Temperature dependence (25-50 °C) of chlorophyll (Chl) fluorescence induction, far-red radiation (FR)-induced relaxation of the post-irradiation transient increase in apparent F0, and the trans-thylakoid proton gradients (ΔpH) was examined in maize leaves. Temperatures above 30 °C caused an elevation of F0 level and an enhancement of F0 quenching during actinic irradiation. Millisecond delayed light emission (ms-DLE), which reflects the magnitude of ΔpH, decreased strikingly above 35 °C, and almost disappeared at 50 °C. It indicates that the heat-enhanced quenching of F0 under actinic irradiation could not be attributed mainly to the mechanism of ΔpH-dependent quenching. The relaxation of the post-irradiation transient increase in apparent F0 upon FR irradiation could be decomposed into two exponential components (τ1 = 0.7-1.8 s, τ2 = 2.0-9.9 s). Decay times of both components increased with temperature increasing from 25 to 40-45 °C. Kinetics of non-photochemical reduction of the photosynthetic intersystem electron transport chain by exogenous NADPH was examined in osmotically lysed spinach chloroplasts by chlorophyll (Chl) fluorescence measurements under anaerobic condition. Upon the addition of NADPH, the apparent F0 increased sigmoidally, and the value of the maximal slope was calculated to give the reduction rate of plastoquinone (PQ) pool. Application of 5 µM antimycin A lowered significantly both the ceiling and the rate of the NADPH-induced Chl fluorescence increase, while the suppressive effect of 10 µM rotenone was slighter. This indicated that dark reduction of the PQ pool by NADPH in spinach chloroplasts under O2-limitation condition could be attributed mainly to the pathway catalysed sequentially by ferredoxin-NADP+ oxidoreductase (FNR) and ferredoxin-plastoquinone reductase (FQR), rather than that mediated by NAD(P)H dehydro-genase (NDH). and Ming-Xian Jin, Hualing Mi.
Reduction kinetics of P700+ after far-red radiation (FR)-induced oxidation in intact tobacco leaves was examined by analysing the post-irradiation relaxation of 810-830 nm absorbance difference. The reduction curve could be de-convoluted distinctively into two or three exponential decaying components, depending on the FR irradiance, the treating and measuring temperatures, and the extent of dark adaptation. The relative size of the pool of electrons accumulated in stroma reductants during actinic irradiation, which can be donated to P700+ via the intersystem chain, was estimated after short-term exposure of intact Zea mays leaves to elevated temperatures.
