12. Observations on sporulation of Eimeria bovis (Apicomplexa: Eimeriidae) from the European bison Bison bonasus: effect of temperature and potassium dichromate solution

- The present study was designed to determine the influence of temperature and the addition of potassium dichromate solution (K2Cr2O7) on the efficiency of sporulation of Eimeria bovis (Zublin, 1908), a coccidian species most commonly diagnosed in European bison Bison bonasus (Linnaeus). Sporulation under conventional conditions (2.5% dichromate solution, 23 °C) was completed on the fourth day of incubation (control), whereas the oocyst development took two days more at the temperature of 18 °C, and two days less at 28 °C. Additionally, experimental sporulation of E. bovis at the temperature of 23 °C in the environment of sterilised tap water (without any preservation) took 9 days more compared to control. Finally, oocyst development took 16 weeks when the faces were stored in the refrigerator (3-5 °C) without any special additives. Overall, our results indicate that K2Cr2O7, besides the temperature, plays a crucial role in the process of sporulation of oocysts under laboratory conditions, as the longest delay in sporogony was observed when the faeces were stored without any other additives in the temperature of the refrigerator., Anna M. 13. Redescription of Eimeria dorcadis Mantovani, 1966 (Apicomplexa: Eimeriidae) from the dorcas gazelle (Gazella dorcas) in Saudi Arabia

- Eimeria dorcadis Mantovani, 1966 is redescribed from dorcas gazelle (Gazella dorcas (L.)) from Saudi Arabia. Oocysts were detected in 7 out of 22 faecal samples (32%) using floatation method. The sporulated oocysts are cylindrical, slightly flattened at the micropylar pole, measure in average 32 × 19 μm (27-36 × 16-24 μm), length/width ratio being 1.7 (1.5-2.1). Oocyst wall is 1.2 μm thick, smooth, double-layered; outer layer is slightly thicker, light blue in colour; inner layer brownish, with micropyle in the inner layer and apparently continual outer one, measures 2.2 μm, but lacks a micropylar cap. The sporocyst elongate-ellipsoidal, measures 14 × 8 μm (12-17 × 6-9 μm), length/width ratio being 1.8, with sporocyst residuum as circular compact, coarse, refractile granules. Stieda body is present, while substieda body is absent. Sporozoites banana-shaped, measure 11 × 2.5 μm, each with a large spheroidal refractile body at the wider pole. Sporulation time is 2-3 days at 25 ± 2 °C.

14. Vipera berus and V. ammodytes (Serpentes: Viperidae) represent new hosts for Caryospora simplex (Apicomplexa: Eimeriidae) in Europe

- During a survey of the coccidian parasites of reptiles, caryosporan oocysts were found in the faeces of wild and captive European viperid snakes Vipera berus (L.) and V. ammodytes (L.). Thirty two of 37 examined V. herus (86%) and 9 of 17 examined V. ammodytes (53%) specimens were found to be passing caryosporan oocysts. Morphological characters of all caryosporan isolates were identical and fitted well with the description of Caryospora simplex Léger, 1904. Experimental inoculation of severe combined immunodeficient (SC1D) mice with seven isolates of C. simplex from V. berus or V. ammodytes confirmed the heteroxenous life cycle pattern, for the first time for isolates of evidently European origin. Caryosporan developmental stages were observed in the connective tissues of the nose, cheeks, ear and scrotum in all inoculated SCID mice. V. berus and V. ammodytes represent new hosts for C. simplex. The present paper represents the first widely based report on coccidian parasites of the genus Caryospora Léger in European viperids. Our findings indicate a wide distribution of C. simplex throughout the range of distribution of snakes of the genus Vipera.
