Six types of sphaeractinomyxon are reported from the coelomic cavity of oligochaetes collected from the Minho River estuary in northern Portugal. Four new types are morphologically and molecularly described from freshwater species belonging to the genera Psammoryctides Hrabě and Potamothrix Vejdovský et Mrázek in the upper estuary, thus significantly increasing the number of known freshwater sphaeractinomyxon. In the lower estuary, sphaeractinomyxon types 8 and 10 of Rangel et al. (2016) are recorded infecting the marine oligochaete Tubificoides pseudogaster (Dahl). A single specimen of T. pseudogaster further displayed infection by one of the four new types found in the upper estuary, suggesting the involvement of sphaeractinomyxon in the life cycles of myxosporean species that infect migratory fish hosts. The acquisition of these second hosts is proposed to have allowed the myxosporean counterparts of sphaeractinomyxon to cross environmental barriers and conquer new habitats. Phylogenetic analyses of the SSU rRNA gene reveal the four new types clustering within the monophyletic clade of mugiliform-infecting myxobolids, strengthening the previously proposed involvement of the sphaeractinomyxon collective group in the life cycles of this specific group of myxosporeans. Endocapsa types also cluster within the latter clade, having actinospores that differ from those of sphaeractinomyxon only in the presence of valvular swellings that do not change when in contact with water. In this study, however, one type was found displaying actinospores with and without valvular swellings in the same oligochaete specimen. Chloromyxum leydigi Mingazzini, 1890, the type species of Chloromyxum Mingazzini, 1890, is redescribed based on material found in the gall bladder of the cartilaginous fish Torpedo marmorata Risso collected from the Portuguese Atlantic coast and its sporogonic development is described. Plasmodia and mature spores were floating free in the bile. Plasmodia are polysporic and highly polymorphic in shape and organization. Mature spores are spherical to subspherical with a pointed anterior end, measuring 12.3 ± 0.5 μm in length and 9.0 ± 0.5 μm in width. The spore wall is composed of two asymmetric shell valves, each bearing 4-5 elevated surface ridges. A bundle of 40-50 tapering caudal filaments extends from the basal portion of the shell valves. Four pyriform equal-sized polar capsules, measuring about 5.3 × 3.2 μm, are observed at the same level in the anterior pole of the spores, each containing a polar filament coiled in 8-9 (rarely 10) turns. Spore morphology, tissue tropism, host species and sequences of the SSU rRNA gene supported species identification. Since its discovery, this species has been dubiously reported from several cartilaginous hosts, namely due to the poor description of its features.
This paper describes the fine structure of oocysts of Nematopsis sp. (Apicomplexa, Porosporidae) found in the abductor muscles of seawater clams, Meretrix meretrix (Linnaeus, 1758) (Veneridae), collected near the city of Dammam (6°17'0''N, 50°12'0''E) in the Arabian Gulf off the coast of Saudi Arabia. Oocysts of an ellipsoidal shape were found among myofibrils of the abductor muscles of infected clams. Each oocyst is composed of an oocyst wall surrounding a single uninucleate vermiform sporozoite located in the lumen of the oocyst wall. The thin oocyst wall (0.70-0.85 µm thick) is composed of homogenous electron-lucent material formed by three layers of equal-thickness. The oocyst wall contains a plano-convex opercular-like structure about 2.5 µm in diameter and 0.75-0.90 µm thick, composed of a homogenous material with moderate electron density. The oocyst is of an ellipsoidal shape and is 15.6 ± 0.6 µm long and 11.1 ± 0.7 µm wide. Externally, the oocyst wall is surrounded by a complex dense network of numerous anastomosed microfibrils, which are attached to the oocyst wall, forming 2-3 layers and extending towards the periphery, at some points penetrating amongst the host cells. The myofibrils in some cases show evident aspects of lysis as a consequence of the appearance of lysosome-like vesicles. Lacking knowledge of a complete life cycle and/or molecular data precluded the conclusive identification of this species.
The first ultrastructural description of Ceratomyxa tenuispora Kabata, 1960 (Myxozoa, Bivalvulida) from Madeira Island (Portugal), a parasite found in the gall bladder of the commercially important black-scabbard fish, Aphanopus carbo Lowe is presented. This parasite possesses spherical to ellipsoidal disporous trophozoites. Spores have a central crescent-shaped body averaging 11.0 µm in length, 28.5 µm in thickness and 12.1 µm in width. The valves have two long opposite lateral processes (ribbon-like structures or tails), each averaging 173 µm in length. The total thickness of the spore averages 375 µm. The spore has two sub-spherical polar capsules (∼5.2 × 4.1 µm), each with a polar filament with 7 to 8 coils. Some ultrastructural aspects of the sporogonic stages are described. The trophozoites develop without contact with epithelial cells. The cytoplasmic membrane has numerous evenly distributed external slender projections about 0.3 to 0.7 µm long. The sporogenesis produces two spores without pansporoblast formation. In the matrix of the capsular primordium, microtubules with an unusual organisation were observed. A binucleate sporoplasm that contains several sporoplasmosomes and dense bodies fills the spore cavity and extends to the tails without penetrating them.
A new microsporidian species of the genus Glugea Thélohan, 1891 parasitising the marine teleost fish Cephalopholis hemistiktos Rüppell, collected from the Red Sea in Saudi Arabia, is described on the basis of microscopic and molecular procedures. Spherical and whitish xenoma were observed adhering to the intestinal wall. The numerous spores contained within these xenoma, were ovoid to pyriform and measured 4.3-6.0 µm (5.1 µm) in length and 1.8-2.9 µm (2.2 µm) in width. The spore's wall was composed of two thick layers, which were thinner in the area contacting the anchoring disk. The latter appeared at the spore's anterior pole, in an eccentric position to the longitudinal axis. A lamellar polaroplast surrounded the uncoiled portion of the polar filament projected to the basal region of the spore, giving rise to 26-29 turns with winding from the base to the anterior zone of the spore. The posterior vacuole, located at the spore's posterior pole, and surrounded by the polar filament coils, was irregular and composed of light material. Molecular analysis of the rRNA genes, including the ITS region, was performed using maximum parsimony, neighbour-joining and maximum likelihood methods. The ultrastructural features observed, combined with the phylogenetic data analysed, suggest this parasite to be a new species of the genus Glugea. This is the first species of this genus to be reported from Saudi Arabia and is herein named Glugea nagelia sp. n., Abdel-Azeem S. Abdel-Baki, Saleh Al-Quraishy, Sónia Rocha, Mohamed A.
