MSG) treatment of neonatal mice results in a selective damage to the arcuate nucleus (ARC) and development of obesity with increased adiposity at sustained body weight in the adulthood. Feeding pattern of the MSG obese mice is unusual. Our previous results showed that after 24-h fasting, MSG mice consumed negligible amount of food in several hours and therefore, it was impossible to register the effect of peptides attenuating food intake such as cholecystokinin (CCK) or cocaine- and amphetamine-regulated transcript (CART) peptide. To overcome this problem, two findings were used: firstly, orexigenic effect of neuropeptide Y (NPY) was attenuated both by CCK or CART peptide in lean fed mice and secondly, orexigenic effect of NPY was preserved in fed rats with MSG obesity. In this study, short-term food intake in fed lean and MSG obese C57BL/6 male mice was measured after simultaneous central administration of orexigenic NPY with either CART peptide or peripherally administered CCK. Anorexigenic action of exogenous CART peptide was preserved in MSG obese mice. On the other hand, satiety effect of exogenous CCK was completely lost in MSG obese mice. This study sought to evaluate whether consumption of polyphenol extract from Cognac (CPC) modulates platelet activation and cardiovascular reactivity in rats. Male Wistar rats were treated daily for 4 weeks by intra-gastric gavage receiving CPC at 80 mg/kg/day or vehicle (5 % glucose). Platelet adhesion and aggregation in response to different activators were assessed. Cardiac and vascular reactivity in response to various agonists as well as NO measurement by electron paramagnetic resonance technique were investigated in isolated heart and thoracic aorta. Oral administration of CPC decreased platelet aggregation induced by ADP but not by collagen. CPC did not affect adhesion to collagen. The chronotropic but not the inotropic response to isoprenaline was reduced without alteration of NO production in hearts from CPC-treated rats. CPC treatment did not affect ex vivo relaxation to acetylcholine nor NO content of rat aorta. CPC did not significantly alter the response to phenylephrine in aorta despite the participation of endothelial vasoconstrictor products. In summary, chronic treatment with CPC has no impact on ex vivo vascular and cardiac reactivity; however, it reduced heart work and platelet aggregation. We measured energy substrates in haemolymph and flight muscles of the large blister beetle Mylabris oculata at rest and after tethered, lift-generating flight. Flight of 1 min duration at an ambient temperature of 38-42°C did not effect a change in the concentration of lipids in the haemolymph, whereas a small, significant decrease in the concentrations of carbohydrates and a 3-fold larger one in the levels of proline were noted, as well as a concomitant increase in alanine. In the flight muscles, glycogen and proline concentrations were diminished slightly but significantly upon flight, whereas alanine levels were increased. Two hours of rest after a flight of 1 min completely reversed the metabolic situation in haemolymph and flight muscles to pre-flight levels. We could isolate two neuropeptides from the corpora cardiaca of M. oculata, which by retention time and mass analyses are characterised as the decapeptide Del-CC (pGlu-Leu-Asn-Phe-Ser-Pro-Asn-Trp-Gly-Asn-NH2) and the octapeptide Tem-HrTH (pGlu-Leu-Asn-PheSer-Pro-Asn-Trp-NH2) previously fully identified from the corpora cardiaca of the blister beetle, Decapotoma lunata (Gäde, 1995). Subsequently, it was unequivocally demonstrated that low doses of Del-CC and Tem-HrTH elicited increases in the concentration of proline and carbohydrates in the haemolymph of D. lunata and M. oculata, but did not change the concentration of lipids in both species. Insect peptidyl-dipeptidase A [angiotensin I - converting enzyme (ACE)] is a soluble single-domain peptidyl-dipeptidase that has many properties in common with the C-domain of mammalian somatic ACE and with the single-domain mammalian ACE. In agreement with a variety of insects, immunocytochemical studies reveal the presence of an ACE-like protein in Locusta migratoria. ACE-like immunoreactivity is present in neurosecretory cells of the pars intercerebralis. These cells have axons projecting into the nervus corporis cardiaci I and into the storage part of the corpus cardiacum, a neuroendocrine organ directly releasing into the aorta. Two anti-peptide antisera (anti-A and anti-B) raised against Gryllus bimaculatus allatostatin A1 (Grb-AST A1) and B1 (Grb-AST B1), respectively, were applied in the peroxidase-antiperoxidase (PAP) immunohistochemical technique on complete series of sections from the brain, suboesophageal ganglion, corpora cardiaca (CC), and corpora allata (CA) of G. bimaculatus. Both antisera yielded intense staining of numerous cells and nerve fibres. Serial sections, alternately stained with anti-A and anti-B confirmed that the anti-B generally stained more cells and nerves. The nature and rate of juvenile hormone (JH) biosynthesis and effects of allatostatins and allatotropin have been investigated in isolated corpora allata (CA) of adults and larvae of the noctuid tomato moth, Lacanobia oleracea. In adult female CA, mean rates of synthesis were relatively constant (10-16 pmol/pr/h) at all times. However, the range of JH synthesis by individual CA of similarly aged insects was quite large (2-30 pmol/pr/h). High performance liquid chromatography (HPLC) separation shows that adult female moth CA synthesise predominantly JH I and JH II. Rates of JH synthesis in vitro are dependent on methionine concentration. Synthetic Manduca sexta allatostatin (Mas-AS) caused a dose-dependent inhibition of JH synthesis by adult female CA but only to a max. of 54%, whilst 10 µM synthetic M. sexta allatotropin caused a 37% stimulation of CA activity.
